饭豆铜锌超氧物歧化酶的纯化及性质

饭豆种子粗提液中有６条Ｃｕ·Ｚｎ—ＳＯＤ活性谱带，经氯仿－乙醇混合液处理，硫酸铵盐析，ＳｅｐｈａｄｅｘＧ－１００凝胶过滤和ＤＥＡＥ－纤维素柱层析被分离，获得了两组ＳＯＤ同工酶组分（ＳＯＤ—１，ＳＯＤ—２）．它们在酶分子结构上可能存在一定差异．ＳＯＤ－１含与粗提液相对的、电泳迁移率较小的活性谱带，它在聚丙烯酰胺凝胶电泳（ＰＡＧＥ）图谱上的活性染色带和蛋白染色带相互对在；在ＳＤＳ－ＰＡＧＥ和等电聚焦电泳图谱上均呈现单一区带，说明该酶已纯化到均一程度．该酶分子量为３１３００，亚基分子量为１６５００，等电点为４．２，在紫外区的吸收值为２６４．０ｎｍ．该必在０—７５℃范围内稳定．纯化的ＳＯＤ—１比活性为１７８９Ｕ／ｍｇ，ＳＯＤ—２比活性为２８４Ｕ／ｍｇ，ＳＯＤ活力回收率为３２％．     

用铜盐法纯化牛血红细胞铜锌超氧物歧化酶

牛血红细胞铜锌超氧物歧化酶（Ｃｕ·Ｚｎ－ＳＯＤ），经溶血，ＣｕＣｌ２处理及离子交换柱层析等步骤被纯化到高度均一程度．纯化酶在聚丙烯酰胺梯度凝胶图谱上的活性染色带与蛋白染色带相对应，在ＳＤＳ－ＰＡＧＥ图谱上呈现单一区带．元素分析表明该酶含有２个锌原子和多于２个的铜原子．分子量和亚基分子量分别为３３５００和１７８００．在紫外区酶有４个明显吸收峰，分别为２７４．６ｎｍ，２６６．２ｎｍ，２５８．８ｎｍ和２４５．４ｎｍ．该酶对热稳定，纯化后酶比活性为１４５２５ｕ／ｍｇ，活力回收率为４５％．本文对经ＣｕＣｌ２处理与氯仿－乙醇处理纯化的牛血Ｃｕ·Ｚｎ－ＳＯＤ进行比较，结果表明，氯仿－乙醇对酶的某些性质有影响     

水牛梭形住肉孢子虫缓殖子抗原的纯化及SDS—PAGE分析

应用葡聚糖凝胶柱层纯化了水牛梭形住内孢子虫缓殖子抗原，并应用ＳＤＳ－ＰＡＧＥ分析了缓殖子可溶性粗抗原及纯化抗原的蛋白组分，结果表明：缓殖子可溶性粗抗原含有１４种蛋白成分，分子量范围为４５．９－７１．２ＫＤ，经过ＳｅｐｈａｄｅｘＧ－１００柱层析纯化扣的抗原在ＳＤＳ－ＰＡＧＥ图谱上显示５条蛋白带，分子量分别为５８ＫＤ，６１ＫＤ，６４ＫＤ，６６ＫＤ，７０ＫＤ。     

花椰菜凝集素的纯化及部分性质的研究

花椰菜凝集素由花蕊组织经生理盐水抽提、硫酸铵沉淀和ＳｅｐｈａｄｅｘＧ－１００、Ｓｅｐｈａｒｏｓｅ－４Ｂ凝胶层析纯化获得．ＰＡＧＥ鉴定和高碘酸－Ｓｃｈｉｆｆ试剂反应均显单一条带，说明该凝集素为纯化的糖蛋白．经ＳｅｐｈａｄｅｘＧ－２００柱过滤分析，其相对分子质量约为９４０００．ＳＤＳ－ＰＡＧＥ分析表明该凝集素含有４个亚基，其相对分子质量分别约为２８０００、２５０００、２２０００和２００００．花椰菜凝集素经５０℃处理５分钟仍保持高的凝集活性，该凝集素对酸处理较碱处理稳定．     

福寿螺β-葡萄糖苷酶的分离纯化及性质的初步研究

以福寿螺内脏为材料,经本实验室开发的工业生产法制备粗酶粉,进一步采用葡聚糖凝胶（ＳｅｐｈａｄｅｘＧ－２００）和ＤＥＡＥ－ＳｅｐｈａｄｅｘＡ－５０柱层析纯化,获得经ＳＤＳ－聚丙烯酰胺凝胶电泳鉴定为单一纯的β－葡萄糖苷酶制剂．测定该酶的最适反应温度为４３℃,最适反应ｐＨ为５．１;在ｐＨ５．１,４５℃下,该酶催化水杨素水解的Ｋｍ值为３．４０ｍｍｏｌ／Ｌ,活化能为５０．６３ｋＪ／ｍｏｌ．研究几种效应物对酶活力的影响,结果表明：Ｍｎ２＋、Ｃｏ２＋对酶有激活作用,而Ｃｕ２＋、Ｈｇ２＋、Ｐｂ２＋、ＳＤＳ及脲对酶有不同程度的抑制作用,其中Ｃｕ２＋和Ｈｇ２＋对该酶的抑制作用最强,其抑制机理均表现为非竞争性抑制

鱼中高温激活蛋白酶的提取纯化

以市售鱼为原料，进行其高温激活蛋白酶（ＨＴＡＰ）的提取纯化通过抽提、高速离心分离、ＤＥＡＥ－ＴＯＹＯＰＥＡＲＬ６５０Ｍ离子交换色谱、ＳＥＰＨＡＣＲＹＬＳ２００凝胶过滤等方法，把ＨＴＡＰ从鱼中分离提取出来运用ＳＤＳ－聚丙烯酰胺凝胶电泳（ＳＤＳ－ＰＡＧＥ）法对分离提纯的ＨＴＡＰ纯度进行鉴定，并通过此法测出了该ＨＴＡＰ的分子量为３９０００     

根瘤菌酯酶SOD和G6PD的凝胶电泳

黑曲霉胞外葡萄糖淀粉酶的纯化及特性

黑曲霉（Ａｓｐｅｒｇｉｌｌｕｓｎｉｇｅｒ７２８）的粗酶液，经硫酸铵沉淀，ＳｅｐｈａｄｅｘＧ－２５柱凝胶过滤脱盐，ＤＥＡＥ－Ｔｏｙｏｐｅａｒｌ离子交换柱层析和ＳｅｐｈａｃｒｙＳ－１００凝胶层析纯化，经ＰＡＧＥ鉴定为一条带ＳＤＳ凝胶电泳测定分子量为７００００．金属离子Ｆｅ３＋对该酶有一定的抑制作用，该酶的等电点为３．７，最适ｐＨ为４，最适温度为６０℃．Ｅ２８０１％为１５．７２，Ｋｍ值为０．１１２×１０－３ｍ．     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。

橡胶颗粒沥青混合料低温抗裂性试验研究

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.
