Allotransplantation of fresh, 1 or 2 week cultured parathyroid glands from Wistar rats (AgB2) to Fischer rats (AgB1) resulted in prompt rejection of the transplant in the muscle site; whereas transplantation into the adrenal site offered slightly prolonged survival, suggesting that the latter is a privileged transplantation site.  相似文献

Summary In rats working in a behavioural schedule of differential reinforcement of low rates (6 or 10 sec), picrotoxin (1 mg kg–1) decreased the number of premature responses and increased (in DRL 10 sec only) the number of rewarded responses. The effect of picrotoxin was antagonized by diazepam (2 mg kg–1). In contrast to picrotoxin, strychnine (1.5 mg kg–1) increased the number of premature responses.This work was supported by a grant of I.N.S.E.R.M. (ATP 39-76-71).  相似文献

Summary The blood pressure and heart rate responses to intravenous dopamine infusion at 2.5, 5.0 and 10.0 g·min–1·100 g–1 were studied in conscious and pentobarbital-anesthetized Sprague — Dawley rats. In the conscious rats, dopamine caused a significant dose-related increase in the mean arterial blood pressure which was abolished in the anesthetized rats. The heart rate increased significantly only at the highest dose infused. The responses to equipressor doses of noradrenaline (40 ng·min–1·100 g–1) and phenylephrine (1.0 g·min–1·100 g–1) were also suppressed in the anesthetized rats. The results suggest that pentobarbital anesthesia depresses the blood pressure response to dopamine infusion in the rat through a depression of activation of alpha-adrenoceptors.16 June 1986  相似文献

The renal effects of an unsaturated fat (UNSAT) diet in mild to moderate two-kidney, one-clip (2K1C) renovascular hypertension were evaluated. An UNSAT diet (37% by energy) prevented the development of hypertension compared to 2K1C rats fed a high saturated fat (SAT) (37% by energy) and a normal fat (CONTROL) (11% by energy) diet. Urinary sodium and fractional sodium excretion increased in 2K1C rats as compared to SHAM operated controls, regardless of the diet received. In the early weeks of the experiment (weeks 2–4 post-surgery to induce hypertension), an enhanced natriuresis occurred in the 2K1C UNSAT as compared to the 2K1C CONTROL and SAT diet groups. This resulted from an increase in the glomerular filtration rate (GFR in mls·min–1) as measured using the single-injection [51Cr] EDTA method (2K1C UNSAT; 1.99±0.18 versus 2K1C SAT; 1.27±0.09, p<0.02; and versus SHAM CONTROL; 1.45×0.01; p<0.02). The increased GFR was not associated with alterations in effective renal plasma flow (ERPF) as measured using the single-injection [125I] Na hippurate method. No differences in sodium excretion; GFR; ERPF or renal blood flow (microsphere technique) were noted between the 2K1C UNSAT and SAT diet groups at weeks 6–8 post-surgery, despite a continued antihypertensive effect of the UNSAT diet. Hence, the antihypertensive effect of an unsaturated fat diet in 2K1C renovascular hypertension in rats is associated with transient glomerular changes leading to an enhanced natriuresis.  相似文献

Summary In perfused livers of 24 hour-fasted rats, PGE1 (prostaglandin E1) infused continuously into the perfusate, was found to cause a 45% increase in the incorporation of 1-14C acetate into liver fatty acids. (6R*)-{(2S*)-2-hydroxyheneicos-12-enyl}-5,6-dihydro-2H-pyran-2-one (1)o is the major constituent of the secretion of freshly dissected Dufour's gland of the urticating antTetramorium aculeatum. In solution, compound 1 is slowly transformed into (1S*, 5R*, 7S*)-7-(nonadec-10-enyl)-2,6-dioxabicyclo[3.3.1]nonan-3-one (2)o on standing. The structures of compounds 1 and 2 have been established on the basis of their spectral and chemical properties. Summary Propanedioldehydrase is shown to convert both (+)-(S)-2-2H-propanediol,3, and (–)-(R)-1-2H2-propanediol,5, to specimens of deuterated propion-aldehyde, for which the (S)-configuration has been established. Thus, in the propanedioldehydrase reaction migration of hydrogen atoms from C–1 to C–2 always occurs with inversion of configuration.  相似文献

Summary It was found that ouabain (10–5 M) was effective in releasing the extraneuronal catecholamine which was taken up by uptake2 process in the guinea-pig vas deferens. This result shows that a Na–K ATPase is essential for the storage of catecholamine in the extraneuronal site.  相似文献

Effects of 17-estradiol (E2) in vitro on Na-dependent Ca2+ efflux from, and depolarization-induced Ca2+ uptake into, the nerve cell were studied with the use of synaptosomes isolated from the brain stem, mesencephalic reticular formation (MRF), caudate nucleus and the hippocampus of long-term ovariectomized adult female rats. It was found that E2 (1) at a concentration of 10 nM or lower, stimulates Na-dependent Ca2+ efflux in the caudate nucleus and hippocampus, and does not affect the efflux in MRF and brain stem; (2) at concentrations above 10 nM has no effect on the Ca2+ efflux in any of the four structures investigated; and (3) produces a biphasic effect on the depolarization-induced Ca2+ uptake, increasing it in all structures except MRF at 10 nM concentration, and decreasing it at concentrations higher than 10 nM, irrespective of the structure investigated. These results suggest that E2, acting at extranuclear sites, modulates synaptic transmission via alterations of Ca2+ transport mechanisms in nerve endings.  相似文献

Summary Autoradiographs of the kidneys of rachitic rats dosed in vivo with 250 pmoles 25-hydroxycholecaliferol (25-OHD3) and3H-leucine showed increased grain counts in portions of proximal renal tubules. On incubation of kidney slices the synthesis of only 1 cytosol protein was found to be stimulated by the steroid. Summary The investigation deals with the successful heterologous transplantation of the Mouse Crocker-Sarcoma on young rats, that were pretreated with the radioactive isotopes P32 and Au198. Summary Tritiated -(dimethylbenzimidazolyl)-Co-5 deoxyadenosyl-cobamide, as obtained in the propanediol dehydrase reaction from (RS)-1-3H-propane-1, 2-diol, is shown to transfer substantial amounts of radioactivity to the substrate when used as a catalyst for the reversible enzymatic conversion of methylmalonyl-CoA to succinyl-CoA. Summary As judged from 2-h blood level curves, adult female rats absorbed more FeII per cm2 of gross duodenal mucosa than adult male rats. By contrast, the 2-h blood level curves per cm2 of mucosa of proximal jejunum did not differ significantly in male and female rats although in both sexes, iron was absorbed more efficiently from the duodenum. Summary The role of vasoactive intestinal peptide (VIP), as a possible neurotransmitter of the intrinsic nerve plexus in the guinea pig gallbladder, was investigated by monitoring spontaneous contractile activity. VIP receptor antagonist (4 Cl-D-Phe6, Leu17)-VIP did not produce any effect on muscular tone and spontaneous activity, whereas (N-Ac-Tyr1, D-Phe2)-GRF-(1-29)-NH2, (14-GRF analog), which is known to stimulate digestive enzyme secretion by interacting with the VIP-preferring receptors, greatly increased the amplitude and frequency of waves as well as the muscular tone. Since VIP receptor antagonist acts selectively as a competitive antagonist for the action of VIP, we conclude that the gallbladder inhibitory intrinsic plexus neurotransmitter is not VIP, but a member of the glucagon-secretin family of peptides.  相似文献

Summary The administration of 15(R)-15-methyl prostaglandin E2 (15(R)-15-M-PGE2) in vivo significantly diminished the uptake of59Fe into blood, spleen, liver, femur and dried intestine of rats, whereas acetylsalicylic acid (ASA) increased the counts significantly. This effect of ASA was counteracted by 15(R)-15-M-PGE2. It is suggested that prostaglandins (PGs) might play an important role in inhibiting iron absorption at the intestinal level.This work was supported by grant No.6638 from CONICET (Argentina). The proliferation ability of satellite cells (considered the 'stem cells' of mature myofibers) declines with increasing age when cultured under standard cell culture conditions of 21% oxygen. However, actual oxygen levels in the intact myofiber in vivo are an order of magnitude lower. No studies to date have addressed the issue of whether culturing satellite cells from old muscles under more 'physiologic' conditions would enhance their proliferation and/or differentiation ability. Therefore, we analyzed satellite cells derived from 31-month-old rats in standard cultures with 21% O2 and in lowered (∼3%) O2. Under the lowered O2 conditions, we noted a remarkable increase in the percentage of large-sized colonies, activation of cell cycle progression factors, phosphorylation of Akt, and downregulation of the cell cycle inhibitor p27Kip1. These data suggest that lower O2 levels provide a milieu that stimulates proliferation by allowing continued cell cycle progression, to result ultimately in the enhanced in vitro replicative life span of the old satellite cells. Such a method therefore provides an improved means for the ex vivo generation of progenitor satellite cell populations for potential therapeutic stem cell transplantation.
