利用RT-PCR技术鉴定人ZP3嵌合肽基因在昆虫细胞中的表达   总被引:3,自引:0,他引:3  
目的:探讨在转录水平上对人卵透明带蛋白3(hZP3)嵌合肽在昆虫细胞中的表达鉴定.方法:用含目的基因的重组病毒感染昆虫细胞,用LiCl法提取RNA,以其为模板,利用一步法RT-PCR反转录出目的DNA.结果:琼脂糖凝胶电泳显示其PCR产物只有一条DNA条带且与目的基因大小一致,而阴性对照未见任何条带.结论:hZP3嵌和肽基因在重组病毒感染的昆虫细胞中成功转录,初步验证了目的基因的表达.  相似文献

目的:用毕赤酵母(Pichia pastoris)表达人透明带hZP3片段,研究其抗生育作用.方法:设计特定引物从hZP3全长序列中PCR扩增794~1282nt基因片段,将扩增片段克隆到酵母表达载体pPICZα上, 构建成重组质粒pPICZα-hZP3CT,线性化后的重组质粒导入毕赤酵母中,筛选出高拷贝菌株,甲醇诱导目的蛋白表达.用SDS-PAGE 和Western blotting 分析表达产物.结果:成功构建酵母表达载体,并在酵母菌株X-33中诱导表达重组蛋白.重组蛋白可以与鼠抗重组人卵透明带hZP3融合肽段的抗血清发生特异结合,表明目的基因成功表达.结论:重组hZP3蛋白已在酵母中成功表达,目的蛋白能与相应的抗体结合.  相似文献

人ZP3B-细胞表位嵌合肽DNA的设计和合成   总被引:2,自引:0,他引:2  
目的:设计和合成适合在昆虫细胞中表达的一个人ZP3 B细胞表位的DNA序列。方法:根据国外的研究基础,设计由人ZP3的B细胞表位肽段和外源Th细胞表位肽段串联而成的45肽嵌合肽序列,并根据昆虫细胞对密码子的偏爱性设计出该嵌合肽的DNA序列。然后人工合成该嵌合肽的DNA链,并克隆到PUC18载体上。结果:通过酶切分析和测序证明,合成的DNA与所设计的嵌合肽DNA序列一致。结论:按设计合成了人ZP3的  相似文献

为了研究猪和鱼卵透明带抗原的免疫交叉反应性,应用免疫印渍术对其抗原组分进行了检测.实验结果表明,鱼卵透明带抗原与已知具有精子受体活性的猪ZP3抗原,甚至猪全ZP抗原均没有交叉反应.提示鱼卵透明带不含有与猪和人等哺乳类的透明带免疫学相类似的抗原决定簇.  相似文献

壳聚糖-卵透明带DNA纳米微囊的制备及其体外表达   总被引:4,自引:0,他引:4  
目的:发展壳聚糖-卵透明带口服DNA粘膜免疫避孕疫苗。方法:首先利用基因工程技术构建含卵透明带特异抗原pZP3α DNA的真核质粒表达载体pVAX1-pZP3α,然后用壳聚糖C390包被pVAX1-pZP3α重组质粒制备免疫微囊,测定免疫微囊对质粒DNA的包裹率,用原子力显微镜(AFM)观察壳聚糖-DNA微囊的形态结构,并体外转染HeLa细胞,用间接免疫荧光法(IIF)检测pZP3α的瞬时表达。结果:pZP30αDNA正确插入到载体pVAX1中并与壳聚糖C390形成纳米免疫微囊,对重组质粒pVAX1-pZP3a的包裹率达90%以上,AFM成像技术显示这些微囊的直径为100nm到300nm不等,平均为224.6nm,形状为球形、椭圆形等。体外转染HeLa细胞用IIF法检测到了抗原pZP3α在细胞内的表达。结论:壳聚糖-卵透明带口服DNA纳米免疫微囊制备成功,为进一步发展更安全、方便、有效的卵透明带DNA避孕疫苗打下了基础。  相似文献

探讨受精后卵透明带糖蛋白的生化特性变化.用SDS-PAGE分析未受精卵和2-细胞胚胎的透明带成分的差异.研究结果表明,受精前后卵透明带的主要带型相似,但2-细胞胚胎透明带在还原电泳中有一条特异的小肽,分子质量在15000~30000之间,约为22000.进一步用HPLC分析两种样品的O-侧糖链的不同,分析显示,两种样品的O-糖链中有3种单糖基的保留时间值很相似,但有些寡糖基明显不同.小鼠透明带精子受体失活的分子机制主要在于O-寡糖链的修饰,O-单糖与精子受体失活没有明显的关系。  相似文献

利用纯化的可溶性人PD-1胞外域(sPD-1)蛋白为免疫原免疫小鼠,获得抗血清.免疫印迹显示该抗血清与原核表达的sPD-1有特异性反应,但与某些菌体蛋白存在交叉反应.经菌体蛋白吸附法处理后,交叉反应明显减弱.以商业化抗人PD-1抗体和小鼠sPD-1抗血清分析经亲和层析富集的活化的Jurkat T细胞表达的PD-1,只有自制的抗血清鉴定到特异反应条带,其表观分子质量为49 ku.  相似文献

详细介绍了抗鱼(花鲢Anstichthysnobilis)卵透明带单克隆抗体淋巴细胞杂交瘤的制备及鉴定方法,用佛氏佐剂乳化热溶花鲢鱼卵透明带,然后免疫雌性BALB/C小鼠,取其脾细胞与骨髓瘤Sp2/0融合产生杂交瘤,经3次再克隆后,用间接ELISA方法筛选出一株抗鱼卵透明带的单克隆抗体杂交瘤,经免疫双扩散试验测得该种单克隆抗体为IgG1,经过染色体检查知其染色体数目在86~108之间,为93.6±3.81,杂交瘤细胞染色体经检查发现有中间和亚中着丝点染色体,符合杂交细胞染色体特征。  相似文献

目的在昆虫细胞中表达出狂犬病毒核蛋白(NP),并探讨重组NP的免疫原性.方法采用杆状病毒表达系统在Sf9昆虫细胞中表达狂犬病毒核蛋白,用直接免疫荧光(DFA)、SDS-PAGE和Western blot对表达产物进行检测和分析,以感染重组杆状病毒的昆虫细胞裂解液免疫小鼠,检测表达产物的免疫原性.结果重组杆状病毒在昆虫细胞中获得表达,免疫小鼠可产生抗核蛋白抗体.结论在Sf9昆虫细胞中表达出狂犬病毒核蛋白,重组产物具有生物活性.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
