Summary Treatment of BHK cells for 8 h with triethylenemelamine (TEM) followed by caffeine for 4 or 8 h, increased the frequency of anaphase-telophase chromatin bridges in relation to controls and TEM-treated cells. These results indicate that TEM-induced chromosome lesions detected as chromatin bridges at anaphase-telophase could be potentiated by caffeine.This work was supported by grants from CIC, CONICET and CAFPTA. 相似文献

In this work the possibility that a mutagenic factor acting in utero or in the perinatal period might lead to elevated mutagenic rates in bone-marrow cells after a considerable period of time was examined. An aromatic hydrocarbon, benzo(a)pyrene was used as the test substance. Benzo(a)pyrene treatments resulted in significantly higher sister-chromatid exchange (SCE)-frequencies in both fetal and neonatal groups in both sexes, even four months after exposure. In a second experiment we examined whether mutagenic exposure suffered in utero could make the individual more susceptible to mutagenic effects in adulthood. Preliminary results indicate that such a possibility could exist. 相似文献

Summary The influence of steroid hormones on the induction of sister-chromatid exchange (SCE) in cultured CHO cells was studied. It was observed that estradiol-17 , estriol, estrone and ethynyl estradiol treatments enhanced SCE rates compared to the controls. Overall, these compounds produced a dose response effect. The importance of a detailed study on the long-term genetic effects of steroids on mammalian cells is emphasized.This work was supported by NIH-Minority Biomedical Research Support Program (MBRS), Grant No. RR 08124-11. Exposures of Chinese hamster cells to pulsing electromagnetic field (PEMF) with 0.18-2.5 mT did not influence the baseline frequency of sister-chromatid exchanges (SCE). Zn2+ in low concentrations (0.005–0.1 mM) inhibited the transient contractions in response to caffeine (25 mM) in a dose-dependent manner in smooth muscle of intact guinea-pig taenia caeci. At Zn2+ concentrations higher than 0.1 mM, caffeine did not elicit any response. After saponin-treatment of the fibres, which leaves the Ca2+ storage sites intact, caffeine contraction was completely inhibited by Zn2+ at a relatively low concentration (0.03 mM). However, in Triton-X-100-treated fibres, in which the Ca2+ release sites are destroyed, the contraction could be induced in the presence of Zn2+ by an increase in Ca2+. In conclusion, Zn2+ can block the intracellular Ca2+ release from caffeine-sensitive release sites in taenia caeci. 相似文献
