Treatment of neonatal mice with an antiviral factor, (AVF), obtained from the leaves of Melia azedarach L. protected them against lethal encephalitis caused by Tacaribe virus inoculation. The degree of protection obtained varied from 66% to 100% depending on the virus dose. Similarly, administration of AVF to nursing mothers protected their offspring from developing virus encephalitis. AVF does not directly inactivate Tacaribe virus; it inhibits an early step (s) in the replication process in cell cultures.  相似文献

A diterpenoid, totarol (1), fromPodocarpus nagi was evaluated as an antioxidant. This diterpenoid inhibited autoxidation of linoleic acid. Mitochondrial and microsomal lipid peroxidation induced by Fe(III)-ADP/NADH or Fe(III)-ADP/NADPH were also inhibited. Nagilactone E (2), a norditerpene lactone isolated from the same source, had no antioxidative activity. Furthermore, totarol protected red cells against oxidative hemolysis. This diterpene was shown to be effective in protecting biological systems against oxidative stresses.  相似文献

The effectiveness of complement-mediated killing ofBorrelia burgdorferi, the causative agent of Lyme disease, in the presence of host-derived tissues was studied. Second and high passage forms ofB. burgdorferi 297 isolate were grown in a LEW/N rat joint tissue co-culture system and in artificial BSK medium. Guinea pig complement and third week immune serum from hamsters with experimental Lyme disease were added to the cultures. Both high and low passage borrelia grown in BSK medium died and did not revive after 3 weeks incubation in BSK medium. However, 5–12% of tissue co-cultured borrelia survived the first complement-mediated lysis. Repeated re-growth and lysis cycles in tissue co-culture resulted in isolation of an 85% complement-resistant population ofB. burgdorferi. Joint tissue culture supernatant collected on the third day of tissue culture, and fibronectin (25 g/ml), also protected spirochetes from complement-mediated lysis in contrast to BSK or fresh co-culture medium. Complement-mediated lysis may not be an effective mechanism in eradication of borrelia, and the chronicity of Lyme disease may be due to resistance ofB. burgdorferi variants to host immune defense mechanisms in the presence of host-derived tissues.  相似文献

The vectors of arthropod-borne viruses (arboviruses) become infected by feeding on the viraemic blood of an infected animal. This theory is based on transmission studies involving artificial infection of vertebrate hosts by syringe inoculation. To reproduce natural conditions of virus transmission, infected and uninfected vectors (ticks) of tick-borne encephalitis virus, the most important arbovirus in Europe, were allowed to feed together on uninfected wild vertebrate hosts. The greatest numbers of infected ticks were obtained from susceptible host species that had undetectable or very low levels of viraemia. The results suggest that nonviremic transmission is an important mechanism for the survival of certain arboviruses in nature.  相似文献

The antiviral activity of Shigyaku-to (TJS-109), a traditional Chinese herbal medicine, was investigated in mice infected with herpes simplex virus type 1 (HSV-1). TJS-109 is a combination of the medicinal plant extracts fromZingiberis siccatum rhizoma,Aconiti tuber andGlycyrrhizae radix in a specific proportion. Mice infected with a 10 LD50 dose of HSV-1 were treated with TJS-109 orally at doses of 1.25 to 20 mg/kg 2 days before, and 1 and 4 days after the infection. The treated groups had 80% (1.25 mg/kg), 40% (5 mg/kg) and 23% (20 mg/kg) mortality rates 25 days after the infection as compared with a 100% mortality rate in control mice treated with saline. When HSV-1 infected mice (recipients) received CD8+T cell fractions derived from spleens of mice treated with TJS-109 (donors), 70% of recipients survived, as compared with 0% survivors in the groups of mice treated with saline, B cell fractions, CD4+ T cell fractions or macrophage-enriched fractions prepared from the same donors. TJS-109 did not show any virucidal activities against HSV-1 or any virostatic activities on the growth of HSV-1 in Vero cells. These results suggest that TJS-109 protected mice exposed to lethal amounts of HSV-1 through the activation of CD8+ T cells.  相似文献

Summary The inhibitory effect of four flavonoid compounds on virus multiplication and their influence on the intracellular cyclic AMP (cAMP) level were studied in cell cultures. Quercetin and quercitrin reduced the yields ofHuman (alpha) herpesvirus 1 (HSV-1) andSuid (alpha) herpesvirus 1 (pseudorabies virus), but hesperidin and rutin had no effect. Further, quercetin and quercitrin elevated the intracellular level of cAMP, whereas hesperidin and rutin did not alter the cAMP level. Both antiviral activity and cAMP-enhancing effect were dependent on the concentrations of the flavonoids, and these effects turned out to be parallel.This study suggests that a relation exists between the antiviral effect and the cAMP-enhancing activity of flavonoids.  相似文献

When infested by herbivorous mites, cotton seedlings produce volatile cues that elicit attraction of predatory mites. Experiments were carried out to elucidate how downwinduninfested conspecific seedlings are affected by these volatiles. It was found that the rate of oviposition of herbivorous mites was reduced on seedlings exposed to volatiles from infested seedlings. Moreover, predatory mites were attracted by exposeduninfested seedlings. These results strongly suggest that uninfested plants are better protected against herbivore attack when exposed to airborne chemicals released by their infested neighbours.  相似文献

Malaria, caused by members of the genusPlasmodia, is still the most prevalent parasitic disease in the world. In an attempt to understand genetic factors conferring resistance to malaria, mouse models of thalassemia, sickle trait, and ankyrin and spectrin deficiency were studied during infection with species of malaria infectious to rodents. Although growth ofP. falciparum is not inhibited in thalassemic erythrocytes in culture, mice carrying a -thalassemia mutation were protected fromPlasmodium chabaudi adami, supporting epidemiologic findings. Transgenic mice expressing s hemoglobin were also significantly protected from two species of rodent malaria. Importantly, a significant role for the spleen in protection in the s transgenic mice was found. Finally, mice deficient in spectrin and ankyrin were studied with respect to their ability to support the growth of malaria. It was found that spectrin deficient mice were almost completely refractory toP. chabaudi adami andP. berghei. These models will allow further study of host factors in resistance to malaria.  相似文献

Summary The lipid fraction, 17%, of air-driedPlexaura homomalla formakükenthali, collected in the Caribbean at Puerto Rico, yielded 50% of its weight as the mammalian prostaglandin (15S)-PGA2 methyl ester. The freeze-dried gorgonian yielded (15S)-PGA2 largely as the acetate of the methyl ester. (15S)-PGA2 was also obtained from material collected at St. Croix and at South Caicos. Field observations indicate thatPlexaura kükenthali may be a species separate fromPlexaura homomalla and that it is abundant on some shallow water reefs in the Caribbean. Prostaglandins could not be detected in the lipid fraction of eggs isolated fromPlexaura homomalla (Esper) formahomomalla.Acknowledgments. Summary Three isolations of a virus of the Hughes group were obtained from seabird ectoparasites,Ornithodoros (Alectorobius) maritimus, on Great Saltee Island, Ireland. The agent is closely related to Soldado virus, originally obtained from related ticks near Trinidad, West Indies, and represents the second recorded tickborne arbovirus in Ireland.These investigations were supported in part by the Royal Irish Academy (Praeger Fund).  相似文献

Summary The effect of cold or isolation stress on mortality rate and brain virus level were investigated in mice infected with West Nile virus (WNV). Exposure of mice for 5 min/day to cold water (1±0.5°C) for 8–10 days resulted in 92% mortality as compared to 47% in control mice (p<0.001). Mice housed in individual cages (isolation stress) were also more susceptible to WN viral infection, as shown by increased mortality rate reaching 85% as compared to 50% in mice housed 6 per cage (p<0.01). Cold or isolation stress increased blood brain and spleen virus levels as early as 2 days after inoculation. After 8 days of isolation or cold stress, mice inoculated with WNV had 8.9 and 9.0 log10 plaque forming units in the brain, respectively, as compared to 6.9 in the control (p<0.01–0.001). Furthermore, lymphoid organs such as spleen and thymus showed severe mass loss. These data suggest that physical or non-physical stress situations enhance WNV encephalitis by accelerating virus proliferation and increase mortality in mice.  相似文献

Summary Monarch butterflies sequester cardenolides from their larval host plants in the milkweed genusAsclepias for use in defense against predation. Of 108Asclepias species in North America, monarchs are known to feed as larvae on 27. Research on 11 of these has shown that monarchs sequester cardenolides most effectively, to an asymptote of approximately 350 g/0.1 g dry butterfly, from plants with intermediate cardenolide contents rather than from those with very high or very low cardenolide contents. SinceAsclepias host plant species are distributed widely in space and time across the continent, monarchs exploit them by migration between breeding and overwintering areas. After overwintering in central Mexico, spring migrants east of the Rocky Mountains exploit three predominantAsclepias species in the southern USA that have moderately high cardenolide contents. Monarchs sequester cardenolides very effectively from these species. First generation butterflies are thus well protected against predators and continue the migration north. Across the northern USA and southern Canada most summer breeding occurs on a fourthAsclepias species and in autumn most of these monarchs migrate back to Mexican overwintering sites. The ecological implications of this cycle of cardenolide sequestration for the evolution of monarch migration are discussed.  相似文献

Summary The herbicide paraquat (1,1-dimethyl-4,4-bipyridylium ion) has been found to exert a growth inhibitory effect on the N2-fixing blue-green algaNostoc muscorum in nitrogen-free (N2) and NO 3 media, without any apparent inhibitory or stimulatory effect on the heterocyst-forming capacity of the organism. With a dose of paraquat permitting about 20 and 50% survival of this alga a reverse mutation (fromhet + nif auxotrophy tohet + nif + prototrophy), a forward mutation (for L-methionine-DL-sulfoximine [MSO]-resistance), and an auxotrophic mutation (for carbon-auxotrophy through methylamine [MA]-resistance) have been obtained. The toxic and mutagenic effects of this agrochemical have been compared with those of the well known mutagen MNNG (N-methyl-N-nitro-N-nitrosoguanidine) and found to be stronger than those of the latter in each case.The author wishes to thank Mr Girish Nath, Lakshami Chemicals Pvt. Summary Males ofPodisus fretus (Hemiptera: Pentatomidae) release a long-range attractant pheromone containing linalool (49.0%), (E)-2-hexenal (34.5%), benzyl alcohol (12.0%), nerolidol (2.0%),-terpineol (1.1%), and traces of several other compounds. The eastern yellowjacket,Vespula maculifrons (Hymenoptera: Vespidae), is attracted to artificial pheromones forP. fretus and for the sympatric species,Podisus maculiventris.The authors thank Mr T.J. Insect-transmitted plant pathogens can have positive, negative or no effects on the vector insect. Effects could be direct (caused by the pathogen) or indirect (mediated by changes in the plant). Pathogen effects on non-vector insects are less well known.Peregrinus maidis (Ashmead) (Homoptera: Delphacidae), an insect that is not a vector of the corn stunt spiroplasma (CSS), weighed less 15 days after feeding on CSS-infected corn plants relative to insects feeding on healthy corn plants. Fecundity of non-vector insects that were removed from diseased plants was higher than for insects coming from healthy plants. ForDalbulus maidis (DeLong & Wolcott) (Homoptera: Cicadellidae), an insect that transmits CSS, there were no differences in weight, longevity, eggs per day, weekly or total fecundity after feeding on healthy or on CSS-infected corn plants. Significant differences in some phloem amino acids were detected between healthy and CSS-infected plants. Infected plants also showed an increased phloem acidity as disease symptoms progressed. Feeding on CSS-infected corn plants by an insect that does not vector the pathogen infecting the plant can have favorable consequences as evidenced by an increased fecundity.Scientific aritcle no. 8819, contribution no. Summary A naturally regenerated young-growth in the Buchser Hochwald area in the Canton of St. Gallen, Switzerland, was used to determine the silviculturally permissible browsing limit for sycamore maple (Acer pseudoplatanus). The investigated area is situated at an altitude ranging from 1280 to 1310 m a.s.l. on anAbieti-Fagetum typicum site. Browsing was done by red deer (Cervus elaphus) and roe deer (Capreolus capreolus). A total of 57 sycamore maples, 1.30 m high, were examined. The actual browse impact on these plants was assessed by determining the frequency of browsing marks on the stem axis. To this end, the plants were cut into sections of 5 cm each and then split radially. On this basis it was possible to calculate permissible shares of browsed plants for four different size categories between 0.10 and 1.30 m. The permissible share of plants with two or more visible browsing marks on the stem axis amounted to 37.8% as an average value for the total risk period. This corresponds to a quota of 26.9% browsed terminal shoots per annum.  相似文献

Summary The effect of 750 second chromosomes ofDrosophila melanogaster on viability was studied. 19.3% of them proved letal or semilethal (=drastics) in homozygous condition. Compared to data obtained in previous years at the same sampling site, a significant frequency decrease of drastics during the past decade could be observed. The dynamic processes taking place in the Korean wild populations ofD. melanogaster are discussed.Acknowledgment.
