Résumé (1) Des adipocytes isolés à partir du tissu adipeux épididymaire du rat ont été incubés dans des milieux contenant du glucose (114C) et des concentrations variables d'insuline. La quantité de glucose (114C) transformée en glycogène (14C), (14C)O2 et triglycerides (14C) a été déterminée. (2) On démontre que la quantité de glucose (114C) transformée en glycogène par des adipocytes peut être évaluée et qu'elle est fonction de la concentration d'insuline. (3) La comparaison entre les effets d'insuline sur les transformations du glucose (114C) en glycogène (14C), en triglycerides (14C) ou (14C)O2 révèle que l'insuline a une action apparemment directe sur l'incorporation du glucose au glycogène. (4) Ce fait serait donc dû à un effet direct de l'insuline sur une réaction dans la synthèse du glycogène par les adipocytes ou à une grande affinité des enzymes participant à la synthèse du glycogène pour le glucose-1-phosphate et à leur faible capacité pour la synthèse du glycogène.  相似文献

Summary Mouse maxillary second molars were removed at either 24 or 96 h of age and maintained in vitro. Half of the teeth, of each age group, were treated with 50 m-units of synthetic salmon calcitonin. By comparing the initial and final calcium concentrations in the medium, the net uptake or release of calcium was inferred. The treated molars took up significantly more calcium than the untreated groups.  相似文献

Mouse embryos explanted at various stages during neurulation were cultured for 20-28 h in the presence of 25-900 micrograms/ml of 5-bromodeoxyuridine (BUdR). BUdR strongly inhibited closure of the cranial neural tube, which was found to be stage-dependent. When mouse embryos were exposed to BUdR after development of the concave curvature in the neuroepithelium of the midbrain to the upper hindbrain regions, they became insensitive to the drug-induced open cranial neural tube. Histological observations showed that BUdR interfered with interkinetic migration and cytokinesis of the neuroepithelial cells. These cellular abnormalities were not dependent on the morphological development of the cranial neural folds. The 3H-BUdR experiment confirmed that the label was mostly incorporated into the DNA fraction.  相似文献

Zusammenfassung Das Serum von Uremikern inhibiert die Glukoseutilisation im Rattendiaphragma. Der Sauerstoffverbrauch ändert sich nicht trotz niedrigerer Glukoseutilisation, da dieselbe durch eine erhöhte Auswertung anderer Substrate im Diaphragma kompensiert wird. Mild oxidation of mouse spleen cells by sodium periodate induces blastogenesis. Concomitantly, the in vitro response of these cells to sheep red blood cells is inhibited. Summary The effect of a potent antiserotonin-antithistaminic compound, cyproheptadine (CPH) on insulin biosynthesis was studied in pancreatic islets isolated from CPH-treated rats. Though insulin content of islets was markedly reduced in CPH-treated rats, the incorporation of radiolabeled leucine into proinsulin and insulin fractions was not affected with respect to the rate and amount.
