Summary Membranes allow the rapid passage of uncharged lipids. Phospholipids on the other hand diffuse very slowly from one monolayer to another with a half-time of several hours. This slow spontaneous movement in a pure lipid bilayer can be selectively modulated in biological membranes by intrinsic proteins. In microsomes, and probably in bacterial membranes, non-specific phospholipid flippases allow the rapid redistribution of newly synthesized phospholipids. In eukaryotic plasma membranes, aminophospholipid translocase selectively pumps phosphatidylserine (PS) and phosphatidylethanolamine (PE) from the outer to the inner leaflet and establishes a permanent lipid asymmetry. The discovery of an aminophospholipid translocase in chromaffin granules proves that eukaryotic organelles may also contain lipid translocators.  相似文献

Incorporation of chinoform-ferric chelate was demonstrable in cultured neural retinal cells of chick embryos after 1 h of incubation, and the lipid peroxide level in the cells was increased strikingly 1 h thereafter. On the other hand, free ferric ions were scarcely incorporated into the cells, and a significant increase in the lipid peroxide level in the cells was not observed. These data indicate that chinoform is carrier of iron for its passage through cell membranes and that the incorporated iron induces lipid peroxidation which in turn leads to neural cell degeneration.  相似文献

Summary This note is a preliminary report on some research being carried out at present on the sediments of Monterosi, a volcanic lake of Central Italy, under the direction of Prof.G. E. Hutchinson, of Yale University, USA. Herein are given some data taken from the pollen analysis of cores of the lake's sediments. The first results seem promising for the stratigraphy of the recent Quaternary Age in Central Italy, and for other related problems.

The role of γ-aminobutyric acid (GABA) as a signal in animals has been documented for over 60 years. In contrast, evidence that GABA is a signal in plants has only emerged in the last 15 years, and it was not until last year that a mechanism by which this could occur was identified—a plant ‘GABA receptor’ that inhibits anion passage through the aluminium-activated malate transporter family of proteins (ALMTs). ALMTs are multigenic, expressed in different organs and present on different membranes. We propose GABA regulation of ALMT activity could function as a signal that modulates plant growth, development, and stress response. In this review, we compare and contrast the plant ‘GABA receptor’ with mammalian GABAA receptors in terms of their molecular identity, predicted topology, mode of action, and signalling roles. We also explore the implications of the discovery that GABA modulates anion flux in plants, its role in signal transduction for the regulation of plant physiology, and predict the possibility that there are other GABA interaction sites in the N termini of ALMT proteins through in silico evolutionary coupling analysis; we also explore the potential interactions between GABA and other signalling molecules.  相似文献

Several neurotransmitters and hormones with potential to trigger a simultaneous contraction of the oviducts and the spermathecal myoepithelium were examined. Saline (0.05 ml), or 0.05 ml saline plus acetylcholine (9 mg), norepinephrine (50 micrograms), arginine-vasotocin (25 units), prostaglandin F2 alpha (3 micrograms) were injected into the spermathecal region of female newts (n = 24 per group). The numbers of sperm present in the cloacae of prostaglandin-injected animals (107 +/- 30 SEM) were significantly greater than the numbers detected in saline (27 +/- 5 SEM) and in uninjected (14 +/- 3 SEM) controls. Smaller and less consistent increases in the numbers of sperm were detected in the vasotocin- and norepinephrine-injected groups. Study of sections from ovulating female newts failed to produce evidence that pressure from the passage of ova through the posterior portion of the oviduct forced sperm from the spermatheca. Observations indicate an active role for the spermathecal myoepithelium in the discharge of stored sperm and of a role for prostaglandin F2 alpha in triggering that discharge.  相似文献

Isolation of a growth-stimulating agent from human skin fibroblast cultures   总被引:1,自引:0,他引:1  
Summary Cell-free supernatants were harvested from cultures of human skin fibroblasts, were applied on to DEAE-cellulose columns, and the first fraction eluted with phosphate-buffered saline contained the growth-stimulating agent. The eluted fraction was then passed through a series of amicon membranes. After passing through PM-10, the filtrate stimulated growth of bovine vascular endothelial, canine myocardial, and human mammary carcinoma cells.  相似文献

Summary Some correlations between the physicochemical properties of drugs and their pharmacokinetic behaviour are outlined. Based on the permeability characteristics of simple model membranes (porous membrane, lipid membrane) permeation and distribution of drugs in the animal body can be described and understood on simple physico-chemical terms. Some clinically important aspects — the absorption of drugs from the intestinal tract, the passage through the blood-brain-barrier and the renal excretion as governed by passive tubular reabsorption — are discussed in more detail. Thereby it appears that the solubility of a drug in lipid material, which may be suitably expressed as partition coefficient between an organic solvent and a buffer solution of pH 7.4, is a major factor in determining its pharmacokinetic behaviour.  相似文献

Summary Large-scale preparations of tissue homogenates are not conveniently obtained with the conventional Potter-Elvehjem homogenizer. Ionic channels are elementary excitable elements in the cell membranes of heart and other tissues. They produce and transduce electrical signals. After decades of trouble with quantitative interpretation of voltage-clamp data from multicellular heart tissue, due to its morphological complexness and methodological limitations, cardiac electrophysiologists have developed new techniques for better control of membrane potential and of the ionic and metabolic environment on both sides of the plasma membrane, by the use of single heart cells. Direct recordings of the behavior of single ionic channels have become possible by using the patch-clamp technique, which was developed simultaneously. Biochemists have made excellent progress in purifying and characterizing ionic channel proteins, and there has been initial success in reconstituting some partially purified channels into lipid bilayers, where their function can be studied.  相似文献

Summary The distribution of alkaline phosphatases has been studied in male and female gonads of some isopod crustaceans by means of Gomori's reaction.Monophosphatases are found on all Feulgen-positive nuclear structures, in the nucleolus and in the cytoplasm of young oocytes. They disappear from the cytoplasm in auxocytosis, during yolk formation, although they persist in the germinal vesicle. The authors think that the cytoplasmic localization is coincident with ribonucleic acid distribution.From the experiments related in the following note, nucleolar phosphatases appear to be in some way different from the nuclear (sensu stricto) and cytoplasmic phosphatases.Diphosphatases do not follow so closely the nucleic acid localization. They are absent from cytoplasm; in the nucleolus they are not distributed uniformly but show characteristic structure; in the nucleus they are not localized in the chromosomes but homogeneously diffused.  相似文献

Photon emission in the visible and near ultraviolet range by samples of human tissue removed during surgery has been measured by means of a low noise photomultiplier coupled to a data acquisition system. The results show that among the 25 analyzed samples the 9 from normal tissues had an emission rate of the order of some tens of photons/cm2 min, while most of the 16 tumor tissue samples had a very much higher rate.  相似文献

Summary A study was undertaken on the variations of the redox potential level produced by streptomycinin vitro andin vivo. We have been able to show that, owing to an oxidative effect, streptomycin produces an increase of the redox potential level. This oxidative effect varies in degree according to the condition of the patient.We also found that in the blood and in the spinal fluid of patients suffering from tubercular meningitis factors are present which inhibit the action of streptomycin.The results of our findings lead to the conclusion that the dose of streptomycin must be varied according to the condition of the patient if the constant level required for an efficient therapy is to be maintained in the blood and in the spinal fluid.  相似文献

Laminin and nidogen (entactin) are major glycoprotein components of basement membranes. At least seven different isoforms of laminin have been identified. Laminin and nidogen form high affinity complexes in basement membranes by specific binding between the laminin γ1 chain and the G3 globule of nidogen. Additional interactions between nidogen and collagen IV, perlecan and other basement membrane components result in the formation of ternary complexes between these matrix components. Nidogen is highly susceptible to proteolytic cleavage, and binding to laminin protects nidogen from degradation. Nidogen is considered to have a crucial role as a link protein in the assembly of basement membranes. Basement membrane components are synthesized at high levels during tissue growth and development, and sites of morphogenesis correlate with localized remodelling of basement membranes. The formation of distinct basement membrane matrices in the developing embryo is influenced by the laminin isoforms produced and by whether laminin and nidogen are co-expressed and secreted as a complex or are produced by cooperation between two cell layers. The potential roles of laminin-nidogen complexes, cell-matrix interactions, and other intermolecular interactions within the matrix in basement membrane assembly and stability are discussed in this review.  相似文献

目的采用酶消化结合组织块培养法对山羊颞下颌关节(temporomandibular joint,TMJ)盘细胞进行体外培养和扩增,探索TMJ关节盘细胞体外培养及扩增的新方法。方法在无菌条件下,切取一月龄山羊TMJ关节盘,剪至1.0mm^3的碎块,用0.25%胰酶、0.01%I型胶原酶消化关节盘组织块,将消化好的组织块置入6孔板中培养。在倒置显微镜下连续观察细胞的形态变化及贴壁率,甲苯胺蓝染色、I型胶原免疫组化染色进行细胞鉴定,测定其生长曲线。结果原代培养的关节盘纤维软骨细胞4天可观察到贴壁细胞,7天贴壁细胞逐渐增多,第10天时细胞彼此相连,铺满平底,细胞以梭形为主,部分多角形。传代后12小时贴壁率达90%,大部分为多角形,4~5天即可长满瓶底。甲苯胺蓝染色可见异染颗粒,胶原免疫纽化染色胞浆内可见棕黄色颗粒。结论酶消化结合组织块培养法培养的山羊TMJ关节盘细胞具有较强的增殖能力,可作为TMJ关节盘组织工程中获取大量原代细胞的实用方法。  相似文献

A method is described for using a fluorescein isothiocyanate concanavalin A conjugate to stain human cell membranes in formalin fixed paraffin embedded tissue. 57 neoplastic and normal tissue sites were examined. In 54 malignant tumours, bright green fluorescence was confined to the cell membranes while in 23 benign tumours and normal tissue sites, the membranes were unstained or showed a diminished level of fluorescence. The distinction between malignant and hyperplastic or normal cells was clear cut and definite.  相似文献
