谷胱甘肽过氧化物酶4(GPX4)是一种能够特异性催化谷胱甘肽将脂质过氧化物转化为类脂醇的硒蛋白,其表达量上调在抑制细胞铁死亡以及相关炎症反应中发挥着重要的作用。为了在青蒿成分中筛选出激动GPX4的潜在活性小分子,首先在“中药系统药理学数据库与分析平台(TCMSP)”检索得到126个青蒿活性小分子;然后,通过类药性筛选出50个青蒿活性小分子;其次,采用GPX4-配体对接模拟和GPX4-配体互作模式对比筛选出8个青蒿活性小分子;继而通过GPX4-配体结合自由能计算分析筛选,发现artemisinin(ARS)、patuletin、kaempferol与阳性对照物1d4(PKUMDL-LC-101-D03)的作用方式相似;最后,进行分子动力学模拟。结果显示:ARS、patuletin、kaempferol、阳性对照物1d4可与GPX4形成稳定性较强的复合物。  相似文献

研究了藤茶多糖组分AGP-3的体外抗氧化活性.用K3[Fe(CN)6]模型测定多糖的还原能力,用试剂盒方法测定丙二醛(MDA)及活性氧(ROS)含量,用分光光度法测定小鼠红细胞溶血和小鼠肝线粒体肿胀度.结果表明,AGP-3在化学模拟体系具有一定的还原能力和清除活性氧能力,一定浓度范围内具有体外清除小鼠血清活性氧的作用,并能减少小鼠肝组织及肝线粒体MDA的生成,可抑制小鼠红细胞溶血和肝线粒体肿胀.  相似文献

为实现单链抗体的可溶性表达, 制备具有谷胱甘肽过氧化物酶(GPX)活力的单链抗体, 在单链抗体表达载体pTMF-2F3上去除原2F3基因N端非必需的18个氨基酸, 采用新的表达载体pRose质粒, 在2F3的N端引入13个氨基酸的前导肽, 转入BL21(lys S)中, 使其分泌到大肠杆菌的周质腔中得以表达, 获得可溶性表达产物. 产物经过分离纯化、 Wester n Blot印迹和硒化测活确定具有GPX活性的鼠单链抗体, 其GPX活力为2 530 U/μmol. 以脂质过氧化、 细胞存活率和细胞膜完整性为指标的抗氧化实验研究表明, 具有GPX活性的单链抗体对大鼠乳鼠表皮细胞有抗紫外线损伤的作用, 是膜脂质过氧化的有效抑制剂.  相似文献

研究哺乳动物谷胱甘肽过氧化物酶基因家族(Gpx)的亲缘关系和进化选择压力性质。从NCBI等网站获取哺乳动物Gpx基因编码区核酸序列和GPX蛋白氨基酸序列,并用MEGA、Clustal构建亲缘关系树,用SignalP和TargetP分析信号肽和亚细胞定位信息;用K estimator和PAML分析其Gpx基因家族进化先后顺序和选择压力;用Diverge分析GPX蛋白家族功能分歧。结果表明,GPX蛋白家族进化中形成两个亚类,其一包括GPX1和GPX2,其二包括GPX3、GPX5和GPX6;各GPX蛋白成员均有高度保守的GPX蛋白家族基序和高度保守的硒代半胱氨酸或半胱氨酸残基,氨基端信号肽分析显示,GPX1蛋白为胞浆线粒体型,其余家族成员均为胞外分泌型;哺乳动物Gpx1基因进化上受到严格的负选择作用,Gpx基因家族成员保守区也受到负选择作用,但用枝位点模型却检出了正选择作用,GPX蛋白家族内存在轻微的功能分歧。哺乳动物GPX蛋白家族的进化上的负选择作用表明其在哺乳动物清除自由基抗氧化方面的关键作用。  相似文献

以体质量为4.9±0.2 g的泥鳅为实验对象,在基础饲料中添加了不同含量(0%、0.3%、0.6%、0.9%和1.2%)的硒化壳聚糖。经过为期60 d的养殖后,比较实验鳅肝脏中过氧化氢酶、谷胱甘肽过氧化物酶和超氧化物歧化酶的活性,以及丙二醛含量和总抗氧化能力的变化。结果发现:硒化壳聚糖对泥鳅过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPX)活性有明显的促进作用,CAT活性随着硒化壳聚糖含量增加而增大,在添加量为1.2%时活性最大,各实验组的GPX活性均显著高于空白组;超氧化物歧化酶(SOD)的活性在硒化壳聚糖添加水平大于0.3%时显著高于对照组,并在1.2%添加水平达到最大;实验组泥鳅肝脏内丙二醛(MDA)含量相比于空白组明显下降,且随着硒化壳聚糖的浓度提升而下降,在1.2%时达到最低;实验组总抗氧化能力也有明显的上升,在0.9%时达到最大。整体上,饲料中添加硒化壳聚糖可以显著提升大鳞副泥鳅的抗氧化能力。  相似文献

考察了紫外线照射小鼠胸腺细胞后, 对细胞存活率、 细胞周期、 细胞凋亡、 脂质过氧化程度以及外源性活性氧含量变化的影响, 表明紫外线对胸腺细胞有严重损伤. 研究了谷胱甘肽过氧化物酶(GPX)的模拟物2-硒桥联β-环糊精和2-碲桥联β-环糊精抵抗紫外线损伤细胞的能力, 结果表明, GPX模拟物能有效地保护细胞, 防止紫外线引起的损伤.  相似文献

合成了尚未见文献报道的铁(Ⅱ)-酪氨酸模拟超氧化物歧化酶。用差动热分析仪(DTA),监测模拟物生成;分析了热稳定性。通过元素分析、红外光谱初步对配合物组成和结构进行了表征,确定了金属离子与氨基酸发生了配位反应;利用改进核黄素光还原氯化硝基四氮唑兰(NBT)法测定了模拟物催化歧化超氧阴离子自由基(O^-2)反应的活性。研究表明:合成配合物结构稳定,水溶液中具有一定的SOD样活性。  相似文献

机体在生命活动的氧化代谢过程中不断产生各种活性氧自由基(ROS),ROS容易损伤机体内的组织,从而引发各种各样的疾病。大量的研究表明,大部分从自然界物质中分离获得的多糖类化合物具有抑制脂质过氧化和清除自由基等抗氧化作用。通过本次实验,研究了黑木耳、香菇多糖的抗氧化活性。结果表明:黑木耳多糖、香菇多糖都具有一定的体外抗氧化能力,其中香菇多糖具有较强的清除羟基自由基(·OH)和1,1-二苯基-2-三硝基苯肼(DPPH·)的作用;并且黑木耳、香菇多糖也具有一定的体内抗氧化活性,均能不同程度的提高血清和肝脏中超氧化物歧化酶(SOD)活力以及谷胱甘肽过氧化物酶(GSH-Px)活力,可以减少肝脏和血清中丙二醛(MDA)含量,增强心脏过氧化物酶(POD)活力以及减少全脑单胺氧化酶(MAO)活力。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献
