The current study was to investigate mRNA expression of cyclin D2 and p16 during the transition from cardiac myocyte hyperplasia to hypertrophy. Cultured cardiac myocytes (CM) and fibroblasts (FC) obtained from 1-day-old Sparague-Dawley rats were used in this study. We have determined (1) hyperplasia by cell growth curve and fluorescence activated cell sorting (FACS); and (2) ultrastructure by electron microscope observation; and (3) expresions of cyclin D2 mRNA and p16 mRNA by using in situ hybridization and image analysis. The results were shown (1) Results of cell growth curve and FACS analysis showed CM could proliferate in the first 3 cultured days (4 days in postnatal development). But the ability decreased quickly, concomitant with the differentiation. (2) The ultrastructure of CM showed the large amount of myofilaments and mitochondrion and FC showed moderate amount of rough endoplasmic reticulum. (3) The expression of cyclin D2 mRNA in 3−, 4−, 5−day CM group was 0.89 times (p<0.05), 0.80 times (p<0.05) and 0.56 times (p<0.01) of that in 1-day group respectively. P16 mRNA in 2−, 3−, 4−, 5−day CM group were 1.63 times (p<0.01), 1.72 times (p<0.01), 1.99 times (p<0.01) and 2.84 times (p<0.01) of that in 1−day group respectively. It can be concluded that cultured neonatal rat cardiac myocytes could proliferate during the first 3 cultured days, but the ability of proliferation decreased, from the fourth day, concomitant with differentiation. Cyclin D2 and p16 have the key roles during the transition from myocyte hyperplasia to hypertrophy. 用H2O2损伤大鼠乳鼠的心肌细胞建立氧化应激损伤模型, 考察谷胱甘肽过氧化物酶模拟物Se-scFv-B3对H2O2诱导的大鼠乳鼠心肌细胞氧化损伤的影响. 结果表明, Se-scFv-B3能部分增加心肌细胞存活率, 减少细胞凋亡, 恢复线粒体膜电位, 下调Caspase-3活力并降低细胞内活性氧的含量. 表明Se-scFv-B3可以保护心肌细胞抵制H2O2诱导的氧化应激损伤.  相似文献

目的 采用长期精神紧张刺激建立动物抑郁症模型。方法 选用C57BL/6 J雄性4周龄小鼠17只,体质量(11. 05±1. 65) g,随机分为对照组、长期精神紧张刺激(LTMS)组。LTMS组单笼饲养并连续21 d每天给予猫惊吓3 h和剥夺睡眠12 h。实验组通过测定摄食量变化和高架十字迷宫实验、旷场实验、强迫游泳行为学变化来判断建模是否成功。结果 与对照组相比,LTMS组摄食量明显降低(P<0. 01),强迫游泳不动时间显著延长(P<0. 01)。旷场试验中,LTMS组小鼠穿越格数和直立次数均明显少于对照组,差异具有极显著性(P<0. 01)。高架十字迷宫实验中,LTMS组小鼠闭合臂停留时间明显延长,差异具有极显著性(P<0. 01);进入开臂次数比例减少,差异具有显著性(P<0. 05)。结论 长期精神紧张模型表现出行为绝望、活动能力下降、兴趣丧失、焦虑与抑郁症临床表现相似的症状,可作为抑郁症模型。  相似文献

在实验室建立了体外培养新生大鼠海马神经细胞方法,观察和研究枣仁皂甙A(JuA)对海马神经细胞生长的影响,用免疫组化和免疫荧光法鉴定神经细胞,M1rr检测神经细胞存活率。试验结果表明,培养细胞为高纯度的海马神经细胞,JuA组的海马神经细胞存活率明显高于对照组(p〈0.01),且呈现一定剂量相关性,提示JuA对体外培养的新生大鼠海马神经细胞的生长有一定的促进作用  相似文献

PURPOSE: To investigate the relationship between expression of cell cycle-related protein cyclin D1, p27kip1 and the pathogenesis of bronchioloalveolar carcinoma (BAC) and the value of prediction of prognosis. METHODS: Cyclin D1 and p27kip1 protein were detected by immunohistochemical En Vision method in 43 BACs. RESULTS: The positivity of cyclin D1 in BAC was 65.1% (28/43), which was significantly higher than that in normal pulmonary tissue (0/13), P<0.01. No statistically significant association was found between cyclin D1 expression data and sex, age, tobacco-use history, histologic subtype (mucinous vs nonmucinous), stromal fibrosis, lymph node metastasis, clinical stage or postoperative survival period (P>0.05), while cyclin D1 expression was found to be negatively correlated with tumor size (P<0.05). The positivity of p27kip1 in BACs was 51.2% (22/43), significantly lower than that in normal pulmonary tissue (12/13), P<0.01. p27kip1 expression level was not associated with sex, age, tobacco-use history, tumor size or histologic subtype (P>0.05), but was negatively correlated with stromal fibrosis, lymph node metastasis and clinical stage (P<0.05); and positively associated with postoperative survival period (P<0.01). The survival rate of p27kip1 positive group was significantly higher than that of p27kip1 negative group (P<0.01). No statistically significant correlation was found between cyclin D1 and p27kip1 expression. CONCLUSIONS: Increased cyclin D1 expression and decreased p27kip1 expression are related to the pathogenesis of BAC; decreased p27kip1 expression is associated with metastasis progression; immunodetection of p27kip1 is useful for assessment of prognosis.  相似文献

研究比较了两种不同类型的冷冻精液在解冻后精子活率、顶体完整率以及受精率方面的差异。实验结果表明,细管型冷冻精液解冻后的活率显著高于颗粒型(n=20,p<0.01)。细管型冷冻精液解冻后的顶体完整率显著高于颗粒型(n=20,p<0.05)。受精实验结果表明,两种类型的冷冻精液的受精率都显著低于以新鲜精液输精的对照组(p<0.01),颗粒型冷冻精液的受精率显著高于细管型冷冻精液(p<0.01)。  相似文献

报道对49例急性颅脑损伤病人检测伤后不同时间血浆脂质过氧化物含量(LPO)。结果表明损伤组血浆LPO含量显著高于对照组(P<0.001—0.05)。脑损伤后8天内,机体自由基水平明显升高,以伤后第4天最高(P<0.001—0.3)。死亡组血浆LP0含量显著高于生存组(P<0.1)。损伤程度与LPO含量呈密切相关(r=3.5.p<0.001)。提示急性颅脑损伤后颅内膜脂质过氧化反应剧增,血浆LPO含量检测对判断脑损伤程度和评估预后有一定的价值。  相似文献

研究初次就业个体在充满变革的组织与工作环境中的职业认同获得及其相关因素。对158名初次就业的组织员工采用了职业认同获得问卷、工作压力源问卷、工作经验来源问卷、自我效能信息来源问卷和中国人自我效能量表,共收回有效问卷152份。研究结果表明,职业认 同获得与工作压力源呈负相关,r=-0.476(p<0.01),与工作经验来源呈正相关,r=0.444(p<0.01),与自我效能信息来源呈正相关,r=0.407(p<0.01)。工作年限对于职业认同获得的间接效应之和为0.21,工作年限解释职业认同获得变异量的4.55%。工作年限对职业认同获得(F(2,151)=10.82,p<0.001)和角色冲突(F(2,151)=21.20,p<0.001)有显著影响,并可能影响它们之间的关系。研究结果提示,个体主动的探索和努力在职业认同获得过程中有着积极的作用。  相似文献

资源是育种的基础,对引进资源进行科学的评价是确定育种方向的前提,也是加快育种进度的关键.该研究通过田间试验(1 a)对引自哥伦比亚的54份木薯资源进行育种特性评价,通过对株高、块根直径、单薯鲜重、薯干率、鲜薯淀粉含量及支链淀粉含量等指标的测定和分析,确定54份资源的生产特性.结果表明:种质CM837、CM2399-4、哥伦比亚16P、19S、SG424-19、SM2300-1、SM2323-6、CMR34-11-3、CMR35-70-1和CMR35-70-1表现最好,是育种的优选材料;种质CM385-6、CM837、CM3993-9、CM4054-40、PER557、CMR26-07-15、CMR36-40-9、CMR37-14-9块根淀粉含量和生长情况较好但产量较低,可作为以高淀粉含量和以茎叶为饲料的育种材料;种质CM92-56-1、CM901、CM7595-1和SM28-80-3木薯产量和质量都较好,可作为以木薯为收获目标的育种材料.  相似文献

Ubiquitin-mediated proteolysis due to the anaphase-promoting complex/cyclosome (APC/C) is essential for separation of sister chromatids, requiring degradation of the anaphase inhibitor Pds1, and for exit from mitosis, requiring inactivation of cyclin B Cdk1 kinases. Exit from mitosis in yeast involves accumulation of the cyclin kinase inhibitor Sic1 as well as cyclin proteolysis mediated by APC/C bound by the activating subunit Cdh1/Hct1 (APC(Cdh1)). Both processes require the Cdc14 phosphatase, whose release from the nucleolus during anaphase causes dephosphorylation and thereby activation of Cdh1 and accumulation of another protein, Sic1 (refs 4-7). We do not know what determines the release of Cdc14 and enables it to promote Cdk1 inactivation, but it is known to be dependent on APC/C bound by Cdc20 (APC(Cdc20)) (ref. 4). Here we show that APC(Cdc20) allows activation of Cdc14 and promotes exit from mitosis by mediating proteolysis of Pds1 and the S phase cyclin Clb5 in the yeast Saccharomyces cerevisiae. Degradation of Pds1 is necessary for release of Cdc14 from the nucleolus, whereas degradation of Clb5 is crucial if Cdc14 is to overwhelm Cdk1 and activate its foes (Cdh1 and Sic1). Remarkably, cells lacking both Pds1 and Clb5 can proliferate in the complete absence of Cdc20.  相似文献

通过HPLC谱图比较不同产地丹参脂溶性成分含量的差异,采用MTT染色法测定缺氧、缺糖1、2、3 h后H9C2心肌细胞的成活率以建立缺氧缺糖模型组,以低、中、高剂量的丹参脂溶性成分加入模型组细胞,观察细胞的状态确定加药浓度。检测各组的乳酸脱氢酶(LDH)含量、总抗氧化活力(T AC)和细胞存活率(SR),以此作为考察丹参脂溶性成分保护作用差异的指标。结果表明,缺氧缺糖组以及不同产地丹参脂溶性成分治疗各组的SR与正常对照组相比有显著性差异(P<0.05),不同产地丹参脂溶性成分治疗各组的SR与缺氧缺糖组相比有显著性差异(P<0.05)。缺氧缺糖组以及不同产地丹参脂溶性成分治疗各组的LDH与正常对照组相比有显著性差异(P<0.05),不同产地丹参脂溶性成分各组的LDH含量与缺氧缺糖组相比有显著性差异(P<0.05)。缺氧缺糖组以及不同产地丹参脂溶性成分各组的T AC与正常对照组相比有显著性差异(P<0.05),不同产地丹参脂溶性成分各组的T AC与缺氧缺糖组相比有显著性差异(P<0.05)。丹参脂溶性成分明显减少心肌细胞缺氧缺糖损伤后LDH的溢出,提高了心肌细胞的T AC和SR,其对缺氧缺糖损伤后心肌细胞确实有修复保护作用,不同产地丹参中脂溶性成分对心肌细胞缺氧缺糖保护作用确实存在药效差别。  相似文献

以不同浓度H_2O_2刺激PC3细胞24h,应用MTT法分析细胞活力变化,并采用蛋白印迹方法检测HSP60蛋白、Akt蛋白(总Akt蛋白)和磷酸化Akt蛋白的表达水平.结果表明,15.625μmol/L的H2O2可诱导PC3细胞内HSP60表达显著升高(p0.05),7.312μmol/L和15.625μmol/L的H_2O_2均会导致磷酸化Akt蛋白的量显著下降(p0.01),但该两种浓度的刺激对细胞内总Akt蛋白量并无显著影响(p0.05).  相似文献

A new dynamic model for non-Fickian diffusion of calcium spark in cardiac myocytes was developed by introducing time lags on the basis of the microscale mass transport theory. Numerical simulation showed that the size of the calcium spark produced by the new dynamic model was larger than that of Fick diffusion and was in more agreement with experimental results. In addition, the time lags of the calcium spark in cardiac myocytes were about 0.1--0.8 ms. These results can be used to understand the mechanism of calcium spark diffusion in cardiac myocytes.  相似文献

目的:了解大学生的完美主义、自杀意念状况,分析影响大学生自杀意念产生的主要因素。方法:采用完美主义量表、自杀意念自评量表对河南省7所高校800名大学生进行统一问卷调查。结果:不同家庭类型大学生在完美主义及维度上存在显著差异(p<0.01),而在自杀意念上不存在显著差异(p>0.05);不同专业满意度大学生在完美主义上不存在显著差异(p>0.05),而在自杀意念上存在显著差异(p<0.001);不同专业大学生在完美主义及其中维度——父母期望、条理性和自杀意念上存在显著差异(p<0.05,p<0.01)。结论:高校教育工作者应采取措施消除完美主义的消极影响,降低自杀意念的产生。  相似文献

研究桂枝茯苓丸在人卵巢癌耐药性裸鼠肿瘤模型中对凋亡的诱导作用,探讨其可能的作用机制。采用实时无标记细胞分析系统检测SKOV3/DDP对顺铂和紫杉醇的耐药性,建立SKOV3/DDP卵巢癌耐药性BALB/c裸鼠肿瘤模型,模型裸鼠随机分为桂枝茯苓丸浓缩液16 g.kg-1.d-1(高)、8 g.kg-1.d-1(中)、4 g.kg-1.d-1(低)剂量组、PI3K抑制剂LY294002组、低剂量桂枝茯苓丸LY294002组(联用组)及对照组,给药10天后剖取肿瘤组织,采用流式细胞术检测肿瘤细胞的凋亡率,采用荧光定量PCR检测肿瘤组织中MTDH、PTEN mRNA的相对表达。结果表明,SKOV3/DDP细胞对顺铂和紫杉醇的耐药倍数分别为4.57和4.36倍。随着桂枝茯苓丸浓缩液剂量的增加,肿瘤细胞的凋亡率逐渐增加,上调PTEN mRNA和下调MTDH mRNA的作用逐渐增强,各组之间差异具有显著性(P < 0.01)。LY294002和低剂量桂枝茯苓丸联用后,抑制凋亡的作用强于二者分别单用的凋亡抑制作用,下调MTDH mRNA和上调PTEN mRNA的作用强于其他各组(P < 0.01)。说明在SKOV3/DDP卵巢癌耐药性BALB/c裸鼠模型中,桂枝茯苓丸能够剂量依赖性地诱导耐药性肿瘤细胞的凋亡,其机制可能与上调PTEN mRNA和下调MTDH mRNA有关。  相似文献
