河套蜜瓜ACC氧化酶基因cDNA部分片段的克隆和序列分析

以河套蜜瓜(Cucumis melo L．ev Hetao)成熟果实为材料，用硫氰酸胍法提取总RNA，以oligo(dT)15为引物，反转录合成cDNA，利用一对甜瓜ACC氧化酶(1-aminocyclopropane-1-earboxylate oxidase，ACO)基因cDNA的特异性引物进行聚合酶链式反应(PCR)扩增。获得了预期大小的cDNA片段．将此片段克隆于pUCl9质粒中，进行DNA序列分析．序列分析结果表明该片段为545bp，编码甜瓜ACO的第126至第306个氨基酸．该河套蜜瓜ACO基因cDNA序列与Andes甜瓜、Cantaloup charentais甜瓜和哈密瓜的相应序列进行比较，其核苷酸序列与Andes甜瓜的同源性为99．4％；与Cantaloup charentais甜瓜和哈密瓜的同源性均为99．6％。与其对应的氨基酸序列的同源性均为99．4％．     

猪肝脏和肌肉肉碱脂酰转移酶I基因片段克隆及序列分析

分别从金皮F2代猪的肝脏、第十肋背最长肌中提取总RNA,并根据报道的猪L—CPT I cDNA和M—CPT I cDNA序列分别设计引物1和引物2,用反转录一聚合酶链式反应（RT—PCR）进行cDNA扩增,获得两条长度分别为733bp和510bp的片段,克隆于pUCm-TVector后进行序列分析。结果表明,长度为733bp的片段为L—CPTI基因cDNA的部分序列,编码243个氨基酸,而长度为510bp的片段则为M—CPTI基因cDNA的部分序列,编码169个氨基酸。得到的两条基因片段与报道的猪L—CPT IcDNA和M—CPT IcDNA部分序列同源性分别为99.59％和99．61％。     

常规PCR与RACE结合法快速从cDNA文库中克隆基因

首先根据Fe(Ⅱ)-转运蛋白基因家族的保守区设计同源的常规PCR引物(F1，R1)，扩增出490bp的特异片段。然后根据RACE法原理，巧妙地设计3′RACE(F2)和5′RACE(R3)特异性引物，使两者既能分别与文库载体上的筛选扩增引物配对扩增cDNA 3′和5′末端序列，又能使扩增出的3′RACE和5′RACE产物序列有重叠部分；另外设计引物时还兼顾到使F2和R3也能配对，从而能对3′RACE和5′RACE扩增产物进行双特异性引物常规PCR鉴定。该方案不但能够直接根据序列鉴定3′RACE和5′RACE产物是否为同一基因序列，还能快捷地用常规PCR鉴定3′RACE和5′RACE扩增产物是否为特异性扩增，避免了对非特异性扩增产物的克隆测序等繁琐的鉴定过程，优化了基因克隆策略。最后根据拼读出的全长序列设计引物(F4，R4)扩增出完整编码区。应用该方法成功地从小金海棠缺铁处理的根系cDNA文库中克隆了Fe(Ⅱ)-转运蛋白基因的cDNA。说明该方案是一种从cDNA文库中克隆目的基因的快捷优化方法。     

应用3’RACE技术扩增仿刺参C型凝集素基因及实验条件的优化

从GenBank上调取刺参C型凝集素（C-type Lectin）基因序列EST,根据此序列设计RACE引物,采用PCR扩增技术得到了仿刺参C-type Lectin基因序列（EST）,根据这段EST序列设计1个基因特异引物（GSPF）,与通用引物（UPM）扩增,成功地克隆到了该基因的3’末端序列.同时,对仿刺参C型凝集素基因3’克隆的实验条件进行了优化.该扩增片段长度为670bp,与已知序列重叠部分为417bp.经测序和比对发现该段序列与预期的目标基因的序列一致.     

人Oligophrenin 1样(OPHN1L)基因的克隆

将人Oligophrenin 1(OPHN1)基因编码区2406 bp与EST数据库进行同源性分析,得到一个363 bp的EST AA035622与OPHN1基因编码区一致性为62%,该EST与一个cDNA序列AB014521完全匹配.在ABO14521中设计引物与cDNA文库载体臂上引物行巢式PCR扩增并进行5′RACE,在胎盘cDNA文库中获得cDNA序列606bp,与AB014521拼接成一个6906 bp的cDNA序列,其中包含一个2442 bp的开放阅读框,编码813个氨基酸.该cDNA序列的GenBank登录号为AF141884,并经GDB命名委员会命名为OPHN1L基因.OPHN1L基因3′端与大规模测序的BAC克隆AC005348及AC004782完全匹配,从而将该基因定位于5q21.2～q21.3,并由此获得它的部分基因组结构.     

禽巴氏杆菌抗链霉素耐药基因StrA的克隆及同原性比较

根据巴氏杆菌基因序列设计合成一对引物，以临床分离的动物源性巴氏杆菌抗链霉素耐药菌株为模板，通过PCR技术，扩增出StrA基因350-1153bp序列．PCR产物及表达载体通过粘端连接，将长约804bp的StrA目的基因片段克隆到pGEM—T载体上，构建了克隆载体质粒pGEM—TstrA．StrA基因片段测序结果与文献报道的结果同源性为99．8％．只有一个碱基发生突变，但所编码的氨基酸没有改变．     

玉米候选抗病相关基因片段的克隆

根据已经克隆的植物抗病基因和候选抗病基因的保守序列P-loop、Kinase-2及GLPLAL设计一系列简并引物，利用同源序列扩增法，对玉米的基因组DNA进行PCR扩增，并对5个扩增产物的克隆进行测序．测序结果在Gen—Bank内进行BLAST检索，发现A9克隆序列与玉米BAC库中的206C17克隆的部分序列有很高的相似性，并且距离GenBank内注册的玉米抗锈病基因rpl位点中的rpl-3基因、rpl-4基因分别约有66Kb、20Kb，且A9克隆序列在玉米基因组中是单拷贝的．这为玉米抗锈病性状的分子标记辅助选择和抗锈病基因的克隆奠定了良好的基础。     

滑膜肉瘤融合基因SYT-SSX cDNA的克隆与序列分析

利用逆转录聚合酶链式反应(RT-PCR)方法获得SYT-SSX的cDNA，克隆入PUCm-T载体后，在美国ABI DNA自动测序仪上以反向引物测序法进行核苷酸序列的测定，扩增出98bp的特异性片段，并筛选出阳性重组克隆质粒PUCm-SYT-SSX。获得的滑膜肉瘤融合基因SYT-SSX的cDNA，为深入揭示滑膜肉瘤分子发生机理奠定基础，同时可进一步完成原核表达。     

人胰岛素样生长因子-Ⅱ基因cDNA克隆与序列分析

本研究通过RT-PCR技术从人胎盘组织中扩增出长为841bp的胰岛素样生长因子-ⅡcDNA,PCR产物经凝胶回收纯化后,克隆在PGEM-T载体的T位点.测序结果和序列分析表明,本人已成功地扩增、克隆了人胰岛素样生长因子-Ⅱ基因cDNA序列.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
