Degradation of [3H]thymidine by a pentosyltransferase (EC 2.4.2.4) in the plasma of man and different animals.

[3H]Thymidine is degraded by an enzyme (thymidine phosphorylase; EC 2.4.2.4) which we have identified in the plasma of man and some animals. The presence of this enzyme in plasma or sera used to supplement culture media may, under certain experimental conditions, limit the validity of measuring the uptake of radiolabeled thymidine as a means of defining DNA synthesis.     

Inhibition by tetanus and botulinum A toxin of the release of [3H]noradrenaline and [3H]GABA from rat brain homogenate

Rat brain homogenate was preloaded with [3H]noradrenaline or [3H]GABA and stimulated with high K+. Tetanus toxin and botulinum A neurotoxin partially prevent the evoked [3H]noradrenaline release in the same range of toxin concentrations starting below 10(-10) M. In contrast, release of gamma-amino butyric acid (GABA) is much more sensitive to tetanus than to botulinum A toxin.

Binding of [3H]GABA and [3H]muscimol to subcellular particles of a neurone-enriched culture of mouse brain

Summary Binding of [³H]GABA and [³H]muscimol, indicative of GABA-receptors, has been demonstrated in a neurone-enriched culture of embryonic mouse brain using a ligand-binding technique. [3H]-Norcocaine and [3H]-pseudococaine: Effect of N-demethylation and C2-epimerization of cocaine on its pharmacokinetics in the rat

Summary After i.v. injections of cocaine, norcocaine, pseudococaine to the rat, the T_1/2 in brain were 0.4, 0.6, 0.2 h respectively and in plasma 0.4, 0.5, 0.2 h respectively. Benzoylnorecgonine and norecgonine were the metabolites of norcocaine in brain. Pseudonorcocaine, pseudobenzoylnorecgonine, pseudobenzoylecgonine and pseudoecgonine were the metabolites of pseudococaine in rat brain. Benzoylnorecgonine and pseudobenzoylecgonine had potent stimulant activity intracisternally in the rats.Acknowledgment. Presence of a protein disulfide isomerase (EC 5.3.4.1) in wheat germ]

The presence of a protein disulfide isomerase (rearrangease) in wheat embryo has been demonstrated by its ability in reactivating randomly cross-linked ribonuclease. This activity requires a dialysable cofactor; after dialysis, the activity is recovered by addition of reduced glutathione. The enzyme can be precipitated by 70% saturation ammonium sulfate.     

Effect of local anaesthetics on the accumulation of [3H]-Metaraminol by rabbit atria and vasa deferentia

Zusammenfassung Es wurde die Wirkung von Lokalanästhetika auf die Anhäufung von [³H]-Metaraminol im Vorhofmuskel und Vas deferens des Kaninchens untersucht. Die von Kokain hervorgerufene Hemmung der [³H]-Metaraminol-Akkumulation ist mindestens hundertmal stärker als diejenige nach Procain, Prilocain und Lidocain.

delta-9-Tetrahydrocannabinol (THC) inhibition of the membrane transport system for thymidine]

THC decreases 3H-thymidine uptake within 15 sec. after addition of the drug to the culture (L1210 murine lymphoma cells and human lymphocytes). Experiments performed at 0 degrees C indicate that THC has no action on thymidine binding to the carrier. THC in micromolar concentration inhibits DNA synthesis through a alteration of membrane configuration.     

Metabolic requirements for the uptake and retention of H3Norepinephrine (H3NE) by isolated frog ventricle

Resumen El tratamiento con iodoacetato (IAA), no modifica la incorporación y retencion de H³Norepinefrina (H³NE) al ventriculo aislado deRana oxigenado y suspendido en ringer con o sin glucosa. Bajo atmosfera de Nitrógeno y ausencia de glucosa, la incorporacion de H³NE es bloqueada en un 58%; en estas condiciones el IAA produce un ulterior bloqueo del 33%.

Summary The biliary excretion of [³H] LSD was studied in Wistar and homozygous Gunn rats. In Wistar rats approximately 46% of the given dose was recovered from bile in 2.5 h whilst in the homozygous Gunn rat 26% was recovered in the same time period. In both strains the main metabolites were glucuronides.     

Degradation of IgG3 monoclonal proteins during storage and in the presence of a thiol (dl-penicillamine)

Résumé Il est bien connu que les immunoglobulines IgG maintenues longtemps à des températures voisines de 0 °C sont dénaturées. L'addition de pénicillamine au sérum peut accentuer cette dénaturation. Dans ces conditions, les immunoglobulines du type IgG3 sont les plus sensibles et le traitement par la pénicillamine peut être utilisé pour les identifier.     

Role played in man by myelinated fibers of different diameters for the evocation of a nociceptive flex reflex and the accompanying pain sensation]

Using simultaneous recordings we have made in Man a comparative study of: the sural nerve afferent volley, the nociceptive flexor reflex of a muscle of the lower limb and the associated painful sensation. Two types of stimulations were used, a single short duration electric stimulus, and a train of electric shocks (100/sec). With a single stimulus, the nociceptive flexor reflex and the painful sensation develop only when A delta fibers are recruited. On the other hand, when the stimulations are given by trains the nociceptive flexor reflex and the painful sensation can develop with a stimulus sub-liminar to the threshold of A delta fibers, when A alpha fibers are recruited. When the stimulus activate both A alpha and A delta fibers, the flexion reflex and the pain disappear when a selective blockade of the A delta group is exerted by means of Lidocain.     

Decrease in [3H]ouabain binding sites in heart and brain from spontaneously hypertensive rats

Summary A decrease in the number of binding sites (B_max) for [³H]ouabain was observed in the heart (37%) and the brain (22%) of spontaneously hypertensive rats (SHR) when compared with age-matched control Wistar Kyoto rats. No variation was detected in the affinity constant (K_D).

Na*-independent binding of [3H] muscimol to a membrane fraction of the brains of normal and dwarf mice

Summary Although their body weights were decreased by about 77% and their brain weights by about 30%, high-affinity [³H] muscimol binding to a cerebral membrane fraction was not altered in hereditary pituitary dwarf mice. Marked changes in the level of pituitary growth-associated hormones do not appear to be associated with a change in cerebral GABA-receptors.Supported by a UNESCO/IBRO Fellowship.
