根据结构动力学理论证明了能力谱曲线的确定与侧向荷载分布模式间存在对应关系,指出对于不同的侧向荷载模式,应采用不同的能力谱曲线转化公式并给出了具体的表达式. 分别采用本文方法、传统方法和弹塑性时程分析对一个六层钢筋混凝土框架结构进行了计算,证明了本文方法计算精度更高. 利用该方法,采用多种侧向荷载模式对结构进行静力弹塑性分析,能将侧向位移反应可能范围限定得更小,使人们把握实际地震反应大小的准确性得以提高. 相似文献

目的探讨nm23-H1基因的表达与白血病细胞HL-60增殖之间的关系。方法:以25ng/mL阿糖胞苷处理HL-60细胞,MTT法测定细胞生长抑制率,NBT还原比色法判断细胞分化状况,RT-PCR检测nm23-H1基因表达的变化;构建nm23-H1基因的真核表达质粒pEGFP-N1-nm23-H1,转染HL-60细胞,通过细胞生长曲线和血清依赖性实验检测nm23-H1基因的过表达对HL-60细胞生长的影响。结果:小剂量Ara-C对HL-60细胞的生长呈时间依赖性抑制,作用4d后细胞NBT还原能力增强且nm23-H1基因的表达下调;转染nm23-H1基因的HL-60细胞生长加快、血清依赖性下降。结论:Ara-C对HL-60细胞增殖的抑制作用与下调nm23-H1基因的表达有-定关系;nm23-H1基因在HL-60细胞中的过表达有促细胞增殖的作用,即增高了HL-60细胞的恶性程度。 相似文献

Cattle yaks are an F1 hybrid between cattle and yaks and exhibit significant hybrid vigor. However, cattle yaks are sterile males. To study the epigenetic aspects of this reproductive isolation, IGF2 mRNA expression in cattle yaks and their parents and the patterns of the IGF2 differentially methylated regions (DMR) were examined in blood and testes. The results showed that IGF2 expression in cattle yaks’ testes was lower than in their parents (P < 0.01). The IGF2 DMR was highly methylated (above 90%) in blood and testes of cattle yaks, yaks and cattle, with the highest level in cattle yaks (P > 0.05). Our study showed that IGF2 plays an important role in bovine spermatogenesis and might be involved in cattle-yak male sterility. The methylation level of the IGF2 DMR was irrelevant to the lower expression of IGF2 in cattle yaks; other factors perhaps play roles in its expression. 相似文献

Dopamine modulates S-potential amplitude and dye-coupling between external horizontal cells in carp retina 总被引:9,自引:0,他引:9
Horizontal cells in the fish retina are electrically coupled and possess gap junctions so that intracellularly injected dye normally diffuses freely to neighbouring cells. Applied dopamine (DA) alters the spatial properties of the horizontal cell responses to light, increasing the amplitude of photopic L-type S-potentials but decreasing their lateral spread. These effects have been attributed to the action of DA on horizontal cell membrane resistance, particularly at the gap junctions, and our present study on the carp retina agrees with this in showing that DA also restricts intracellular Lucifer yellow (LY) to single injected horizontal cells, an effect, like those of DA on the S-potentials, which is antagonized by the dopamine blocker haloperidol. In addition, we present evidence that dopaminergic interplexiform cells in fish normally function to regulate the spatial properties of responses in horizontal cells, possibly acting on their junctional resistance via a DA-receptor-mediated mechanism. Previous destruction of the interplexiform cells with 6-hydroxydopamine (6-OHDA) resulted in much reduced L-type S-potentials to centred lights but wider lateral spread of these responses, while the dye injected spread extensively to neighbouring cells. After 6-OHDA treatment, however, applied DA retained its normal activity, restoring large-amplitude, narrow receptive-field S-potentials and restricting LY to the injected cells, effects which were both closely mimicked by dibutyryl cyclic AMP. 相似文献

不透水面指数可以有效提取不透水面信息,反映城市硬化地表的覆盖度,但不能直接表征地表覆盖度的复杂度,而遥感图像信息容量是一种可以表征地表景观复杂度的指标.该文结合二者各自特点,计算出遥感图像信息容量和不透水面指数,对二者进行耦合分析,发现不同波段的信息容量与城市不透水面指数均方差的相关系数均高于0.8,二者关系密切,说明信息容量可以表征城市不透水面的变化,进一步研究还发现,信息容量值的大小与城市化进程较为一致. 相似文献

Protein-protein interaction is a physical interaction of two proteins in living cells. In budding yeast Saccharomyces cerevisiae, large-seale protein-protein interaction data have been obtained through high-throughput yeast two-hybrid systems (Y2H) and protein complex purification techniques based on mass-spectrometry. Here, we collect 11855 interactions between total 2617 proteins. Through seriate genome-wide mRNA expression data, similarity between two genes could be measured. Protein complex data can also be obtained publicly and can be translated to pair relationship that any two proteins can only exist in the same complex or not. Analysis of protein complex data, protein-protein interaction data and mRNA expression data can elucidate correlations between them. The results show that proteins that have interactions or similar expression patterns have a higher possibility to be in the same protein complex than randomized selected proteins, and proteins which have interactions and similar expression patterns are even more possible to exist in the same protein complex. The work indirates that comprehensive integration and analysis of public large-seale bioinformatical data, such as protein complex data, protein-protein interaction data and mRNA expression data, may help to uncover their relationships and common biological information underlying these data. The strategies described here may help to integrate and analyze other functional genomic and proteomic data, such as gene expression profiling, protein-localization mapping and large-scale phenotypic data, both in yeast and in other organisms. 相似文献

In addition to conventional neurotransmitters such as acetylcholine, dopamine, glycine and gamma-aminobutyric acid (GABA), a number of peptide-immunoreactive substances have recently been localized in the vertebrate retina. The functional roles of these retinal peptides and their interactions with conventional neurotransmitters are largely unknown. We have previously shown that exogenous opiates affect both the release of GABA and the firing patterns of ganglion cells in the goldfish retina, and we have now begun a systematic characterization of the opioid pathways in the chicken retina, because, among vertebrate retinas, avian retinas contain the highest concentration of enkephalins. Monoclonal antibodies specific for enkephalin have been used to demonstrate that a subpopulation of enkephalin-containing amacrine cells exists in the chicken retina. This retina also synthesizes Met-enkephalin and releases it on cell depolarization. The enkephalin-induced inhibition of GABA release in goldfish retina led us to examine whether similar interactions occur in chicken, and if so, whether enkephalins and GABA coexist in the same amacrine cells. Our results, presented here, indicate that exogenous enkephalins do indeed inhibit GABA release in the chicken retina. Surprisingly, we found that although some amacrine cells contain both enkephalin and GABA, others contain only one or the other. 相似文献

探讨NHEJ信号通路关键基因XRCC4,XRCC5,XRCC6,LIG4,PRKDC及DCLRE1C的mRNA表达水平与宫颈鳞癌同步放化疗敏感性的关系。以42例宫颈鳞癌活检样本为研究对象,采用荧光定量PCR(qRT-PCR)技术分析上述6个基因的mRNA表达水平,并将检测结果与患者的同步放化疗敏感性进行关联分析。XRCC6 mRNA高表达患者应答率为47.6%,低表达患者应答率为90.5%(P=0.003);其他5个关键基因的mRNA表达水平与患者的应答率无显著性关联;多因素Logistic回归分析显示XRCC6 mRNA表达水平与患者同步放化疗敏感性有显著性关联(P=0.008)。XRCC6 mRNA表达水平与宫颈鳞癌患者同步放化疗敏感性密切相关,可作为评估其临床疗效的分子标志物。 相似文献
