自体造血干细胞移植后的胸腺近期输出功能   总被引:1,自引:1,他引:0  
目的:了解血液病患者接受自体造血干细胞移植(auto-HSCT)后外周血中的T细胞受体重排删除环(TRECs)水平,从而了解其胸腺近期输出功能和T细胞免疫重建情况。方法:采用实时定量PCR法检测9例血液病患者经auto-HSCT治疗后外周血单个核细胞(PBMNCs)中的TRECs水平;22例正常人外周血样本作为对照。结果:PBMNCs中的TRECs水平有着较大的个体差异,年龄与PBMNCs中的TRECs水平呈负相关;auto-HSCT组的每1 000个PBMNCs中的TRECs平均拷贝数(2.533±4.194)明显低于正常组(4.367±3.626)(P<0.05),但部分病例的TRECs水平已基本达到正常水平。结论:在一定时间内,auto-HSCT后的胸腺近期输出功能仍较低,不同个体T细胞免疫重建时间存在差异。  相似文献

目的:分析免疫性血小板减少症(ITP)中T细胞活化信号通路相关分子Cbl-b和c-Cbl基因的表达情况.方法:利用SYBR Green实时荧光定量聚合酶链式反应(RT-PCR)方法检测18例ITP患者外周血单个核细胞(PBMC)中Cbl-b和c-Cbl基因的表达水平,20例健康成人作为对照.结果:ITP组Cbl-b的表达水平(中位数为0.091%)与健康成人组Cbl-b的表达水平(中位数为0.366%)相比,明显降低(P0.001);而c-Cbl基因在ITP组中的表达水平(中位数为0.372%)与健康成人组的表达水平(中位数为0.298%)相比没有统计学差异,但c-Cbl基因在ITP女性患者样本中的表达水平(中位数为0.236%)明显低于男性患者样本c-Cbl的表达水平(中位数为0.479%)(P=0.039).结论:ITP患者外周血中Cbl-b表达下调,可能是其T细胞免疫异常活化的原因之一.  相似文献

目的:析再生障碍性贫血(AA)和真性红细胞增多症(PV)患者外周血T细胞中TCR ζ链基因表达水平,从而了解患者T细胞活化过程的相关变化情况.方法:利用SYBR Green Ⅰ荧光定量PCR,相对定量检测25例AA患者、12例PV患者及30例正常人的外周血单个核细胞(PBMCs)的TCR ζ链基因表达情况,β2微球蛋白基因(β2M)为内参照,根据相对定量公式:2- △△Ct计算患者与正常人TCR ζ链基因表达差异倍数.结果:与正常人相比,TCR ζ链基因表达水平有40% AA(10例)和41.7% PV(5例)患者呈表达上调,其它的(60?和58.3%PV患者)则呈表达下调情况,TCR ζ链基因表达水平的变化与患者年龄无相关性.结论:AA和PV患者可能存在T细胞活化相关的信号通路紊乱的情况,TCR ζ链基因表达模式改变的意义仍需进一步研究.  相似文献

目的:在慢性粒细胞白血病病人(CML),CD3ζ链表达明显下降,分析与CD3ζ存在互补关系的FcεRⅠγ基因在CML患者中表达水平,以了解T细胞免疫中TCR信号转导的变化情况.方法:利用SYBR Green Ⅰ实时荧光定量RT-PCR法测定CML患者和健康人各20例的外周血单个核细胞(PBMCs)的FcεRⅠγ基因表达水平,以β2微球蛋白基因(β2MG)作为内参照,采用相对定量公式:2-△Ct×100%,计算FcεRⅠγ链的相对mRNA表达量.结果:CML患者PBMCs中FcεRⅠγ基因表达水平明显高于健康对照组(P<0.001).FcεRⅠ γ表达水平变化与病人外周血CD3+T细胞比例无相关性.结论:CML病人中FcεRⅠ γ表达上升,提示FcεRⅠγ可能在一定程度调节CD3ζ链缺陷所带来的T细胞免疫异常.  相似文献

目的:检测不明原因病毒性脑炎(VE)患者外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中博尔纳病病毒(Boma Disease Virus,BDV)p24基因,探讨BDV感染的病毒性脑炎患者临床特征.方法:用荧光定量巢式逆转录聚合酶链反应(FQ-n RT-PCR)方法检测病毒性脑炎患者及正常体检者PBMCs中BDV p24基因片段,同时用β-肌动蛋白(β-actin)作为内参照,总结出临床特征.结果:52例病毒性脑炎PBMCs标本BDV p24基因片段FQ-nRT-PCR检出率13.46%(7/52),拷贝数>102kb/μl.对照组30例正常体检者均为阴性(0%,0/30),经比较两者间阳性率差异有显著性(P<0.05).BDV p24阳性脑炎患者主要以精神行为异常为临床特征.结论:贵州省遵义市部分病毒性脑炎的发生与BDV感染有关,主要以精神行为异常为临床特征.  相似文献

目的:建立体外扩增系统性红斑狼疮(SLE)患者外周血γδT细胞的方法,并初步分析其表型和功能,探索γδT细胞在SLE发病中的作用。方法:采用单克隆抗体固相法,对15例SLE患者和8例正常人外周血γδT细胞进行体外扩增建系,以流式细胞仪检测γδT细胞表型,并用MTT法观察γδT细胞对Daud i细胞的细胞毒作用。结果:建立了SLE患者外周血γδT细胞系,其平均纯度为(58.1±11.2)%,较正常对照组(80.3±9.2)%偏低(P<0.05);其细胞表型为:Vδ1(34.4±24.5)%、Vδ2(61.9±28.6)%、Vδ3(16.1±10.6)%、Vγ9(76.4±11.8)%,其中Vδ2表达较对照组降低,而Vδ1和Vδ3表达增加(P均<0.05);其细胞毒作用在二组间无明显差别。结论:SLE患者外周血γδT细胞的Vδ、Vγ基因的取用表达存在差异,提示其在SLE的发病中可能起一定作用。  相似文献

目的:检测T细胞免疫球蛋白粘蛋白分子-1在女性系统性红斑狼疮(SLE)患者外周血的表达情况,并探讨与SLE疾病活动的相关性.方法:采用酶联免疫吸附测定(ELISA)方法检测SLE治疗前后和健康对照组外周血中T细胞免疫球蛋白粘蛋白分子-1的表达量,SLE活动性以系统性红斑狼疮病变活动指数(SLEDAI)表示,并进行相关性分析以明确T细胞免疫球蛋白粘蛋白分子-1与系统性红斑狼疮患者疾病活动性的关系.结果:治疗前SLE患者外周血中T细胞免疫球蛋白粘蛋白分子-1的表达水平明显高于对照组和治疗后SLE患者外周血中的表达水平,有统计学差异(P0.05),且其表达水平与SLEDAI评分呈正相关(r=0.639,P0.01).结论:T细胞免疫球蛋白粘蛋白分子-1的表达水平与SLE的发病有关,且可反映SLE患者的疾病活动状态.  相似文献

目的:检测急性髓性白血病(AML)患者外周血中CR Vβ2-、Vβ5-和Vβ17-Dβ1 sjTRECs的存在情况。方法:利用半巢式PCR扩增31例不同亚型AML患者外周血单个核细胞DNA中TCRB基因重排时形成的T细胞受体DNA删除环(sjTRECs)中的Vβ2-、Vβ5-和Vβ17-Dβ1 sjTRECs,10例正常人外周血作为对照。结果:TCR Vβ2-、Vβ5-和Vβ17-Dβ1 sjTRECs在正常人和AML患者中均可检测到,AML患者三者的检出率分别为45.16%、41.94%和32.26%,均略低于正常人,但差别未显示出统计学意义。≤30岁的AML患者3种Vβ sjTRECs的检出率均高于30岁以上的患者,以Vβ2-Dβ1sjTRECs最明显。结论:率先提供AML患者外周血T细胞中TCR Vβ2-、Vβ5-和Vβ17-Dp1 sjTRECs的存在情况,为研究AML患者胸腺近期各Vβ亚家族幼稚(naive)T细胞的输出情况提供资料。  相似文献

目的探讨锰超氧化物歧化酶(manganese superoxide dismutase,MnSOD)在乳腺癌患者和正常人外周血中的表达差异及其临床意义.方法采用实时荧光定量PCR方法检测218例乳腺癌患者和77例正常女性外周血中MnSOD基因表达水平,并进行分析.结果乳腺癌患者外周血中MnSOD基因表达水平为4.38±0.74,正常女性外周血中MnSOD基因表达水平为4.16±0.64,P0.001,有统计学意义.淋巴转移患者组和临床早期患者组与正常女性组比较,P0.001.结论淋巴转移和临床早期乳腺癌患者外周血中MnSOD基因表达水平显著低于正常女性,提示MnSOD的低表达与肿瘤的发生和发展有关.应用荧光定量PCR方法检测外周血MnSOD基因水平对监测乳腺癌有一定意义.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
