Afferent activity has an important role in the formation of connections in the developing mammalian visual system. But the extent to which the activity of target neurons shapes patterns of afferent termination and synaptic contact is not known. In the ferret's visual pathway, retinal ganglion cell axons from each eye segregate early in development into eye-specific laminae in the lateral geniculate nucleus (LGN). The dorsal laminae (termed laminae A and A1) then segregate further into inner and outer sublaminae that retain input from on-centre and off-centre retinal axons, respectively. Thus, individual retinogeniculate axons form terminal arbors within laminae A and A1 that are restricted to one inner or outer sublamina. We report here that blockade of N-methyl-D-aspartate (NMDA) receptors on LGN cells with specific antagonists during the period of sublamina formation prevents retinal afferents from segregating into 'On' and 'Off' sublaminae. Retinogeniculate axons have arbors that are not restricted appropriately, or are restricted in size but inappropriately positioned within the eye-specific laminae. NMDA receptor antagonists may specifically disrupt a mechanism by which LGN neurons detect correlated afferent and target activity, and have been shown to reduce retinogeniculate transmission more generally, causing LGN cells to have markedly reduced levels of activity. These results therefore indicate that the activity of postsynaptic cells can significantly influence the patterning of inputs and the structure of presynaptic afferents during development.  相似文献

Monocular lid suture during the sensitive period early in the life of a kitten disrupts normal development of inputs from the two eyes to the visual cortex, causing a decrease in the fraction of cortical cells responding to the deprived eye. Such an ocular dominance shift has been assumed to depend on patterned visual experience, because no change in cortical physiology is produced by inequalities between the two eyes in retinal illumination or temporally modulated diffuse light stimulation. A higher-level process, involving gating signals from areas outside striate cortex, has been proposed to ensure that sustained changes in synaptic efficacy occur only in response to behaviourally significant visual inputs. To test whether such a process is necessary for ocular dominance plasticity, we treated 4-week-old kittens with visual deprivation and monocular tetrodotoxin (TTX) injections to create an imbalance in the electrical activities of the two retinas in the absence of patterned vision. After 1 week of treatment we determined the ocular dominance distribution of single units in primary visual cortex. In all kittens studied, a significant ocular dominance shift was found. In addition to this physiological change, there was an anatomical change in the lateral geniculate nucleus, where cells were larger in laminae receiving input from the more active eye. Our results indicate that patterned vision is not necessary for visual cortical plasticity, and that an imbalance in spontaneous retinal activity alone can produce a significant ocular dominance shift.  相似文献

When dissimilar images are presented to the two eyes, they compete for perceptual dominance so that each image is visible in turn for a few seconds while the other is suppressed. Such binocular rivalry is associated with relative suppression of local, eye-based representations that can also be modulated by high-level influences such as perceptual grouping. However, it is currently unclear how early in visual processing the suppression of eye-based signals can occur. Here we use high-resolution functional magnetic resonance imaging (fMRI) in conjunction with a new binocular rivalry stimulus to show that signals recorded from the human lateral geniculate nucleus (LGN) exhibit eye-specific suppression during rivalry. Regions of the LGN that show strong eye-preference independently show strongly reduced activity during binocular rivalry when the stimulus presented in their preferred eye is perceptually suppressed. The human LGN is thus the earliest stage of visual processing that reflects eye-specific dominance and suppression.  相似文献

To understand conscious vision, scientists must elucidate how the brain selects specific visual signals for awareness. When different monocular patterns are presented to the two eyes, they rival for conscious expression such that only one monocular image is perceived at a time. Controversy surrounds whether this binocular rivalry reflects neural competition among pattern representations or monocular channels. Here we show that rivalry arises from interocular competition, using functional magnetic resonance imaging of activity in a monocular region of primary visual cortex corresponding to the blind spot. This cortical region greatly prefers stimulation of the ipsilateral eye to that of the blind-spot eye. Subjects reported their dominant percept while viewing rivalrous orthogonal gratings in the visual location corresponding to the blind spot and its surround. As predicted by interocular rivalry, the monocular blind-spot representation was activated when the ipsilateral grating became perceptually dominant and suppressed when the blind-spot grating became dominant. These responses were as large as those observed during actual alternations between the gratings, indicating that rivalry may be fully resolved in monocular visual cortex. Our findings provide the first physiological evidence, to our knowledge, that interocular competition mediates binocular rivalry, and indicate that V1 may be important in the selection and expression of conscious visual information.  相似文献

The majority of neurones in the visual cortex of both adult cats and kittens can be excited by visual stimulation of either eye. Nevertheless, if one eye is deprived of patterned vision early in life, most cortical cells can only be activated by visual stimuli presented to the nondeprived eye and behaviourally the deprived eye is apparently useless. Although the consequences of monocular deprivation can be severe, they can in many circumstances be rapidly reversed with the early implementation of reverse occlusion which forces the use of the initially deprived eye. However, by itself reverse occlusion does not restore a normal distribution of cortical ocular dominance and only promotes visual recovery in one eye. In an effort to find a procedure that might restore good binocular vision, we have examined the effects on acuity and cortical ocular dominance of a short, but physiologically optimal period of reverse occlusion, followed by a period of binocular vision beginning at 7.5 weeks of age. Surprisingly, despite the early introduction of binocular vision, both eyes attained acuities that were only approximately 1/3 of normal acuity levels. Despite the severe bilateral amblyopia, cortical ocular dominance appeared similar to that of normal cats. This is the first demonstration of severe bilateral amblyopia following consecutive periods of monocular occlusion.  相似文献

Cytochrome oxidase (cytochrome c oxidase; ferrocytochrome c: oxygen oxidoreductase, EC 1.9.2.1) has been introduced as an oxidative metabolic marker for neurones in the central nervous system. Previous studies have shown that mature neurones remained sensitive to altered functional demands, and that both developing and adult neurones responded to sensory deprivation or deafferentation by reducing their cytochrome oxidase (Cyt. Ox.) activity. More recently, we showed that the blockage of retinal impulse transmission with tetrodotoxin led to a reversible reduction in Cyt. Ox. staining of affected lateral geniculate (LGN) and striate neurones in adult cats. The present study sought to extend these findings to adult monkeys, where Cyt. Ox. 'puffs' or 'blobs' are uniquely present in the visual cortex. We found that, while the retina remained histologically intact, with only moderate decreases in Cyt. Ox. staining of large ganglion cells and the two plexiform layers, subtle changes occurred in the LGN as early as 1 day post-tetrodotoxin injection, and clear reduction in enzyme levels was evident in both the LGN and the visual cortex by 3 days. Changes became progressively more severe up to 4 weeks post-injection. Within area 17, alternating bands of high and low Cyt. Ox. staining occurred in lamina IV, with alternating rows of dark and lightly reactive puffs superimposed in exact register. Thus, the mature visual neurones in the primate remain extremely sensitive to the cessation of retinal impulse transmission, and plastic metabolic changes occur through several synapses along the sensory pathway.  相似文献

The adult brain has a remarkable ability to adjust to changes in sensory input. Removal of afferent input to the somatosensory, auditory, motor or visual cortex results in a marked change of cortical topography. Changes in sensory activity can, over a period of months, alter receptive field size and cortical topography. Here we remove visual input by focal binocular retinal lesions and record from the same cortical sites before and within minutes after making the lesion and find immediate striking increases in receptive field size for cortical cells with receptive fields near the edge of the retinal scotoma. After a few months even the cortical areas that were initially silenced by the lesion recover visual activity, representing retinotopic loci surrounding the lesion. At the level of the lateral geniculate nucleus, which provides the visual input to the striate cortex, a large silent region remains. Furthermore, anatomical studies show that the spread of geniculocortical afferents is insufficient to account for the cortical recovery. The results indicate that the topographic reorganization within the cortex was largely due to synaptic changes intrinsic to the cortex, perhaps through the plexus of long-range horizontal connections.  相似文献

Sensory experience begins when neural circuits in the cerebral cortex are still immature; however, the contribution of experience to cortical maturation remains unclear. In the visual cortex, the selectivity of neurons for oriented stimuli at the time of eye opening is poor and increases dramatically after the onset of visual experience. Here we investigate whether visual experience has a significant role in the maturation of orientation selectivity and underlying cortical circuits using two forms of deprivation: dark rearing, which completely eliminates experience, and binocular lid suture, which alters the pattern of sensory driven activity. Orientation maps were present in dark-reared ferrets, but fully mature levels of tuning were never attained. In contrast, only rudimentary levels of orientation selectivity were observed in lid-sutured ferrets. Despite these differences, horizontal connections in both groups were less extensive and less clustered than normal, suggesting that long-range cortical processing is not essential for the expression of orientation selectivity, but may be needed for the full maturation of tuning. Thus, experience is beneficial or highly detrimental to cortical maturation, depending on the pattern of sensory driven activity.  相似文献

Binocular neurons in the visual cortex are thought to perform the first stage of processing for the fine stereoscopic depth discrimination exhibited by animals with frontally located eyes. Because lateral separation of the eyes gives a slightly different view to each eye, there are small variations in position (disparities), mainly along the horizontal dimension, between corresponding features in the two retinal images. The visual system uses these disparities to gauge depth. We studied neurons in the cat's visual cortex to determine whether the visual system uses the anisotropy in the range of horizontal and vertical disparities. We report here that there is a corresponding anisotropy in the cortical representation of binocular information: receptive-field profiles for left and right eyes are matched for cells that are tuned to horizontal orientations of image contours. For neurons tuned to vertical orientations, left and right receptive fields are predominantly dissimilar. Therefore, a major modification is required of the conventional notion of disparity processing. The modified scheme allows a unified encoding of monocular form and binocular disparity information.  相似文献

Although the influence of electrical activity on neural development has been studied extensively, experiments have only recently focused on the role of activity in the development of the mammalian central nervous system (CNS). Using tetrodotoxin (TTX) to abolish sodium-mediated action potentials, studies on the visual system show that impulse activity is essential both for the normal development of neuronal size and responsivity in the lateral geniculate nucleus (LGN), and for the eye-specific segregation of geniculo-cortical axons. There have been no anatomical studies to investigate the influence of action potentials on CNS synaptic development. We report here the first direct evidence that elimination of action potentials in the mammalian CNS blocks the growth of developing axon terminals and the formation of normal adult synaptic patterns. Our results show that when TTX is used to eliminate retinal ganglion-cell action potentials in the cat from birth to 8 weeks, the connections made by ganglion cell axons with LGN neurones, retinogeniculate synapses, remain almost identical morphologically to those in the newborn kitten.  相似文献

The cellular mechanisms by which the axons of individual neurons achieve their precise terminal branching patterns are poorly understood. In the visual system of adult cats, retinal ganglion cell axons from each eye form narrow cylindrical terminal arborizations restricted to alternate non-overlapping layers within the lateral geniculate nucleus (LGN). During prenatal development, axon arborizations from the two eyes are initially simple in shape and are intermixed with each other; they then gradually segregate to form complex adult-like arborizations in separate eye-specific layers by birth. Here we report that ganglion cell axons exposed to tetrodotoxin (TTX) to block neuronal activity during fetal life fail to form the normal pattern of terminal arborization. Individual TTX-treated axon arborizations are not stunted in their growth, but instead produce abnormally widespread terminal arborizations which extend across the equivalent of approximately two eye-specific layers. These observations suggest that during fetal development of the central nervous system, the formation of morphologically appropriate and correctly located axon terminal arborizations within targets is brought about by an activity-dependent process.  相似文献

Visual information from the retina is transmitted to the cerebral cortex by way of the lateral geniculate nucleus (LGN) in the thalamus. In primates, most of the retinal ganglion cells that project to the LGN belong to one of two classes, P and M, whose axons terminate in the parvocellular or magnocellular subdivisions of the LGN. These cell classes give rise to two channels that have been distinguished anatomically, physiologically and behaviourally. The visual cortex also can be subdivided into two pathways, one specialized for motion processing and the other for colour and form information. Several lines of indirect evidence have suggested a close correspondence between the subcortical and cortical pathways, such that the M channel provides input to the motion pathway and the P channel drives the colour/form pathway. This hypothesis was tested directly by selectively inactivating either the magnocellular or parvocellular subdivision of the LGN and recording the effects on visual responses in the cortex. We have previously reported that, in accordance with the hypothesis, responses in the motion pathway in the cortex depend primarily on magnocellular LGN. We now report that in the colour/form pathway, visual responses depend on both P and M input. These results argue against a simple correspondence between the subcortical and cortical pathways.  相似文献

The massive visual input from the eye to the brain requires selective processing of some visual information at the expense of other information, a process referred to as visual attention. Increases in the responses of visual neurons with attention have been extensively studied along the visual processing streams in monkey cerebral cortex, from primary visual areas to parietal and frontal cortex. Here we show, by recording neurons in attending macaque monkeys (Macaca mulatta), that attention modulates visual signals before they even reach cortex by increasing responses of both magnocellular and parvocellular neurons in the first relay between retina and cortex, the lateral geniculate nucleus (LGN). At the same time, attention decreases neuronal responses in the adjacent thalamic reticular nucleus (TRN). Crick argued for such modulation of the LGN by observing that it is inhibited by the TRN, and suggested that "if the thalamus is the gateway to the cortex, the reticular complex might be described as the guardian of the gateway", a reciprocal relationship we now show to be more than just hypothesis. The reciprocal modulation in LGN and TRN appears only during the initial visual response, but the modulation of LGN reappears later in the response, suggesting separate early and late sources of attentional modulation in LGN.  相似文献

To investigate the effects of short-term intraocular pressure (IOP) elevation on the receptive field properties of lateral geniculate nucleus (LGN) cells, responses of the LGN cells to annulus, disc and drifting gratings with high or low spatial frequencies have been recorded extracellularly in the eat with the retinal perfusion pressure kept stable (30mmHg). Our results indicated that the responses of the X and Y type LGN cells were significantly weakened during IOP elevation. And the responses varied with the different mechanisms of receptive fields. Specifically, while using annulus and disc as stimuli, the responses of Y cells were more tolerant than X cells to lOP elevation. The surround area of the receptive field was more sensitive to lOP elevation than the center. The mean responses during IOP elevation decreased more than the peak responses did. lOP elevation has more influence on the responses of X cells than on the response of Y cells to the drifting gratings with high spatial frequency. These results may reflect different degrees of ischemia on corresponding retinal structures caused by IOP elevation.  相似文献

The classical neuro-ophthalmologic literature describes the organization of the primate's optic tract as containing a single topographic representation of the complete contralateral visual hemifield. In contrast, cats have separate visual field representations for the optic axons of the functionally distinct retinal ganglion cell classes. As the line of decussation for each ganglion cell class in the cat occupies a different location on the retinal surface, whereas in primates they are all superimposed, such a species difference might be expected. We report that implants of horseradish peroxidase placed in either the deep or superficial extremes of the monkey's optic tract produce retrograde labelling of distinct retinal ganglion cell classes, and produce anterograde labelling confined to distinct laminae of the lateral geniculate nucleus. Hence, the optic tract of the primate cannot contain a single representation of the contralateral visual hemifield; rather, independent visual field representations for the functionally distinct optic axons must exist. Their anatomical segregation may account for the clinical observation of selective impairments of distinct visual abilities following partial interruption of the optic tract in man.  相似文献

A hallmark of mammalian brain evolution is the disproportionate increase in neocortical size as compared with subcortical structures. Because primary visual cortex (V1) is the most thoroughly understood cortical region, the visual system provides an excellent model in which to investigate the evolutionary expansion of neocortex. I have compared the numbers of neurons in the visual thalamus (lateral geniculate nucleus; LGN) and area V1 across primate species. Here I find that the number of V1 neurons increases as the 3/2 power of the number of LGN neurons. As a consequence of this scaling law, the human, for example, uses four times as many V1 neurons per LGN neuron (356) to process visual information as does a tarsier (87). I argue that the 3/2 power relationship is a natural consequence of the organization of V1, together with the requirement that spatial resolution in V1 should parallel the maximum resolution provided by the LGN. The additional observation that thalamus/neocortex follows the same evolutionary scaling law as LGN/V1 may suggest that neocortex generally conforms to the same organizational principle as V1.  相似文献

When the two eyes are confronted with sufficiently different versions of the visual environment, one or the other eye dominates perception in alternation. A similar situation may be created in the laboratory by presenting images to the left and right eyes which differ in orientation or colour. Although perception is dominated by one eye during rivalry, there are a number of instances in which visual processes nevertheless continue to integrate information from the suppressed eye. For example the interocular transfer of the motion after-effect is undiminished when induced during binocular rivalry. Thus motion information processing may occur in parallel with the rivalry process. Here we describe a novel example in which the visual system simultaneously exhibits binocular rivalry and vision that integrates signals from both eyes. This apparent contradiction is resolved by postulating parallel visual processes devoted to the analyses of colour and motion information. Counterphased gratings are viewed dichoptically such that for one eye the grating is composed of alternating yellow and black stripes (luminance) while for the other it is composed of alternating red and green stripes (chrominance). When the gratings are fused, a moving grating is perceived. A consistent direction of motion can only be achieved if left and right monocular signals are integrated by the nervous system. Yet the apparent colour of the binocular percept alternates between red-green and yellow-black. These observations demonstrate the segregation of processing by the early motion system from that affording the perception of colour. Although, in this stimulus, colour information in itself can play no part in the cyclopean perception of motion direction, colour is carried along perceptually (filled in) by the moving pattern which is integrated from both eyes.  相似文献

In the visual cortex of mammals, response properties of single neurons can be changed by restricted visual experience during early postnatal development. Covering one eye for four to eight hours when kittens are at the peak of the sensitive period is sufficient to weaken the influence of the occluded eye on cortical neurons resulting in a noticeable shift of ocular dominance towards the open eye. The underlying changes in synaptic connections do not occur so readily when a kitten is anaesthetized and paralysed. We report here that an ocular dominance shift is prevented in alert kittens that receive repeated brief monocular exposures when these are followed by ketamine-xylazine anaesthesia. This retrograde effect on cortical plasticity suggests that the process by which synaptic activity is converted into structural changes has been disturbed.  相似文献

Systematic changes in neuronal connections have been observed during the development of many vertebrate neuronal systems. These changes have usually involved a refinement from an initial exuberance of connections or a response to some experimental perturbation. Here we report on a system of neuronal connections, which, during a protracted developmental period, undergo ordered changes in response to normally occurring changes in functional requirements. In the frog Xenopus laevis, interocular alignment changes markedly during late larval and post-metamorphic life, producing a progressive enlargement of the binocular portion of the visual field. An intertectal system links the two mid-brain optic tecta and is concerned with the neural representation of binocular visual space. In the adult animal, connections in this system link corresponding points (points receiving information from one locus of binocular visual space) on the two tecta. Changes in eye position with development, however, change the set of corresponding points. Therefore, if the intertectal connections link corresponding tectal points throughout development, they must undergo an ordered change with time. We present electrophysiological evidence that the intertectal connections do, indeed, undergo such changes in response to changes in eye alignment, and that the changes are major.  相似文献

Retinal ganglion cells are the projection neurons that link the retina to the brain. Peptide immunoreactive cells in the ganglion cell layer (GCL) of the mammalian retina have been noted but their identity has not been determined. We now report that, in the rabbit, 25-35% of all retinal ganglion cells contain substance P-like (SP) immunoreactivity. They were identified by either retrograde transport of fluorescent tracers injected into the superior colliculus, or by retrograde degeneration after optic nerve section. SP immunoreactive cells are present in all parts of the retina and have medium to large cell bodies with dendrites that ramify extensively in the proximal inner plexiform layer. Their axons terminate in the dorsal lateral geniculate nucleus, superior colliculus and accessory optic nuclei, and these terminals disappear completely after contralateral optic nerve section and/or eye enucleation. In the dorsal lateral geniculate nucleus large, beaded, immunoreactive axons and varicosities make up a narrow plexus just below the optic tract, where they define a new geniculate lamina. The varicosities make multiple synaptic contacts with dendrites of dorsal lateral geniculate nucleus projection neurons and presumptive interneurons in complex glomerular neuropil. This is direct evidence that some mammalian retinal ganglion cells contain substance P-like peptides and strongly suggests that, in the rabbit, substance P (or related tachykinins) may be a transmitter or modulator in a specific population or populations of retinal ganglion cells.  相似文献
