鸡传染性法氏囊病的RT—PCR诊断方法的研究

在研究鸡法氏囊病毒（HBDV）的A片断cDNA结构的基础上，建立了RT－PCR早期诊断技术，结果表明，使用PrimerDesign引物设计软件，在VP5和VP2的重叠基因区设计的一对引物具有非常高的同源性和保守性，采用的四种IBDV的标准毒株，一种野毒株和一例病鸡标本通过此引物进行RT－PCR检测均得到了明显的阳性扩增结果，而两种其他鸡病病毒和一例健康鸡的法氏囊组织扩增均为阴性结果。     

BICP0激活三种复杂牛反转录病毒LTR

BICP0在病毒生活周期中具有重要功能，参与对病毒以及宿主的许多启动子的激活．BHV—1超感染所引发的BIV基因表达水平的提高即与该蛋白有关．为探究其激活特性，选取三种典型牛反转录病毒BIV、BLV和BFV，对其LTR上NF—KB及AP—1位点进行单独或共缺失，通过Luc瞬时表达分析发现：BICP0可激活所选三种牛反转录病毒的启动子，且激活功能与其NF—κB和AP—1位点有关．同时发现：在BIV中BICP0介导的两转录因子间具有相互协同作用，而在BFV中却表现相互抑制。     

反义PARP基因真核表达重组体的构建及其HeLa转化细胞系的建立

利用DNA体外重组技术将PARP基因cDNA部分序列反向克隆到真核表达载体pMAMneo上,构建成重组质粒pMAMneo－C1．4和pMAMneo－C0．3．将重组质粒pMAM－neo－C1．4转染HeLa细胞,经G418筛选,建成细胞系HeLa－C1．4－neo,Southern杂交结果表明,外源PARP基因cDNA部分序列已稳定整合到受体细胞基因组中,该细胞系的建立为研究聚ADP核糖基化作用在细胞内的功能打下了基础．     

运用HSV—tk／ACV系统进行肿瘤基因治疗研究

构建了含ｐｇｋ启动子驱动的ＨＳＶ－ｔｋ基因的反转录病毒载体ｐＬＮＴＫ，将ＨＳＶ－ｔｋ基因转移至人脑胶质瘤ＳＨＧ４４细胞及小鼠黑色素瘤细胞Ｂ１６。体外实验证实核苷类似物ＡＣＶ对ＳＨＧＬＮＴＫ细胞和Ｂ１６ＬＮＴＫ细胞的杀伤敏感性分别高于亲本高于１０００和４００倍。转ＨＳＶ－display status     

人p16^INK4cDNA探针在非小细胞肺癌研究中的应用

制备总RNA,RT－PCR克隆p16^INk4cDNA,测序验证,制备探针,进行PCR产物Southern杂交检测非小细胞肺癌组织标本中p1^^INK4基因第二外显子阴性杂交率为12．9％（4／31）,原位杂交显示p16^INK4基因转录阴性率为22．6％（7／31）,结果说明克隆的p^INK4cDNA是正确的。可用于临床基因诊断,p^16INK4基因变异及表达在非小细胞肺癌的、发展中起作用。     

草鱼生长激素基因在COS7细胞中转染表达的研究

应用RT-PCR技术克隆草鱼生长激素(gcGH)的cDNA,将此cDNA定向插入真核表达载体VR1020中,构建成重组真核表达质粒VgcGH.利用脂质体法使质粒VgcGH转染哺乳动物细胞COS7,对转染后的COS7细胞进行RT-PCR、ELISA和免疫荧光检测,分别在转录和翻译水平证实gcGH基因在COS7细胞中得到了持续和正确的转染表达.     

亚洲一型口蹄疫病毒YNAs1.1株结构蛋白VP1基因的核苷酸序列分析

提取BHK21细胞增殖的亚洲一型口蹄疫病毒（foot-and-mouth disease virus Serotype Asial)强毒株YNAs1.1的RNA,用一对引物P7,P13经反转录（RT）－PCR法扩增了约674bp的DNA片段。克隆目的基因后,采用双脱氧DNA链末端终止法测得了YNAs1.1的VP1基因36－633核苷酸序列。分析表明,病毒VP1基因的核苷酸序列与以色列以及印度已报道的Asia1型FMDV的同源性分别为82.11%与88.07%,对应的氨基酸序列同源性为87.94％与93.47％。该序列在GeneBank登陆号为AF241566。     

逆转录病毒载体设计对ANTI—RAS核酶基因转移效率的影响

将ａｎｔｉ－ｒａｓ核酶基因Ｒ８克隆于不同的逆转录病毒载体，并导入逆转录病毒包装细胞质ＰＡ３１７，得到具一次感染性的缺失性病毒，通过测定病毒滴度选择Ｒ８基因的高效重组转录病毒载体。     

PDXl与PAX4双基因表达腺病毒载体的构建与表达

运用pADxsi系统制备可表达人PDX1与PAX4双基因的重组5型腺病毒。从pEGFP-N1-PDX1质粒上酶切下目的基因PDX1并酶连到腺病毒穿梭质粒pShuttle-EGFP-CMV上,替换EGFP而得到pShuttle-CMV-PDX1;再将PAX4从pEGFP-N1-PAX4质粒上酶切下连到pShuttle-CMV-PDX1的多克隆酶切位点而得到穿梭质粒pShuttle-CMV-PDX1/CMV-PAX4;双酶切pShuttle-CMV-PDX1/CMV-PAX4将CMV-PDX1/CMV-PAX4转移到ADxsi骨架质粒上,得到pADxsi-CMV-PDX1/CMV-PAX4腺病毒质粒,然后在293细胞中进行包装并扩增重组腺病毒,并进行病毒滴度测定;体外感染人间充质干细胞。依据酶切、测序和PCR的结果均证明重组人PDX1与PAX4双基因表达腺病毒载体构建正确;而RTPCR与WB结果也显示目的基因在细胞中稳定表达。应用重组技术成功构建人PDX1与PAX4双基因表达5型腺病毒载体,转录因子PDX1与PAX4在间充质干细胞内稳定表达且定位于细胞核内。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
