采用反相高效液相色谱法分离测定手性药物前体扁桃酸及其底物苯乙酮酸.反相色谱柱为VarianC18柱,以甲醇-磷酸盐缓冲液(体积分数=1∶9)作为流动相,流速为1.0mL·min-1,检测波长为297nm,柱温25℃.该法测定的灵敏度高,可用于发酵液中苯乙酮酸和扁桃酸的定量测定.扁桃酸及苯乙酮酸浓度在5~20mmol·L-1范围内呈现良好的浓度-峰面积线性关系.  相似文献

交联海藻酸钙凝胶固定化酵母醇脱氢酶研究   总被引:2,自引:0,他引:2  
对用海藻酸钙包埋、戊二醛交联法固定酵母醇脱氧酶催化苯乙酮酸合成(R)-扁桃酸的过程进行研究,比较游离酶与固定化酶的酶学性质.实验结果表明:固定化酶的热稳定性显著提高,游离酶在70℃时酶蛋白变性失去活力,而固定化酶在65℃保温1 h的能保持64%的酶活力,在70℃时酶活力仍町保留48.6%;固定化酶的最适温度由30℃升至40℃,最适反应pH值由6.8下降为5.8;固定化酶保留了62.72%的游离酶活性, 固定化酶的表观米氏常数和最大反应速率分别为37.33 mmol/L和358.42 nmol/min.该固定化酶具有良好的储存稳定性和操作稳定性.  相似文献

选用酵母菌、乳酸菌、粪链球菌、假丝酵母等4个种属共15株菌株,研究其对苯乙酮酸甲酯的不对称还原催化特性.以具有R(-)-扁桃酸甲酯较高转化活力的菌株S.cNo.1、S.cNo.3、S.cNo.9作为出发菌株,采用紫外与微波复合诱变的方法,筛选获得S.c3.5.16突变株.考察培养温度、pH值对该突变株转化活力的影响,结果表明,S.c3.5.16菌株的R(-)-扁桃酸甲酯生物转化最适反应条件为培养基pH6.5、温度38℃,苯乙酮酸甲酯转化为扁桃酸甲酯的得率达到99.3%,R(-)-扁桃酸的光学纯度达到95.5%e.e..  相似文献

建立一种同时测定生物不对称转化发酵液中扁桃酸及苯乙酮酸含量的反相离子对高效液相色谱方法.色谱柱为HypersilC18柱(4.6mm×250mm,5μm),流动相为甲醇-25mmol·L-1磷酸二氢钾缓冲液(内含6mmol·L-1四丁基溴化铵,NaOH调pH至7.0)(体积分数15∶85),流速为1.0mL·min-1,检测波长220nm,柱温为室温.扁桃酸与苯乙酮酸在0.5~20mmol·L-1内,其浓度与峰面积呈现良好的线性关系(r=0.9994,0.9997).二者的加样回收率均为96.5%~100.6%.当信噪比(S/N)为3时,二者的最低检测浓度分别为2.4和8μmol·L-1.该方法简便、准确、灵敏度高,可用于手性生物转化中扁桃酸与苯乙酮酸的定量分析.  相似文献

采用DEAE Sepharose离子交换层析、Butyl Sepharose疏水层析和Blue Sepharose亲和层析等分离方法,从高产D-(-)-扁桃酸的酵母菌株BY1.1b中纯化得到电泳纯的D-(-)-扁桃酸脱氢酶.采用Sephacryl S-200分子筛层析柱测得该酶的相对分子质量约60 kDa,用SDS-PAGE电泳测得该酶亚基分子量为32 kDa,表明该酶属于同型二聚体.该酶的最适反应温度为30℃,最适反应pH为6.0.该酶在低于30℃,pH5.5~8.0较为稳定.以NADH和苯乙酮酸为底物,其Km值分别为Km(NADH)=99.2μmol/L和Km(苯乙酮酸)=263.3μmol/L.  相似文献

为获得稳定高效生物脱硫催化剂,采用海藻酸钠(SA)包埋法制备生物脱硫菌UP-1的固定化细胞,使用环镜扫描电镜(ESEM)对固定化细胞进行分析和表征.结果表明:固定化细胞可以重复使用5次以上,而未固定化细胞只能有效重复使用2~3次;固定化小球内部的网状结构、表面的致密表层和固定化细胞初始阶段存在覆盖物,说明SA是固定UP-1的合适载体,反应底物和产物在固定化载体中的传质阻力增加和包埋材料对UP-1的初始覆盖导致了固定化UP-1脱硫活性的降低;最佳条件下得到的固定化细胞菌体在载体内部均匀饱和分布,这从微观结构上证明固定化可以提高生物催化剂总脱硫能力和稳定性.  相似文献

考察了酵母菌株FD11b不对称还原苯乙酮酸生产R-(-)-扁桃酸的重复使用性能.菌株FD11b在持续6批次(14 d)的还原反应中,一直保持着较高的催化还原活性,且目标产物R-(-)-扁桃酸的对映体过量值(e.e.值)均高于96.5%.在30 L发酵罐上进行Fed-Batch放大试验,当菌体浓度(细胞干重,以下同)为12.5 g/L,底物苯乙酮酸浓度控制在20~30 mmol/L时,还原反应49 h,产物浓度可达125 mmol/L,产物得率为96.1%,e.e.值为98.5%.  相似文献

本研究以辽宁东部山区野生植物资源—山楂果为原料,利用海藻酸钠(Alginate)载体包埋法制备固定化酵母(Hansenula anomala)细胞,经增殖后,采用自制加工的2000毫升生物反应器进行了半连续发酵试验,证明在pH5.1—5.5,温度控制在32—35℃,装量1∶1(v/v),发酵时间为16—20小时,乙醇的转化率最高可达到理论值的27.5%.通过固定化与非固定化细胞的乙醇发酵对比试验,征明固定化细胞具有发酵速度快、周期短、发酵能力强,并且可以连续使用等特点.发酵后所制得的饮料颜色呈淡金红,香味纯正,酸甜适口,具有山楂发酵饮料的独特风格.经测试,含多种氨基酸和维生素,是一种全天然、高营养的强身健体饮料.  相似文献

微生物全细胞转化靛蓝的过程中,靛蓝易被游离细胞吸附,造成产物提取困难,是微生物产靛蓝工业化进程中面临的主要问题之一.为解决该问题,尝试了不同固定化方法将基因工程菌固定,其中海藻酸钠包埋法取得了最好的效果,最佳包埋条件为3.5%海藻酸钠、1%氯化钙,固定化细胞在有机-水两相体系中可以重复利用两次,首次转化6h后,靛蓝浓度为15.9mg/L.另外,两相体系的引入有效避免了固定化细胞产靛蓝过程中伴随的产物分解现象,选择正十二烷作为有机相,优化得到其最佳比例为体积分数40%.体系对吲哚的最佳耐受浓度为400mg/L,在此条件下,靛蓝产率为0.28mg/mg.  相似文献

采用海藻酸钠包埋法和海藻酸钠-壳聚糖包埋-交联法固定化漆酶,探讨了固定化条件、固定化漆酶及游离酶的酶学性质,结果表明:包埋法和包埋-交联法固定化漆酶的最佳条件分别为海藻酸钠浓度3%、CaCl2浓度1.5%和海藻酸钠浓度2%、CaCl2浓度2%、壳聚糖浓度1.5%、戊二醛浓度1%.两种固定化漆酶的最适pH和最适温度相同,分别为5.0和30℃,游离酶pH为4.6,温度20℃.包埋法固定化漆酶、包埋一交联法固定化漆酶和游离酶的米氏常数分别为3.3,2.8,1.22mmol/L.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
