The effect ofγ-irradiation on soil enzyme stability

Summary Arylsulphatase, -1,3 glucanase, phosphatase and urease responded differently to -irradiation (5–50 Mrad) in air-dried and moist soils. In all instances phosphatase was the most stable. The variability between enzymes may be due to inherent biochemical and stuctural characteristics or to their location within the soil microenvironment.Supported by a grant from the NERC.     

The effect of whole body röntgen irradiation on the megakaryocytic system in rat femur

Zusammenfassung Das hämopoietische Gewebe des Femurs junger Albinoratten wurde in einer Lösung, die 5% Na-Zitrat (1 Teil) und 0,7% Na-Chlorid (4 Teile) enthielt, suspendiert. Die Megakaryozyten, deren Durchmesser 24 µ überschritt, wurden in einer Bürker-Kammer gezählt. Der Effekt der Ganzkörperbestrahlung auf das Megakaryozytensystem wurde untersucht durch Festlegung des femoralen Megakaryozytengehaltes während den ersten 7 Tagen nach der Bestrahlung.     

The effects of γ-aminobutyric acid and picrotoxin on the junctional potential and the contraction of crayfish muscle

Résumé L'acide-amino-butyrique et la-alanine reproduisent l'action du transmetteur inhibiteur du système neuromusculaire de l'écrevisse. Ces deux acids aminés diminuent la contraction musculaire. L'acide-amino-butyrique réduit la différence de potentiel et augmente sa décomposition. L'action de ces acides aminés est bloquée par la picrotoxine comme l'est celle du transmetteur inhibiteur.

The effect of αB-crystallin and Hsp27 on the availability of translation initiation factors in heat-shocked cells

The mechanism of the translational thermotolerance provided by the small heat shock proteins (sHsps) αB-crystallin or Hsp27 is unknown. We show here that Hsp27, but not αB-crystallin, increased the pool of mobile stress granule-associated enhanced green fluorescent protein (EGFP)-eukaryotic translation initiation factor (eIF)4E in heat-shocked cells, as determined by fluorescence recovery after photobleaching. Hsp27 also partially prevented the sharp decrease in the pool of mobile cytoplasmic EGFP-eIF4G. sHsps did not prevent the phosphorylation of eIF2α by a heat shock, but promoted dephosphorylation during recovery. Expression of the C-terminal fragment of GADD34, which causes constitutive dephosphorylation of eIF2α, fully compensated for the stimulatory effect of αB-crystallin on protein synthesis in heat-shocked cells, but only partially for that of Hsp27. Our data show that sHsps do not prevent the inhibition of protein synthesis upon heat shock, but restore translation more rapidly by promoting the dephosphorylation of eIF2α and, in the case of Hsp27, the availability of eIF4E and eIF4G. The effect of cyclic cytidine 3′,5′-monophosphate (cCMP) on the in vitro development,hatching and attachment of the mouse blastocyst

Summary The in vitro development and attachment of hatched mouse blastocysts on the untreated substratum was enhanced by 10 M dibutyryl cCMP (dbcCMP). The result suggests that cCMP has an effect on embryonic development and on the blastocyst attachment process.25 November1986     

The effect of bile salts on thyroxine 5′-monodeiodination in rat liver homogenate

Summary Unconjugated and conjugated bile salts inhibited the conversion of thyroxine to 3,3,5 triiodothyronine in rat liver homogenate.This work was supported in part by a grant from the Imanaga Foundation, Nagoya, Japan.     

The effect of o,p′-DDD on the adrenal cortex in sheep

Summary The effect of o,p-DDD (200 mg/kg/day given p.o. for 100 consecutive days) on the sheep adrenal gland was studied. The results suggest that this ruminant species is highly resistant to the adrenocorticolytic activity of o,p-DDD when compared with dogs.This work was supported by a grant of Bristol Laboratories, Syracuse, New York 13201, USA.     

The effect of chronic marginal vitamin C deficiency on the α-tocopherol content of the organs and plasma of guinea-pigs

Summary Chronic marginal vitamin C deficiency lasting 270 days induced hypertriglyceridaemia, an abrupt fall of ascorbic acid in all organs and a significant decrease of -tocopherol in the liver and the lungs in guinea-pigs.     

The effect of ischæmia on nucleoproteins in liver cells

Résumé Chez la souris, 24 heures après la ligature du pédicule d'un lobe hépatique, la basophilie cytoplasmique disparaît alors que les noyaux conservent une colorabilité normale par la méthode deFeulgen. Les dosages chimiques établissent que le taux de l'acide ribonucléique s'abaisse considérablement, tandis que celui de l'acide désoxyribonucléique ne subit pas de modifications appréciables. Ces phénomènes sont liés à la destruction des granules et microsomes séparables par ultracentrifugation. Une partie de ces nucléoprotéines se retrouve, après ultracentrifugations répétées, dans le liquide surnageant.     

The action of 5-hydroxy-dl-tryptophan and 5-hydroxy-tryptamine on the cortical electrical activity of the ‘midpontine pretrigeminal preparation’

Isotope effect studies on the dehydrogenation of Δ1-tetrahydrocannabinol in the rat

Summary Isotope effect studies on the metabolic dehydrogenation of ¹-tetrahydrocannabinol in rats are described and its is shown that this process is confined to a very short period following i.v. administration. The in vivo expression of the globin genes of theβ cistron in γ-,δ-, andδβ-thalassemia heterozygotes

There is considerable evidence suggesting that the switch from to and chain production after birth is due, in part, to silencing of the genes by stage-specific factors which bind to their promoters and to the competition from the adult ( and ) genes for a common enhancer element located in the locus control region. As a consequence one can expect that the increased Hb F production in adults with hereditary persistence of fetal hemoglobin or -thalassemia is directed mainly by -globin genes in cis to the deletion(s) responsible for these conditions. Here we review data on heterozygotes with -, -, or -thalassemia, who also had an^AT mutation, in cis or in trans, which was used as a marker of gene expression. The results show that a deletion affecting adult genes favors the expression of genes in cis, while the deletion of a single gene does not affect the expression of the gene in cis but leads to a faster switch postnatally.     

Effect of dexamethasone on Fc γ receptor expression in foetal and neonatal rat gut

Summary When injected into 12-day-old suckling rats, dexamethasone caused a precocious disappearance of Fc receptors from enterocytes of the proximal small intestine. However, dexamethasone appeared to be necessary for the maintenance or production of such receptors in foetal rat gut cultured in vitro.Supported by an award from the Science and Engineering Research Council.     

The partitioning ofΔ 1-tetrahydrocannabinol into erythrocyte membranes in vivo and its effect on membrane fluidity1into erythrocyte membranes in vivo and its effect on membrane fluidity1

Summary ¹-Tetrahydrocannabinol ( ¹-THC) has been quantified directly in erythrocyte membranes from drug-treated mice using gas chromatography/mass spectrometry. Concentrations of approximately 6 ng ¹-THC/mg membrane protein (10^–5 M) were found when effects of the drug on behavior were prevalent. At these concentrations the drug produced a decrease in membrane order as measured by ESR.This work was supported by grants from the Medical Research Council, the Wellcome Trust and the E. P. The effect ofβ-adrenergic stimulation and blockade on the efflux of PGE1-like material from the isolated perfused rabbit heart

Summary Prostaglandin (PG) release was measured from the isolated perfused rabbit heart. The effects of -adrenergic stimulation and blockade suggest that PG synthesis is regulated in part by adrenergic mechanisms.     

The product of the γ-secretase processing of ephrinB2 regulates VE-cadherin complexes and angiogenesis

Presenilin-1 (PS1) gene encodes the catalytic component of γ-secretase, which proteolytically processes several type I transmembrane proteins. We here present evidence that the cytosolic peptide efnB2/CTF2 produced by the PS1/γ-secretase cleavage of efnB2 ligand promotes EphB4 receptor-dependent angiogenesis in vitro. EfnB2/CTF2 increases endothelial cell sprouting and tube formation, stimulates the formation of angiogenic complexes that include VE-cadherin, Raf-1 and Rok-α, and increases MLC2 phosphorylation. These functions are mediated by the PDZ-binding domain of efnB2. Acute downregulation of PS1 or inhibition of γ-secretase inhibits the angiogenic functions of EphB4 while absence of PS1 decreases the VE-cadherin angiogenic complexes of mouse brain. Our data reveal a mechanism by which PS1/γ-secretase regulates efnB2/EphB4 mediated angiogenesis.     

Sex determination in mammals — Before and after the evolution of SRY

Therian mammals (marsupials and placentals) have an XX female: XY male sex chromosome system, which is homologous to autosomes in other vertebrates. The testis-determining gene, SRY, is conserved on the Y throughout therians, but is absent in other vertebrates, suggesting that the mammal system evolved about 310 million years ago (MYA). However, recent work on the basal monotreme mammals has completely changed our conception of how and when this change occurred. Platypus and echidna lack SRY, and the therian X and Y are represented by autosomes, implying that SRY evolved in therians after their divergence from monotremes only 166 MYA. Clues to the ancestral mechanism usurped by SRY in therians are provided by the monotremes, whose sex chromosomes are homologous to the ZW of birds. This suggests that the therian X and Y, and the SRY gene, evolved from an ancient bird-like sex chromosome system which predates the divergence of mammals and reptiles 310 MYA.
