谷氨酰胺合成酶是应用广泛的生物酶类,以LNU0165为实验出发菌株,对不同pH、温度条件下产谷氨酰胺合成酶的最佳发酵条件进行了研究,通过测定其谷氨酰胺合成酶酶活力,确定LNU0165为高产菌株.从分子水平上对该菌株进行16S rRNA序列分析及同源性比较,以及生理生化的鉴定.根据LNU0165菌株16SrRNA与GenBank数据库中Arthrobacter globiformis的16S rRNA的序列具有99%的同源性,确定LNU0165为球形节杆菌(Arthrobacter globiformis).  相似文献

一株脂肪酶的筛选和产酶条件及其酶性质的研究   总被引:4,自引:0,他引:4  
从东营油污样中分离获得一株产脂肪酶的菌株BYCM-1,经鉴定为根霉属.研究了该菌株的产酶条件及其酶性质.研究表明该酶最适的产酶条件为:培养基初始pH 6.5,温度40℃,培养时间72 h.酶作用的最适pH值为8.0,最适温度为40℃.Ca2 和K 对酶活性有一定的激活作用,而Mg2 ,Cu2 ,Fe2 对酶活性有不同程度的抑制作用.  相似文献

目的:从土壤中筛选产a-淀粉酶活力较强的菌株并对其酶学性质进行初步研究。方法:结合选择性分离培养,碘液检测法,酶活力测定和形态学检测等方法从土壤中分离出产酶活力较强的菌株,并对其所产仅一淀粉酶的最适反应温度和pH值进行研究。结果:共分离到4株能产a-淀粉酶的菌株,其中菌株4产酶活力最高。该菌株所产仅一淀粉酶最适反应温度为50~60℃,最适pH值为6.2。结论:分离到1株产a-淀粉酶酶活力较强的芽孢杆菌,所产仪一淀粉酶属于中温淀粉酶,最适pH值为6.2。  相似文献

对肉色曲霉产碱性脂肪酶的发酵条件进行优化,摇瓶实验表明,该菌株最适产酶培养基组成为(%):黄豆饼粉2,NH4NO3 0.5,玉米粉1,糊精1,柠檬酸钠0.1,K2HPO4 0.5,FeSO4 0.006,大豆油0.6,吐温-800.5mL,pH 9.0.最适产酶温度为28℃,产酶高峰出现在96小时,最佳装液量为35mL,碱性脂肪酶菌株的酶活可达9.3U/mL.脂肪酶的最适作用pH为9.0,最适作用温度为30℃.  相似文献

【目的】筛选分离新的纤维素酶产酶菌株,并对其生长特性和产酶特性进行研究。【方法】从堆肥样品中分离得到一株产纤维素酶菌株,定名为CP1,利用16SrRNA序列对比和Biolog微生物鉴定系统进行鉴定。通过测定菌株生长速率确定其最适生长条件。采用CMC发酵培养基进行纤维素酶的研究,确定其产酶曲线。测定粗酶液最适作用温度和pH值,热稳定性和金属离子对其影响。【结果】经鉴定该菌株为梭形芽孢杆菌(Lysinibacillus fusiformis),其最适生长温度为30℃,最适生长pH值为6。在含CMC-Na的培养基培养,该菌株的生长与产酶同步进行,培养48h菌株生长量达到最大,培养液的CMC酶活力同时达到最大值,为0.46U/mL。该菌株所产的纤维素酶既有酸性CMC酶活力,又有碱性CMC酶活力,并以碱性CMC酶为主,酸性CMC酶的活力只有碱性CMC酶的71.4%。其酸性CMC酶的最适作用pH值为6.0,碱性CMC酶的最适作用pH值为8.0。另外,该菌株所产碱性CMC酶活的最适作用温度为40~50℃,而且0.5%的Cu2+使其酶活降低45%。【结论】菌株CP1为首次报道的梭形芽孢杆菌产纤维素酶新菌株,具有独特的酸碱性环境下的纤维素酶活力。  相似文献

以假单胞菌(Pseudomonas sp.)TS1138为供试菌株,对微生物酶法生产L-半胱氨酸的条件进行了初步研究。通过对产酶培养基中的碳氮源进行研究,得到了TS1138菌株产酶的最佳碳氮源分别为葡萄糖和尿素,DL—ATC的最适添加量为5g/L;通过对产酶培养基的产酶条件进行研究,得出了最适的种子接种量为10%,产酶培养基的最适初始pH为8.0,500mL三角瓶的最适装液量为40mL。  相似文献

从低等木食性白蚁——黑胸散白蚁肠道中通过七叶苷与柠檬酸铁铵的显色反应筛选出14株产β-葡萄糖苷酶的菌株.采用选择培养复筛出1株在60℃, pH值为8的培养条件下,具有耐高温耐碱性的产β-葡萄糖苷酶的菌株BH1.通过对BH1菌株进行形态学鉴定,结合16S rRNA序列对比分析,初步判断BH1属于芽孢杆菌属(Bacillus sp.).运用单因素和正交试验法对其在产酶过程中的培养条件,如时间、温度、 pH等进行优化.通过单因素试验可知BH1菌株产酶最佳碳源是麦麸,氮源是酵母膏,无机盐是NaCl,最适pH值为8.0,最适温度为60℃. BH1菌株最佳酶活力为36.24 U/mL,在pH值为9.0,温度为70℃时还具有相对较高的酶活,也进一步说明了该菌株所产的β-葡萄糖苷酶具有耐高温耐碱的特性.  相似文献

以普鲁兰糖为唯一碳源,利用平板涂布法从海泥中分离到一株产碱性普鲁兰酶的细菌M25,结合菌株形态特征观察和16S rDNA基因序列分析,确定该菌株为假交替单胞菌(Pseudoalteromonas sp.).该菌在30℃、200 r/min条件下培养18 h达到产酶最高峰,其合成普鲁兰酶的模式属于同步合成型.酶学性质研究表明:该普鲁兰酶的最适作用温度是50℃,在70℃下保温2 h活力残留50%以上;最适作用pH值为8.0,在pH值6.0~9.0较稳定.实验结果显示该菌所产普鲁兰酶具有较好的温度稳定性和pH稳定性.  相似文献

从腈纶厂污水处理系统的活性污泥中分离和筛选到一株丙烯酰胺转化菌.经初步鉴定,菌株T3属于红球菌属(Rlwdococcus sp.).对菌株T3产生腈水合酶的最适条件进行研究,结果表明,该菌株的最适产酶条件为培养基初始pH值7.5,培养温度35℃,培养时间72h,同时加入0.05g/L Co2+也会明显提高酶活性.在最适产酶条件下,菌株,13培养液的最高比酶活可达128.3U/mg,比优化前提高55.1%.  相似文献

为提高海洋细菌THN1产右旋糖酐酶的能力,本研究利用单因素试验和响应面试验对菌株THN1的发酵条件进行优化,并探究菌株THN1右旋糖酐酶的酶学性质。优化结果表明,菌株THN1的最佳产酶条件为麸皮5 g/L、胰蛋白胨20 g/L、NaCl 10 g/L、右旋糖酐T20 5 g/L,陈海水配置,pH值为8.2,30℃培养24 h。在此条件下,菌株THN1的发酵液酶活力达到3.71 U/mL,是优化前(初始酶活力为0.12 U/mL)的31倍。酶学性质分析表明,右旋糖酐酶的最适反应温度为40℃,在30℃条件下放置5 h酶活力基本保持稳定;最适反应pH值为7.5,在pH值为5.0-9.0的条件下相对酶活力能保持50%以上。本研究成果可为缩短右旋糖酐酶生产周期和降低成本提供数据支撑,并认为菌株THN1右旋糖酐酶在口腔护理方面具有良好的应用潜力。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
