本文报道了溶菌酶水溶液的激光拉曼谱。酰胺Ⅰ振动谱带在1657cm~(-1),酰胺Ⅱ振动谱带在1302和1253cm~(-1),酪氨酸残基的特征谱带在854和837cm~(-1)。硫—硫桥键的振动谱带在505cm~(-1)。并由此定性地证明了该蛋白质分子的肽链主要由α—螺旋和无规则卷曲组成,酪氨酸残基在分子中呈三分之二“暴露”式,硫—硫键部位的几何构型为扭式—扭式—扭式。  相似文献

对丙酮处理酵母菌吸附铅前后的红外光谱进行了分析比较。生物材料的红外光谱图主要由蛋白质的吸收带、碳水化合物的吸收带组成。1652cm^-1、1534cm^-1、1239cm^-1处的吸收峰是蛋白质的三个特征吸收峰。1455cm^-1处的吸收峰为CH3和CH2的弯曲振动峰;1397cm^-1处中等强度的吸收归属于羧基的对称伸缩振动。1200-940cm^-1之间出现的强且复杂的谱带归属于醚和碳水化合物中的C-O伸缩振动。915cm^-1、890cm^-1处两个弱的吸收峰属于细胞壁中多糖的糖环键的振动。吸附pb^2 后,3375cm^-1处的吸收峰紫移至3388cm^-1,1398cm^-1处的峰红移到1384cm^-1,说明羟基和羧基在结合pb^2 起重要作用丙酮处理酵母菌的主要成份和结构吸附前后保持完整。  相似文献

利用紫外可见光谱和红外光谱研究了Ag(Ⅰ)与溶菌酶之间的相互作用和酶微观结构的变化.紫外可见光谱测试表明Ag+与溶菌酶肽链上的C=O存在相互作用,红外光谱测试表明Ag+与溶菌酶结合位点可能为—OH和—NH基团,利用二阶导、自退卷积和谱线拟合技术对溶菌酶红外谱图的酰胺Ⅰ带进行处理并推测其二级结构变化,结果表明:α-螺旋二级结构含量降低,β-折叠二级结构含量增加,与Ag+作用后,溶菌酶的活性没有明显改变.  相似文献

分析了绿色木霉产的纤维二糖酶在pH 5.0和pH 2.0下的拉曼光谱图.实验结果表明:纤维二糖酶在pH 5.0时具有酶活功能,蛋白主链结构主要为α-螺旋和无规则卷曲,侧链中酪氨酸和色氨酸均为"埋藏式";纤维二糖酶在pH 2.0时无酶活,酶蛋白主链结构主要为α-螺旋,而无规则卷曲发生较大变化,铡链中的酪氨酸和色氨酸均为"暴露式".根据光谱特征分析,该纤维二糖酶中可能不舍二硫键.  相似文献

利用从头算方法探索蛋白质模型分子——丙氨酸二肽在氯仿溶剂环境下的二级结构布居特征.借助对分子结构敏感的振动探针(酰胺振动吸收带)的光谱表象,寻求振动光谱参数与分子结构之间的联系.研究结果表明,丙氨酸二肽分子倾向于形成β折叠、PPⅡ、C7eq和C5等构型.简正模式分析结果表明分子内酰胺振动带对分子骨架变化十分敏感,因此可以将酰胺振动吸收带作为结构探针研究蛋白质分子的二级结构信息.  相似文献

花生、大豆种子中的SOD粗提液经聚丙烯酰胺凝胶电泳及NBT活性染色,结果表明,花生种子有6条SOD同工酶谱带,其中1条为Mn-SOD,其余为Cu,Zn-SOD.大豆种子有8条SOD同工酶谱带,其中3条为Mn-SOD,其余为Cu,Zn-SOD.不同pH值对SOD活性有一定的影响,pH=1.0时,Mn-SOD同工酶谱带消失;pH=12.0时,全部SOD同工酶谱带消失.变性剂DTT、SDS和β-巯基乙醇对SOD同工酶有较大影响,SDS使Mn-SOD谱带丢失,DTT、β-巯基乙醇使全部SOD同工酶谱带消失.  相似文献

在pH5.0的缓冲溶液中,叶酸与酪氨酸、色氨酸和苯丙氨酸3种芳香族氨基酸反应,使得上述芳香族氨基酸的荧光发生猝灭,最大猝灭波长分别位于304 nm(酪氨酸体系),353 nm(色氨酸体系),284 nm(苯丙氨酸体系),芳香族氨基酸的荧光猝灭值在一定范围内与叶酸浓度成正比.由此,建立荧光猝灭法测定叶酸的新方法,该方法的线性范围和检出限分别为:1.0× 10-5-1.0×10-2 g/L,5.2×10-6 g/L(酪氨酸体系);1.0×10-5-1.5 ×10-2 g/L,5.6×10-6 g/L(色氨酸体系);5.0×10-6-8.0×10-3g/L,2.9×10-6 g/L(苯丙氨酸体系).该方法已应用于实际样品的测定,并对荧光猝灭的机理进行讨论.  相似文献

通过对栗黄枯叶蛾核型多角体病毒TvNPV(Trabala vishnou Nuclear Po-lyhedrosis Virus)的病毒粒子的拉曼光谱研究,讨论了它的蛋白质分子空间构象.其蛋白质主要为β折叠结构、α螺旋结构以及少量无规卷曲(其中包括β回折)结构.蛋白质中酪氨酸残基多数暴露于分子表面,少数埋藏在分子内部;色氨酸残基大多埋藏在疏水环境中.蛋白质分子侧链C—C—S—S—C—C构型为反式—扭曲—反式.  相似文献

合成了3μm无孔脲醛树脂-二氧化锆(UF-ZrO2)复合微球,并用扫描电子显微镜对其进行表征.利用基体表面上的酰胺键与丙烯酸甲酯和乙二胺进行三次马氏加成反应和胺化反应,形成末端为胺基的聚胺基酰胺星型树状间隔臂.再用咪唑法将牛血清白蛋白(BSA)对其表面进行修饰,制成BSA-UF-ZrO2固相萃取剂.以溶菌酶和卵清蛋白作为测试蛋白,分别考察了时间,pH值,洗脱试剂及其浓度对BSA-UF-ZrO2固相萃取剂吸附量的影响.结果表明,溶菌酶和卵清蛋白最佳吸附时间是4 h,缓冲溶液的pH值是6.0和5.0,最佳洗脱试剂是0.0625 mol.mL-1的KSCN(溶菌酶)和0.1250 mol.mL-1的KSCN(卵清蛋白).  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献
