Demonstration of a receptor for 5 alpha-androstane-3 beta, 17 beta-diol in the cytosol of the anterior pituitary of prepubertal male rats]

The presence of a specific receptor for 5alpha-androstan-3beta, 17beta-diol (3beta-diol) in the pituitary cytosol from prepubertal male rats was demonstrated. Its characteristics were: Ka = 5.2.10(7) M-1 KD = 1.9 X 10(-8) M, number of specific binding = 8.7 10(-14) moles per mg of proteins. Its sedimentation constant was 3 S. Competition assays showed that only 3beta-diol itself and estrogens were able to compete for the binding sites for 3beta-diol. Androgens, including 3alpha-diol, were inefficient. This receptor was found only in pituitary cytosol, it was missing from hypothalamic or cortical cytosols. This special localization seemed to foreshadow a specific role for 3beta-diol in the anterior hypophysis.

Incubation of 4-14C-5 alpha-androstane-3 beta, 17 beta-diol with human normal and hyperplastic prostate minces led to the NADPH-dependent production of a major polar radiometabolite which was identified as 5 alpha-androstane-3 beta, 7 alpha, 17 beta-triol and proved the presence of a 3 beta-hydroxysteroid-7 alpha-hydroxylase in prostate. Such evidences may bring new leads for the study of the mechanisms of androgenic steroid action in the prostate gland.

The experiments carried out showed the presence- in sympathetic nerve endings ot the carotid sinus- of alpha and beta adrenoceptors which by means of respective negative and positive feedback processes, modulated NA release induced by a sympathetic nerve stimulation. Similarly, Pgs acted by means of a negative feedback mechanism to regulated adrenergic neuro-transmission in carotid sinus but they could not be considered as the chemical mediators of either alpha or beta adrenoceptors.

The intravenous injection that was given to rabbits which consisted of 0,5 mg of a common heptapeptide to ACTH4-10, BETA MSH11-17 hormones, and to beta lipotropic47-53 hormone (beta LPH47-53) was followed at the end of an hour by an augmentation of 70% of total lipids, and at the end of two hours by a lowering of calcemia (around 27%) and of phosphoremia (20%). The injection of a tetrapeptide corresponding to only ACTH7-10 bound to a nitrobenzylhydralamin and in presence of a Freund adjuvants has provoked the formation of corresponding antibodies.     

Alteration in the secretion of anterior pituitary hormones during dimethylbenzathracene induction of mammary tumors in rats]

In a sensitive strain of Rat (Sprague-Dawley) a deregulation of the secretion of some pituitary hormones was noted in the female during induction of mammary tumors by dimethylbenzanthracene administration. The deregulation occurred quickly and depended on the stage of the estrous cycle. It mainly consists of hyposecretion of gonadotropins. TSH and GH and of hypersecretion of prolactin.     

Effect of hypothalamic crude extract on the utilization of D-[14C]-glucose by the anterior pituitary,in vitro

Resumen Los efectos de extractos crudos de hipotalamo en la utilizacion deD-[¹⁴C]-glucosa por hipófisis anteriores fueram estudiados in vitro en condiciones identicas a aquelas en las cuales los extractos inducen la liberación de hormonas al medio de incubación. Los resultados indican que los extractos hipotalámicos aumentan la producción de¹⁴CO₂ por la hipófisis a partir de glucose y la incorporación de¹⁴C en proteinas del medio de incubación. Los extractos no modificaron la cantidad de¹⁴C glucose utilizada para sintesis de proteinas tissulares.

Investigations regarding the chemical composition of the volatiles in male mouse urine have recently enabled the structural elucidation of a hitherto unreported urinary component, 7-exo-ethyl-5-methyl-6,8-dioxabicyclo[3.2.1]-3-octene. This compound's uniqueness to mouse urine and its dependence on testosterone levels in the male suggest its probable role as a mouse pheromone or pheromone adjuvant.
