B cells express immunoglobulins on their surface where they serve as antigen receptors. When secreted as antibodies, the same
molecules are key elements of the humoral immune response against pathogens such as viruses. Although most antibodies are
restricted to binding a specific antigen, some are polyreactive and have the ability to bind to several different ligands,
usually with low affinity. Highly polyreactive antibodies are removed from the repertoire during B-cell development by physiologic
tolerance mechanisms including deletion and receptor editing. However, a low level of antibody polyreactivity is tolerated
and can confer additional binding properties to pathogen-specific antibodies. For example, high-affinity human antibodies
to HIV are frequently polyreactive. Here we review the evidence suggesting that in the case of some pathogens like HIV, polyreactivity
may confer a selective advantage to pathogen-specific antibodies. 相似文献

Summary The inclusion of vitamin C in the drinking water of BALB/c mice was without effect on the humoral antibody response to sheep red blood cells and bacterial lipopolysaccharide. However, there was a significantly increased cell-mediated immune response as determined by increased T-lymphocyte responses to concanavalin A. This might suggest a mechanism, along with interferon enhancement, for the possible protection by vitamin C against some viral infections.We thank Miss Wendy Treat for excellent technical assistance. 相似文献

Cellular and humoral immune mechanisms recruited to defend the host from infectious agents depend upon the early immune events
triggered by antigen. The cytokine milieu within which the immune response matures is the most important of many factors that
govern the nature of the immune response. Natural T cells, whose function is controlled by CD1d molecules, are an early source
of cytokines that can bestow type 1 or type 2 differentiative potential upon helper T lymphocytes. This review attempts to
illuminate the glycolipid antigen presentation properties of CD1d, how CD1d controls the function of natural T cells and how
CD1d and natural T cells interact to jump start the immune system. CD1d is postulated to function as a sensor, sensing alterations
in cellular lipid content by virtue of its affinity for such ligands. The presentation of a neo-self glycolipid, presumably
by infectious assault of antigen-presenting cells, activates natural T cells, which promptly release pro-inflammatory and
anti-inflammatory cytokines and jumpstart the immune system.

Summary Daily administrations of tryptophol to laboratory rodents resulted in significantly depressed antibody production to heterologous red blood cell challenge, but did not alter cellular-mediated responses to oxazalone. The involvement of heat shock proteins in immune response is categorized into four distinct paradigms. In the First Paradigm, HSP derived from foreign organisms act as classical foreign antigens, and they elicit immune response to the non-conserved HSP epitopes. The Second Paradigm refers to instances where the host responds to self HSP to which there is no central or peripheral tolerance. The Third Paradigm involves molecular mimicry, where cross-reactivity between an HSP and another protein leads to an immune response to the latter under conditions which elicit an immune response to the former, such as infection with a bacterium whose immunodominant antigen is an HSP. The Fourth Paradigm refers to situations where an HSP-antigen complex elicits an effective response to the antigen andnot to the HSP. Thus the HSP acts as a carrier for the antigenic peptide. The role of HSP in recognition by γδ T cells may also fall into this paradigm. In this article, the Fourth Paradigm is considered as a crucial element in the development of vaccines against cancers and infectious diseases, and is analyzed through the prism of the observed association of hsp70 species with antigenic peptides. 相似文献

Humoral immune reactions to heat shock proteins (hsp) from microorganisms are one aspect of microbial infections in humans. The production of antibodies which are specific to epitopes present on procaryotic hsp leads also to the appearance of cross-reactive serum antibodies in the host organism that react with human hsp. This article discusses the consequences of such autoreactive antibodies for the host in context with the development of immune tolerance and autoimmune diseases, especially rheumatoid arthritis (RA), and in experimental animal models for arthritis such as adjuvant arthritis in rats. On the basis of epitope cross-reactivity between hsp and other host proteins, a hypothesis is presented for the development of autoimmune disease following the production of hsp-specific antibodies. 相似文献

T lymphocytes, which are central to almost every immune response, frequently recognize microbial hsp60. Such cells could provide an early defense mechanism against pathogenic microbes. However, T cells also recognize epitopes of hsp60 shared by microbe and host. Not only conventional / T cells respond to hsp60; / T cells do so, as well. In fact, certain / T cells seem to have a particular preference for this molecule. Recognition of stressed host cells expressing hsp60 could facilitate the scavenger function of the T cell system. Reversible tyrosine phosphorylation is a key posttranslational regulatory modification of proteins in all eukaryotic cells
in normal and pathological processes. Recently a pivotal janus-faced biological role of the low molecular weight protein tyrosine
phosphatase (LMWPTP) has become clear. On the one hand this enzyme is important in facilitating appropriate immune responses
towards infectious agents, on the other hand it mediates exaggerated inflammatory responses toward innocuous stimuli. The
evidence that LMWPTP plays a role in oncological processes has added a promising novel angle. In this review we shall focus
on the regulation of LMWPTP enzymatic activity of signaling pathways of different immunological cells, the relation between
genetic polymorphism of LMWPTP and predisposition to some type of inflammatory disorders and the contribution of this enzyme
to cancer cell onset, growth and migration. Therefore, the LMWPTP is an interesting target for pharmacological intervention,
thus modifying both inappropriate cellular immune responses and cancer cell aggressiveness.

Suppression of the immune response by drugs interfering with the metabolism of serotonin 总被引:1,自引:0,他引:1
The work was based on the assumption that neurohumoral control of the immune response, particularly in stressed animals, involves central serotoninergic mechanisms. Rats immunized with sheep erythrocytes were stressed by repeated restraints and/or treated with a precursor of serotonin (5-hydroxytryptophan, 5-HTP) or with an inhibitor of serotonin synthesis (parachlorophenylalanine, PCPA). As expected, repeated stresses reduced the plaque-forming cell (PFC) response. Treatment with 5-HTP also reduced the PFC response, and potentiated the immunosuppressive effect of stress. This was accompanied by increased metabolism of serotonin in the brain, as indicated by increased concentration of its metabolite, 5-hydroxyindoleacetic acid (5-HIAA), in cerebral tissue. Treatment with PCPA also suppressed the PFC response, but this suppression was accompanied by decreased levels of brain serotonin and of 5-HIAA. Plasma corticosterone levels were elevated in rats treated with PCPA. It seems that putative central effects of PCPA on serotoninergic regulation of the immune response were outweighed by its effects on corticosterone secretion and/or on lymphoid cells. 相似文献
