Transmembrane potential of J774.2 mouse macrophage cells measured by microelectrode and ion distribution methods

The transmembrane potential (Em) of J774.2 macrophage cells measured by microelectrodes was -24.1 +/- 0.7 mV (mean +/- SEM). Em measured by lipophilic ion distribution was -35 +/- 2 mV or -40 +/- 2 mV, using a cation or anion, respectively. By any method, colchicine reduced Em by approximately 3 mV.     

3[H]-Sulpiride labels mesolimbic non-dopaminergic sites that bind antidepressant drugs

3[H]-(-)-Sulpiride and 3[H]-spiperone binding was compared in rat amygdala, nucleus accumbens and striatum, using (+/-)-sulpiride to define specific binding. 3[H]-(-)-Sulpiride bound to twice as many sites in amygdala and nucleus accumbens as 3[H]-spiperone. 3[H]-(-)-Sulpiride binding was directed to these additional sites by using 1 microM spiperone to mask dopaminergic binding. The binding of 3[H]-(-)-sulpiride to these sites was high affinity, reversible, Na+-dependent, but not stereospecific. Metoclopramide, tiapride and antidepressant medications, but not other neuroleptics, ADTN, or serotonin displaced 3[H]-(-)-sulpiride binding to these sites. These data suggest that 3[H]-(-)-sulpiride labels mesolimbic sites other than dopamine receptors which may mediate antidepressant effects.     

Induction of lymphomas by urethane in combination with diethylstilboestrol in CFLP mice

Protracted analgesia in young and adult rats maternally exposed to methadone

Summary The analgesic response to the hot-plate test was studied in 21-, 45-, 60-, 120- and 300-day-old rats maternally exposed to methadone (5 mg/kg). An elevation in nociceptive threshold, in the absence of further exposure to methadone, was observed in young and adult rats perinatally subjected to methadone.This research was supported by grant DA 01618.     

Potentiative effects of sulfhydryl compounds on carrageenin-induced oedema in rats and relationship to their potencies as inhibitors of angiostin-coverting enzyme in vivo.

Carrageenin-induced oedema in rats was potentiated by oral administration of (4R)-3-[(2S)-3-mercapto-2-methylpropanoyl]-4-thiazolidinecarboxylic acid (SA291) and related sulfhydryl compounds, and the effect was closely correlated with their potencies as inhibitors of angiotensin-converting enzyme in vivo.     

Influence of cyclic nucleotides on protein synthesis in vascular smooth muscle.

The incorporation of leucine-14C into protein in bovine mesenteric arteries was augmented by cyclic GMP (10-3 M) and decreased by cyclic AMP (10-3 M). There was no effect of 5'AMP (10-3 M). The phosphodiesterase inhibiting drugs theophylline (10-3 M) and papaverine (5 x 10-5 g/ml) both decreased the leucine-14C incorporation.     

Alpha 1-adrenergic stimulation of ketogenesis and fatty acid oxidation is associated with inhibition of lipogenesis in rat hepatocytes

The effect of norepinephrine on fatty acid synthesis (3H2O incorporation into fatty acids), on fatty acid oxidation to CO2 and on ketogenesis was studied in isolated hepatocytes of fed rats. After incubation with norepinephrine (50 microM), lipogenesis was lower (5.7 +/- 1.1 nmoles 3H2O incorporated into fatty acids/mg dry weight/30 min) than in controls (7.5 +/- 1.7; n = 6, p less than 0.02). In contrast, (1-14C) palmitate conversion into total ketone bodies was increased to 10.9 +/- 1.8 nmoles/mg/30 min with norepinephrine, vs 8.5 +/- 1.6 in controls (p less than 0.05), and more (1-14C) palmitate was converted to 14CO2 with norepinephrine than in controls (1.48 +/- 0.10 nmoles/mg/30 min vs 1.06 +/- 0.11, p less than 0.05). The inhibitory effect of norepinephrine on lipogenesis was abolished by addition of the alpha 1-receptor blocker prazosin, but not by alpha 2 or beta-blockers. The results demonstrate that the ketogenic effect of norepinephrine is coupled with an inhibitory effect on lipogenesis which may be explained by diminished activity of acetyl-CoA carboxylase, diminished formation of malonyl-CoA and decreased activity of carnitine palmitoyl transferase I.     

Bacterial lipopolysaccharide (LPS) enhances concanavalin A reactivity of thymocytes from the low-LPS-responder mouse strain C3H/Hej

Summary The capacity of LPS to enhance Con A reactivity of thymocytes was studied comparatively in the low-LPS-responder C3H/Hej mice and the high-LPS-responder CBA mice. The extent of synergism LPS+Con A was found similar in both strains.This work was supported by a grant from the INSERM (CRL: 76-5-101-1).     

Mechanical parameters determined in dispersed ventricular heart cells

A high-resolution laser diffraction system suitable for studying the basic mechanical properties of small contractile single cells has been developed. This method was used to establish the mechanical behavior of 95 ventricular cells isolated from adult guinea pig hearts. During contraction, the sarcomere length shortened from 1828 +/- 43 nm (mean +/- SD) to 1518 +/- 99 nm. The maximal velocities were 1.98 +/- 0.64 micron/s for shortening and 1.93 +/- 0.54 micron/s for re-lengthening. The twitch duration from 20% shortening to 80% re-lengthening was 622 +/- 120 ms.     

Sympathetic influence on cerebral blood volume: Time-dependent effect of decentralization of the superior cervical ganglia in mice

Zusammenfassung Neue Befunde über die Neuronale Regulation der Hirndurchblutung. Durchschneidung der präganglionären Fasern des Hals-Sy. verursachen eine starke Initiale und eine etwas weniger ausgeprägte dauerzunahme der Durchblutung, was durch Unterbechung der innervation und Sensitivierung der Rezeptoren erklärt wird.

Serum cholesterol levels in a Nigerian population sample

Serum cholesterol was determined in 104 healthy Nigerians (57 males and 47 females). The average cholesterol concentration was 187 +/- 72.8 mg/100 ml (mean +/- SD) with a range of 60-480 mg/100 ml. Cholesterol levels were higher in males than in females and increased with age. Distribution of cholesterol level was generally skew to the left.     

The effects of time of equilibration with cryoprotectants at 0 degree C prior to freezing on the survival of mouse embryos frozen by the two-step method

Mouse embryos were frozen by the two-step method after equilibration for 0.1-60 min with cryoprotectants at 0 degree C. No survival or a very low survival was obtained after equilibration for only 0.1 min. The morulae showed the highest survival rates when equilibration time was 5-30 min with 2 M DMSO, 20-30 min with 2 M glycerol, 5-10 min with 2 M ethylene glycol and 20-30 min with 2 M propylene glycol, respectively.     

Lipid accumulation by a cellulolytic strain of Aspergillus niger.

Lipid accumulation by a cellulolytic mold, Aspergillus niger, was studied. The amount of lipid accumulated ranged from 13.6-16.6% on various carbon sources, namely glucose, xylose, avicel (microcrystalline cellulose) and bagasse (a natural lignocellulosic substrate). Neutral lipids, phospholipids and glycolipids of the mycelia varied from 41.0-46.2%, 34.9-38.4% and 18.7-22.6% of total lipids, respectively. Unsaturated fatty acids comprised around 80% of total fatty materials with linoleic and oleic acid predominating. Of the four nitrogen sources tested, NH4Cl was the best source for lipid synthesis from cellulose (bagasse). Optimum temperature range for growth and lipid synthesis was 25-30 degrees C.     

Pharmacological studies of a new analgesic, dl-erythro-1-phenyl-2-(o-chlorophenyl)-2-[4-(p-methoxybenzyl)-1-piperazinyl] ethanol dihydrochloride, in experimental animals.

dl-Erythro-1-phenyl-2-(o-chlorophenyl)-2-[4-(p-methoxybenzyl)-1-piperazinyl] ethanol dihydrochloride showed orally a definite analgesic activity, without producing the significant morphine-like physical dependence liability, and its analgesic potency was about a half that of codeine and far superior to aminopyrine in experimental animals.

Induction of mouse hepatic ornithine decarboxylase by skin application of 12-0-tetradecanoylphorbol-13-acetate

Summary Topically applied 12-0-tetradecanoylphorbol-13-acetate on mouse skin was able to induce liver ornithine decarboxylase. Maximum induction occurred 10 h after a single application. Whereas no induction was noticeable at doses of 0.17 and 1.7 nmoles, 17 and 177 nmoles of 12-0-tetradecanoylphorbol-13-acetate caused about 30- and 57-fold increases respectively.This work was supported by NCI grants CA-09020, CA-07175 and P01-CA-22484.     

(S)-(+)-4,4,4-Trifluoro-3-(indole-3-)butyric acid,a novel fluorinated plant growth regulator

Racemic 4,4,4-trifluoro-3-(indole-3-)butyric acid (TFIBA) has been synthesized and shown to inhibitAvena coleoptile elongation. (S)-(+)-TFIBA (fig. 1), which was prepared by an enzymatic method and markedly promotes root growth of Chinese cabbage, lettuce and rice plants, is a novel fluorinated plant growth regulator. Activity of the (S)-(+)-enantiomer of TFIBA was 10-fold greater than that of the (R)-(–)-enantiomer in the first two plant species and 5-fold greater in rice.     

Occurrence of trichochromes in the urine of a melanoma patient

Summary The presence of two phaeomelanic pigments, trichochrome B and C, was demonstrated in the urine of a patient with malignant melanoma metastases.Supported by grants from the Swedish Cancer Society (No. 626-B75-04XA), 869-B75-01P, the Swedish Medical Research Council (No. B76-04X-00056-12), and the Walter, Ellen and Lennart Hesselman Foundation for Scientific Research. Age distribution of circulating alpha-interferon

A sensitive radioimmunoassay showed that circulating alpha-interferon in the plasma of healthy individuals was low in children and reached the highest level in the young adult, then declined gradually with age. Circulating alpha-interferon was 0.201 +/- 0.059 ng/ml in males (n = 19) and 0.184 +/- 0.076 ng/ml in females (n = 14) at ages 30-39 years old. It was noted that circulating alpha-interferon was maintained up to a certain level even in elderly individuals.
