利用PCR方法,将羊生长激素(oGH)成熟蛋白编码序列扩增,再将扩增产物接入分泌型表达载体pET12a(SalI位点)中,利用宿主菌自身的OmpT信号肽引导羊生长激素在E.coli BL21(DE3)中分泌,获得正向插入重组质粒pET12-oGH8,对重组子及对照菌进行诱导表达,薄层色谱扫描(SDS-PAGE)检测结果发现,oGH在OmpT信号肽引导下未见明显分泌,推测oGH自身的氨基酸序列可有是  相似文献

利用PCR方法扩增得到枯草杆菌本身的表达调控序列SP(含有信号肽)和Pro序列(包含信号肽和前肽),分别与血小板生成素(TPO)结构基因及枯草杆菌载体片段连接,构建两个重组TPO枯草杆菌表达载体,转化受体菌,得到DB403(SP-TPO)和DB403(Pro-TPO)重组克隆,比较有无前肽对其表达分泌的作用.用ELISA和Western Blot检测TPO的表达分泌,发现DB403(Pro-TPO)能够表达分泌TPO,而DB403(SP-TPO)未发现TPO的分泌.对小鼠腹腔注射DB403(Pro-TPO)发酵浓缩液,发现能够明显增加血小板数目,与自身对照相比增加84.2%,与生理盐水对照比较增加99.7%.以上实验说明Pro-TPO能够表达分泌有明显活性的TPO蛋白,而且前肽对外源蛋白的分泌是必要的.  相似文献

以野生型枯草芽孢杆菌基因组DNA为模板,PCR扩增获得带有SD序列及谷氨酸棒状杆菌信号肽ΔS0949的BTG基因.将其与大肠杆菌-谷氨酸棒状杆菌穿梭表达载体pXMJ19连接,构建重组质粒pXMJ19-Sbtg转化谷氨酸棒状杆菌ATCC13032.经IPTG诱导后该重组菌发酵液具有交联酪蛋白的能力,表明该重组菌能够实现分泌表达.  相似文献

为获得人A组轮状病毒重组结构蛋白VP7及其高效价的卵黄抗体,使用PCR技术扩增VP7 DNA片段并将其克隆到原核表达载体pET28a上形成重组质粒pET28a-VP7。将重组质粒转化基因工程菌E.Coil BL21(DE3),用0.5 mM IPTG诱导VP7重组蛋白的表达。通过包涵体纯化和蛋白复性后,将VP7重组蛋白与弗氏佐剂混匀免疫产蛋鸡,收集鸡蛋。用水稀释-盐析法纯化卵黄抗体IgY。进一步应用SDS-PAGE、ELISA和点杂交技术,分析该卵黄抗体的纯度、效价和特异性。结果表明在基因工程菌E.coli BL21(DE3)中能够实现VP7重组抗原的表达,将重组抗原VP7免疫产蛋鸡,提取纯化得到滴度为1:100 000的抗VP7卵黄抗体,且特异性的识别野生型A组轮状病毒。  相似文献

采用PCR法从含有人可溶性BAFF基因的pET30a( )扩增得到人sBAFF基因,通过rPCR将抗菌肽Cecropin CM4基因的2条单核苷酸链进行扩增得到CM4基因,再采用over-lap PCR法通过linker将hsBAFF与Cecropin CM4融合基因相连接.经纯化和鉴定后,定向插入到原核表达载体pET30a( )中,然后转化E.coliBL21(DE3),通过实验确定了表达该融合基因的最佳诱导条件:IPTG终浓度为1.0 mmol/L,诱导时间为5 h.温度为30℃,其表达量占全菌蛋白的40%.表达产物经SDS-PAGE分析,得到相对分子质量约为22 000的重组蛋白并且存在超声裂解后的上清中.重组蛋白经Western blot检测,结果显示重组蛋白可被鼠抗人可溶性BAFF的抗体识别.采用分子筛Sephadex G-75对重组融合蛋白进行纯化,并经SDS-PAGE对其鉴定.通过对其生物学功能的检测得知,纯化后的重组融合蛋白对大肠杆菌K12D31和真菌有明显的抑菌能力.  相似文献

利用PCR技术从宁夏地方三黄肉鸡肝组织中扩增出α干扰素基因,构建成原核重组表达质粒pET-28a/(ChIFN-α从埃希氏菌属大肠杆菌(Escherichia coli)中扩增出L-天(门)冬酰胺酶Ⅱ(L-Asmraginase,ASP)信号肽基因,并与原核重组质粒pET-28a/ChIFN-α连接,构建成分泌型表达重组质粒pET-28a/ASP-ChIFN-α将重组质粒经IPTG诱导5 h.用SDS-PAGE分析表达蛋白含量,并用鸡α-干扰素ELISA(Western-blot)试剂盒检测表达的重组蛋白的特异性.结果表明,包涵体型重组质粒pET-28a/ChIFN-α和分泌型重组质粒pET-28a/ASP-ChIFN-α在23 kD处均表达出目的蛋白,蛋白含量分别为27%和38%,pET-28a/ASP-ChIFN-α的蛋白表达含量比pET-28a/ChlFN-α的蛋白表达含量要高出11%,表达的重组蛋白具有特异性.实验实现了鸡α-干扰素成熟蛋白基因的高效表达,筛选出了可应用于临床的鸡α-干扰素高效表达菌株BL21(DE3)(pET-28a/ASP-ChIFN-α).  相似文献

为解决异源表达酮还原酶域EryKR1的重组大肠杆菌Escherichia coli BL21(pET28a-eryKR1)催化环己酮还原时消耗的氢供体NADPH再生的问题,构建了克隆枯草芽孢杆菌葡萄糖脱氢酶基因gdh的重组菌E.coli BL21(pET28a-gdh),其中的gdh基因经Nucleotide BLAST功能分析显示与枯草芽孢杆菌9902的gdh基因序列(登录号为EF626962.1)的一致性达到100%。SDS-PAGE检测显示该重组菌经IPTG诱导后可以高效表达出葡萄糖脱氢酶(GDH),其表达量占全菌可溶性蛋白质的64%。GDH粗酶液的比酶活为137.90U/mg。通过气相色谱检测添加了E.coli BL21(pET28a-gdh)的E.coli BL21(pET28a-eryKR1)环己酮还原反应体系中的环己醇含量,结果显示加入重组GDH的双重组菌耦合反应体系中环己醇的产率为82.21%,是未添加GDH的反应体系对应值的3.23倍,表明重组GDH可以为EryKR1还原环己酮系统解决辅酶再生问题。  相似文献

为了获得重组猪生长激素抗体,对重组猪生长激素融合基因的真核表达产物进行免疫印迹(Western blot)检测,将利用DNA重组技术构建的重组质粒pPGH020在大肠杆菌(E.coli)BL21中进行诱导表达.表达产物经Ni+亲和层析柱纯化,获得纯化的重组猪生长激素融合蛋白.然后以此为抗原免疫新西兰大白兔,获得多克隆抗体,抗体再经硫铵沉淀、透析和亲和层析纯化.Western印迹结果表明,该纯化抗体显示了良好的免疫反应,其灵敏度比兔抗rpGH抗血清提高50倍,为猪生长激素融合基因在真核细胞的表达及功能鉴定奠定了基础.  相似文献

通过PCR从红斑丹毒丝菌C43065株基因组DNA中扩增出编码信号肽除外的成熟SpaA蛋白基因spaA,将其克隆到表达载体pET32a的BamHⅠ和Hind Ⅲ位点上,构建重组表达质粒pET-spaA,转化大肠杆菌BL21,在IPTG诱导下表达N端带有Trx标签的融合蛋白rSpaA,SDS-PAGE检测表达蛋白.DNA测序结果表明,spaA基因大小为1794 bp,编码由597个氨基酸残基组成的成熟SpaA蛋白,SDS-PAGE结果显示在大肠杆菌BL21中成功表达了分子量约为86 kDa的重组rSpaA,为进一步开展SpaA保护区域的研究奠定基础.  相似文献

目的:研究新型抗糖尿病药物exendin-4的分子作用机制,为exendin-4的大规模生产奠定基础.方法:采用重叠PCR法扩增出exendin-4的完整序列,并将其克隆至表达载体pET22b上,得到重组质粒pET-Exn4.重组质粒转化入大肠杆菌BL21(DE3)中,经IPTG诱导表达后,超声破碎表达产物,Tricine-SDS-PAGE分析exendin-4表达.结果:PCR、酶切鉴定和测序结果证明成功构建了重组表达载体pET-Exn4,Tricine-SDS-PAGE结果表明ex-endin-4基因在大肠杆菌中获得了分泌表达.结论:成功构建了exendin-4的重组表达载体并在大肠杆菌中实现分泌表达.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
