用苦丁茶当年生枝条的顶芽和侧芽作外植体,培养在附加了各种浓度BA的MS培养基上,当BA浓度为3.0mg/L时诱导芽形成及增殖效果最佳,GA2.0+NAA0.2mg/L和BA配合使用有利于壮苗;在1/2MS培养基中,附加NAA0.5mg/L,生根效果最佳。试管苗高2 ̄3cm、根数2 ̄3条时开盖炼苗2d ̄3d移入培养土中,试管苗移栽成活率高。  相似文献

通过对以马铃薯(Solanum tuberousm L.)块茎为受体的农杆力菌转化系统进行优化,筛选出使三个品种(鲁引1号、克新3号、优金)的受体均可得高频分化的培养基:MS+2.0mg/L BA+3。0mg/L ZT+0.5mg/L NAA+0.5mg/L GA。对几种农杆菌转化条件的比较研究发现,菌体用Ms+0.5mg/L GA流体重悬并稀释4倍屿受体于28℃,100r/min振荡侵染20~3  相似文献

通过实验,筛选出银杏愈组织的最佳培养基为N6+NAA3.0mg/L+BA1.0mg/L,其次为MS+NAA3.0mg/L+BA1.0mg/L,分别对NAA、2,4-D、GA、BA和KT等在培养过程中的作用与浓度进行了探索,结果表明NAA、BA结合使用,既适合诱导又利于继代培养,2,4-D适合诱导,GA不利于诱导,KT有利于分化。  相似文献

在成功构建了谷脱甘肽(GSH)合成酶基因表达质料(pTrc-gsh)的基础上,对不同大肠杆蓖突主菌进行了比较,筛选出高效、稳定表达GSH合成酶的工程菌并进行了最佳酶表达条件的研究。结果表明:重组工程菌E.coliBL21(pTrc-gsh)表达量最高,质粒稳定性好;以5%接各量于37℃、pH7.2培养至OD550=0.5左右,加入诱导剂IPTG(0.mmol/L),同时切换培养条件为34℃、pH6  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
