目的:针对地衣初生、次生代谢产物丰富的特点,提出了一种适于地衣总DNA的提取方法。方法:应用改良的CTAB法,对3种常见地衣进行了总DNA提取,经过琼脂糖电泳、紫外吸收分析DNA质量。结果:3个样本DNA的A260/A280值均在1.8~2.0之间,琼脂糖电泳图像清晰。结论:改良的CTAB法简便、省时、价廉,适合于地衣总DNA的提取,获得的DNA含量高,纯度好。  相似文献

以酒精酵母,IFFI1300 材料,建立了以”蜗牛酶+机械振荡“复俣自理破碎细胞的方法;先提取并纯化线粒体,后从线粒体中提取线粒体DNA的方法,得到了紫外吸收A260/280为1.823,琼脂糖电泳为一条带且分子量大λDNA的线粒体DNA。  相似文献

用碱变性法和核酸酶消化法从黑斑蛙的肝脏和肌肉中提取和纯化线粒体DNA,经琼脂糖凝胶电泳及OD值测定显示,所得黑斑蛙线粒体DNA的结构完整,并避免了核DNA、线粒体RNA、蛋白质、有机溶剂等的污染.结果表明:黑斑蛙mtDNA的分子量大小约为17.167kb.其得率和纯度均能满足mtDNA的各种分析要求.  相似文献

为了更加快速地获得糯米酒发酵过程中高质量的微生物基因组DNA,本实验通过测定基因组DNA浓度和进行聚合酶链式反应(PCR),利用溶菌酶-SDS-蛋白酶-K(A法),改良CTAB(B法)和改良CTAB-溶菌酶(C法)三种实验方法获得微生物基因组DNA,通过比较三种方法发现,C法提取效率最高,获得的基因组DNA浓度为1 573.97±711.51ng/μL;B法获得的基因组浓度为1 436.03±128.0ng/μL;A法效率最低,获得基因组浓度为411.67±133.29ng/μL.以基因组DNA为模板进行PCR,将扩增产物在0.6%琼脂糖凝胶中进行电泳检验.结果显示C法获得的条带最清楚明亮,表明DNA浓度最高、结构完整.基因组DNA浓度和PCR产物琼脂糖凝胶电泳条带的亮度表明,提取糯米酒发酵过程中微生物基因组DNA的最优方法为改良CTAB-溶菌酶法(C法).  相似文献

采用动物基因组DNA快速抽提试剂盒法、SDS法、CTAB法、NaCl法4种方法提取摇蚊基因组DNA.通过紫外分光光度计分别检测所得样品DNA在波长为260nm和280nm的吸光值,根据A260/A280的比值检测其纯度和浓度.采用PCR技术获得摇蚊COI基因,用琼脂糖凝胶电泳检测PCR-COI.综合分光光度法与电泳结果筛选出最适合提取摇蚊基因组DNA的方法.结果表明:SDS法所得样品DNA纯度最高,试剂盒法所得DNA浓度最大.同时综合比较DNA纯度与浓度分析得出:试剂盒法SK8222(改良)提取摇蚊基因组DNA为最佳方法,其次为SDS法或CTAB法(改良).  相似文献

以兰考泡桐组织培养苗叶片为材料,分别采用CTAB-Ⅰ、CTAB-Ⅱ、SDS-Ⅰ、SDS-Ⅱ四种方法提取叶片DNA,并对不同提取结果进行紫外分光光度、电泳、酶切、RAPD等方法的鉴定。结果表明,用四种方法提取的泡桐叶片DNA得率约为3.5-5.2μg/10mg,A260/A280约为1.7-1.8,A260/A230在2.0以上,分子量在23Kb以上,能够被EcoRⅠ、HindⅢ酶切,能够进行RAPD分析。综合考虑得率、纯度、质量等因素,以SDS-Ⅰ法为最佳方案。  相似文献

以披碱草属老芒麦(Elymus sibiricus)为材料,分别用简易提取法、十六烷基三乙基溴化铵(CTAB)法、十二烷基硫酸钠(SDS)法提取其基因组DNA,利用分光光度计测定其OD260与OD280的吸光值,计算比率估计核酸的纯度和浓度,并利用0.8%琼脂糖凝胶电泳观察DNA片段在凝胶中的位置.结果表明,3种方法都可以提取到基因组DNA,但SDS法所提取的DNA质量明显优于简易提取法和CTAB法,且分离的DNA产率高,纯度高,质量好,纯化后电泳检测带型清晰.因此,SDS法是实验室提取老芒麦基因组DNA的较有效而实用的方法.  相似文献

利用吸光值、琼脂糖电泳及PCR差异扩增等指标,对酚一氯仿法、水煮法、碱裂解法3种方法制备的鳗鲡病原菌DNA模板进行了差异分析.结果表明:1)不同方法提取的病原菌模板DNA的浓度、纯度及分子质量大小存在差异,但以这3种方法提取的病原菌DNA为模板,均能成功扩增到细菌16SrD.NA和外膜蛋白的保守序列.其中,酚一氯仿提取的模板DNA浓度较低、纯度较高;水煮法和碱裂解法提取的模板DNA浓度较高、纯度较低.2)电泳显示,3种方法获得的模板DNA扩增出的16SrDNA保守序列条带均一,没有显著差异,但在外膜蛋白保守序列的扩增中却因菌种和模板提取方法的不同而存在差异.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
