A fluorescence in situ hybridization (FISH) procedure was adopted to physically map two rice BAC clones 24E21 and 4F22 linked to Gm-6 and Pi-5(t) in O. offi-cinalis. FISH results showed that the two BAC clones were located at 4L. The percentage distance from the centromere to the hybridization sites was 72 + 2.62 for 24E21 and 54+ 5.43 for 4F22, the detection rates were 52.70% and 61.2%. The results obtained from the BAC and plasmid clones, RG214 and RZ565 of cultivated rice and O. officinalis were the same. This suggested that the markers, RG214 and RZ565 of cultivated rice and O. officinalis were in the same BAC clones. The homologous sequences of Gm-6 and Pi-5(t) in O. officinalis were positions that signals existed on the 4L. Many signals were observed when no Cot-1 DNA blocked. This also showed that repetitive sequences were some ho-molgous between cultivated rice and O. officinalis. The identification of chromosome 4 of O. officinalis is based on Jena et al. (1994). In our study, we discussed the possibility of physical map in O. officinalis with rice BAC clones.  相似文献

Oryza granulata Nees et Arn. ex Watt. is one of the three wild relatives of rice,which are the most valuable for study and utilization in China.In this study,the homology and physical locations of three rice resistance genes,Glh,Bph-3 and xa-5 are comparatively analyzed between O.sativa and O. granulata by Southern blotting and fluorescence in situ hybridization (FISH).The results of Southern blotting indicate that there exist homologous sequences of the tested RFLP markers in O. granulata.By using three bacterial artificial chromosome (BAC) clones scanned by the tested RFLP as probes, FISH signals are detected on both mitotic and pachytene chromosomes in O.sativa and O.granulata.Dual-color FISH demonstrates that two of the three BAC clones (14E16 and 38J9) are located on the short arm of the same chromosome pair in O. granulata. Additionally, colinearity is shown for the two clones between O.sativa and O.granulata. Another BAC clone 44B4 is located on the end of the short arm of other chromosome pair in these two species.Although the phylogenetic relationship between O.sativa and O.granulata is the most distinct in Oryza and these two species have evidently different biological features and ecological habits, the relative lengths and arm ratios of the detected chromosomes and the relative positions of the tested clone signals on chromosomes in O. granulata are quite similar to those in O. sativa.  相似文献

以Ce(NO3)3·6H2O和(NH4)2CO3为沉淀源,搀杂稀土氧化物Sm2O2,利用柠檬酸作为络合剂和分散剂,制备出纳米(CeO2)0.86(Sm2O3)0.14.并采用TEM、XRD等方法对其热分解所得纳米(CeO2)0.86(Sm2O3)0.14的形貌及结晶性等进行了表征.结果表明:随着柠檬酸加入量的增加,焙烧生成的纳米(CeO2)0.86 (Sm2O3)0.14颗粒的形貌从纳米线向类球形颗粒变化,所得(CeO2)0.86(Sm2O3)0.14为立方萤石结构,且随焙烧温度的提高,晶化程度增大.  相似文献

对临床上确认为M2-ANLL的病例,首先进行骨髓细胞染色体的核型分析,经G显带和R显色确定为t(8;20)(q22;p23)易位,是一种新的变异型,为进行分子生物学研究,分别设计引物,反转录扩增检测ETO基因cDNA 3‘0端顺序和ETO融合转录物检测为阴性,从分子水平上排除了经典的t(8,21)易位的可能性。说明了该病例为t(8;20)易位,ETO基因受累的新变异型白血病。  相似文献

采用XRD,SEM、氧浓差电池、电化学工作站等方法对高温固相反应法合成的(La0.97Yb0.03)2Mo2O9固体电解质材料的电性能进行了研究.结果表明,在600~ 800℃下,制得的样品氧浓差电池电动势的实测值与理论值吻合得很好,氧浓差电池放电性能证实了样品在该温度范围内为一纯氧离子导体.在测定温度范围内,不同气氛下的电导率随温度升高基本呈线性增大,湿润空气中表现出较高的电导率,800℃时达到最大值0.022 S·cm-1.  相似文献

用紫外-可见分光光度计对Na2S与Na2[Fe(CN)5NO]反应得到的紫色混合水溶液的稳定性进行跟踪检测,结果表明此紫色化合物极不稳定.进一步用显微熔点测定仪、红外光谱等仪器对该紫色混合液的分解产物-黄色针状晶体进行了鉴定,最终确定该晶体为黄血盐Na4[Fe(CN)6].3H2O.这一研究对Na2[Fe(CN)5NO]鉴定S2-的实验现象有更进一步的解释.  相似文献

作者采用化学共沉淀法制备ZrO_2(Y_2O_3)超细粉末,在沉淀反应时添加表面活性剂作为分散剂.测试了粉末的物理性能,结果表明:表面活性剂吸附在沉淀物粒子上,有效地阻止了沉淀物粒子的团聚,所制得的粉末性能与乙醇处理工艺制得的粉末性能相近,这大大降低了氧化锆超细粉末的成本.  相似文献

本文基于密度泛函理论的第一性原理,采用广义梯度近似的PBEsol+U方法计算了ThO2﹑PaO2﹑UO2﹑(Th, Pa)O2和(U, Pa)O2的结构、力学、电子和光学性质. 结构优化的结果表明PBEsol+U方法能对锕系氧化物ThO2﹑PaO2和UO2提供较好的晶格参数和力学参量. 计算ThO2﹑PaO2和UO2的晶格参数和带隙与实验值和相关理论值做了比较,且与实验值吻合较好. 同时,(Th, Pa)O2和(U, Pa)O2的晶格参数处于ThO2与UO2之间,同时预测(Th, Pa)O2和(U, Pa)O2的带隙也处于ThO2与UO2的带隙值之间. 电子性质计算表明PaO2和UO2是Mott绝缘体,而ThO2是电荷转移绝缘体,这些是和实验和理论结论是一致的. 而(Th, Pa)O2和(U, Pa)O2在费米能级附近的电子态有显著的自旋极化效应,且导带分别主要是Pa-5f和U-5f电子态占据. 最后,对比分析了这些体系的光学介电函数的实部和虚部以及光学参数.  相似文献

Genetic mapping of microsatellite markers was carried out in a rice DH population derived from across between Zaiyeqing 8 (indica) and Jingxi 17 (japonica). A total of 89 microsatellite markers, including 84 (GA)n, 2(TCT)n, 2(ATT)n, and, l(ATC)n motifs, were integrated relatively evenly into the established genetic map of the DH population. 合成了一个镍-多吡啶配合物[Ni(dmbpy)3](H2O)(ClO4)2(dmbpy=4,4’-二甲基2,2’-联吡啶),并通过室温挥发法培养了该配合物的单晶.单晶X-衍射结果表明,该配合物属三斜晶系,P-1空间群,主要晶体学参数为:a=11.192(13),b=14.444(18),c=14.468(18),α=84.86(2)°,β=76.691(19)°,γ=68.24(2)°,V=2114(4)3,Z=2.配合物分子由1个单核镍阳离子单元[Ni(dmbpy)3]2+、2个高氯酸根阴离子和1个水分子组成,配合物中镍原子的配位环境为六配位的八面体构型,3个dmbpy分子以双齿形式和1个镍原子配位.通过紫外光谱和荧光光谱研究了该配合物与小牛胸腺DNA(CT-DNA)的相互作用,结果表明该配合物与CT-DNA之间存在静电和插入两种作用模式.  相似文献

To test the resistant spectrum of the Xa-min(t) gene introgressed from Oryza minuta, thirty-four isolates of different bacterial blight pathogen, Xanthomonas oryzae pv. oryzae (Xoo), from 11 countries were used to inoculate the Xa-min(t) introgression line 78-15. Four rice cultivars, IR24, C64 (IRBB21), Nipponbare and Zhonghua 11 were used as controls. The results showed that the Xa-min(t) gene was broad-spectrum and highly resistant to diverse Xoo isolates. The methods of bulk segregant analysis (BSA), randomly amplified polymorphic DNA (RAPD) and sequence characterized amplified regions (SCAR) were used to analyze F2 individuals of the hybrid IR24×78-15 and molecular genetic markers linked to Xa-min(t) gene were identified. A total of 800 arbitrary decamer oligonucleotide primers were used for RAPD analysis. Two RAPD markers, BE05300 and BE061400, produced by primers BE05 and BE06 respectively, were closely linked to the Xa-min(t) gene. Based on the sequences of these two markers, sequence specific primers were designed and used to screen all F2 plants. One RAPD marker, BE05300, was converted into a stable SCAR marker (ScBE05300). Linkage analysis was carried out using markers ScBE05300 and BE061400 on 948 and 719 F2 individuals of the hybrid IR24×78-15. Our results indicate that the genetic distances from Xa-min(t) to ScBE05300 and BE061400 are 2.2 cM and 3.7 cM respectively on the same side. This study may facilitate the construction of the fine physical map of the Xa-min(t) gene.  相似文献

本文研究了3,5—二溴—PADAP在酸性条件下与Cr_2O_7~(2-)和SCN~-形成三元离子缔合物的最佳条件。缔合物的表现摩尔吸光系数为1.44×10~5L·mol·cm~(-1),从而提供了测定微量Cr_2O_7~(2-)的新方法,可用于合金钢铬含量及饮用水Cr(vl)含量的测定,结果满意。  相似文献
