Human colon adenocarcinomas from 52 patients were investigated for the presence of the colon polymorphic antigens WZ. The patients were typed for their WZ phenotype, using the immunofluorescence method on non tumoral colon mucosa sections: 27 patients were found W+ Z+, 18 W- Z+, and 7 W- Z-. The tumors were tested for the presence of the WZ phenotypes, using the immunofluorescence method and a radio-immunoassay. The WZ phenotypes were not expressed in the non secreting tumors, whatever the patient's phenotype. They were expressed in the secreting tumors and had the same phenotype as found in the corresponding normal mucosa. The WZ phenotypes were present in human developed into "nude" Mice inoculated either with differentiated colon carcinomas, or with a human colon carcinoma cell line (HT-29).  相似文献

An antiserum raised in Rabbits against brain glycoprotein precipitated an identical antigen in faetal dermis and intestine extracts, and also in non nervous tumors (breast adenocarcinomas and adenofibromas, ovarian cystadenoma, gastric and colonic adenocarcinomas, hepatomas, malignant melanomas, rhabdomvosarcoma, fibrosarcomas). By the indirect immunofluorescence technique, this antigen was always found associated with the mesenchymal proliferation, either malignant (fibrosarcoma) or reactive (fibrous stroma reaction of carcinomas).  相似文献

The morphogenesis and cytodifferentiation of human colon cancer cells (LS174T and HT29) were examined by combining cancer cells with fetal rat digestive-tract mesenchyme in organ culture. LS174T cells migrated into the mesenchyme to form glandular structures composed of single columnar cells with their nuclei oriented basally, while HT29 cells formed cell masses with little lumen formation. Immunohistochemical studies with antibodies against carcinoembryonic antigen and secretory components showed that the composition of cell surface glycoproteins was not necessarily reversed to the normal type, even when neoplastic cells exhibited normal glandular structures.  相似文献

The insoluble pellet of human mammary carcinomas was solubilized by an acid buffer. Antiserum prepared with this acidosoluble fraction, after suitable absorption gave one precipitin line with the immunizing extracts: this line is different from those given by the tumor associated antigens actually known. The same antiserum reacted only with sections of human mammary carcinomas by immunofluorescence . It did not stain sections of normal mammary glands or benign mammary diseases. Reactivity with cancers of other organs was absent or doubtful. Hence it is likely that an antigen associated to human mammary carcinomas was characterized.  相似文献

3,5,3'-triiodothyronine (T3) and 3,3',5'-triiodothyronine (rT3) were measured by radioimmunoassay in saline extracts of neonates and human adult thyroid tissues and of fetuses, Piglets and adult Swine thyroid tissues. In all these extracts, T3 content was higher than rT3 content whatever the period of development. Both triiodoamino acids represent a small percentage of the iodinated protein in thyroid tissues.  相似文献

The continuous cell lines T 24 and HT-29, derived from human bladder and colon carcinomas, produce term-placental and intestinal alkaline phosphatase, respectively. Growth in hyperosmolar medium or exposure to prednisolone or sodium butyrate induces increased enzyme levels, and combinations of inducers elicit synergistic activity increases. The effect of the inducing agents is strikingly diminished when cells are grown in the presence of high concentrations of human serum, and the synergistic increases are essentially abolished. Major human serum protein fractions do not affect alkaline phosphatase induction.  相似文献

The kinesin-related protein HsEg5 plays essential roles in mitotic spindle dynamics. Although inhibition of HsEg5 has been suggested as an aid in cancer treatment, the effects of such inhibition on human cells have not been characterized. Here we studied the effects of monastrol, an allosteric HsEg5 inhibitor, on AGS and HT29 cell lines and compared them to those of taxol. While both cell lines were similarly sensitive to taxol, AGS cells were more sensitive to monastrol. The differences in sensitivity were determined by the degree of inhibitory effect on cell proliferation, reversibility of monastrol-induced G2/M arrest, intracellular phenotypes and induction of apoptosis. In both cell lines, monastrol-induced apoptosis was accompanied by mitochondrial membrane depolarization and poly-ADP-ribose polymerase 1 cleavage. In AGS, but not HT29 cells, monastrol-induced apoptosis involved a prominent cleavage of procaspases 8 and 3. While in AGS cells, monastrol induced the formation of symmetric microtubule asters only, in HT29 cells, asymmetric asters were also formed, which may be related to specific HsEg5 functions in HT29 cells.Received 18 February 2004; received after revision 30 May 2004; accepted 16 June 2004  相似文献

In excised inside-out membrane patches of the human colon carcinoma HT-29cl.19A cells a large conductance (373±10 pS) chloride channel was found. Channel activity could only be observed after excision of patches from cells incubated with calcium ionophore. The channel was never observed in cell-attached patches. The channel was strongly voltage dependent, being open only between +30 and –30 mV clamp potentials. The selectivity sequence among anions, deduced from reversal potentials, was I>Br>Cl>F>gluconate. The PNa/PCl was 0.09. Although a similar type of channel, has been described earlier, this is the first report stating its appearance in patches of intestinal epithelial cells requiring the combined action of Ca2+ ionophore and excision, suggesting its control by an intracellular compound.  相似文献

Summary The intestine of the ferret (Putorius furo) is unusual in that there is no external anatomical division between ileum and colon. Up to 8–10 cm from the anus the electrical activity was organized into migrating myoelectric complexes typical of the small intestine. At this point the pattern of electrical activity changed abruptly to that characteristic of the colon, namely short and long spike burst activity. Histological examination showed that at this point the muscular layers were interupted by a band of connective tissue sufficient to permit the functional autonomy of the last part of the intestine.Acknowledgments. Small doses, repeated for months of anti-esterase-lipoprotein antibodies, from several antisera raised against a crude lipoprotein-lipase from Rabbit adipose tissue (2), induce exocrine pancreatic carcinomas in mice, without any specific barrier. This seems to be the first case of a quite elective carcinogenic process of the pancreas, by an immunological mechanism, with a very high incidence, up to 100%. Such carcinomas are highly metastatic, produce a pancreatic carcinomatous ascites, and are deadly. This immunological pathogeny is discussed., more particulary on an action of immuno-complexes in an abnormal physiological environment.  相似文献

目的探索人结直肠癌细胞系形成肿瘤细胞球的培养方法及球体内间质化标志物的表达检测。方法结直肠癌细胞系SW480、SW620、LOVO、HT29及HCT116在添加了不同细胞生长因子的无血清培养基(SFM)中悬浮培养,传代更新,诱导分化。免疫荧光技术和RT-PCR分别检测细胞球中间质化标志物N—cadherin及Vimentin的表迭情况。结果5种细胞系均能在特制的SFM中形成细胞球,并稳定的传代增殖。在用血清诱导分化后,又重新贴壁生长,且与亲代细胞保持一致的细胞形态。细胞球中间质化标志物N—cadherin、Vimentin表达均上调。结论结直肠癌细胞系在低黏附及添加了细胞生长因子R—Spondin 1、Noggin的无血清环境中更容易形成悬浮生长的肿瘤细胞球,细胞球细胞比亲代细胞更具有转移侵袭能力。  相似文献

Summary The presence of a hyperglycemic factor in the optic tentacles of the snailC. ligulata is reported here. A preliminary characterization based on crude extracts indicates the factor to be water-soluble, heat labile and to be an albumin. The ablation of optic tentacles and injection of optic tentacle extract into operated and normal snails caused a rise in blood sugar, total carbohydrate and glycogen in the foot muscle and mantle and a decrease in hepatopancreatic glycogen. The ablation also caused a fall in blood free amino acids and a rise in the tissues, which was reversed in the blood and foot muscle by injection of the extract. Possible conversion of amino acids to total carbohydrates and glycogen by gluconeogenesis is suggested.  相似文献

Summary High-titer antibodies against bradykinin were raised in rabbits. 2 different conjugates of bradykinin were used for immunization: bradykinin coupled to human serum albumin via 1,5-difluoro-2,4-dinitrobenzene and bradykinin coupled to edestin via 1-ethyl-3(3-dimethyl-aminopropyl)-carbodiimide. The sensitivity of the radioimmunoassay method is in the range of 1–50 pg of bradykinin. Cross-reaction of anti-bradykinin antisera occurred with kallidin and met-lysbradykinin.  相似文献
