从安徽省合肥市西郊的大蜀山上采集了40余株木腐真菌,将其在含有α-萘酚的麸皮汁平板上分离纯化后,根据菌落周围产生紫色氧化圈的显著程度筛选出4株漆酶活性较高的菌株,将这4株真菌进行摇瓶培养,测定并比较它们所产漆酶的活性,得到一株产酶活力最高的菌株SY04.菌株SY04在液体培养3d后粗酶液漆酶活力为330U/mL,在对其漆酶粗酶液进行活性分析后发现,该菌所产漆酶在60℃下保温30min后,活性不受明显影响,其最适反应温度为60℃.该菌所产漆酶具有较高的最适反应温度和热稳定性,具有一定的研究和应用价值.  相似文献

以筛选与鉴定尉犁县黑湖产淀粉酶细菌,并对筛选出的细菌进行产酶条件优化为研究目的,从43株细菌中筛选出淀粉酶高产菌株HM-22,并对其进行了菌株形态学鉴定、16S r DNA分子鉴定以及产酶条件优化。从尉犁县黑湖采样的30个水、土和泥样样品中分离筛选43株细菌,从中筛选出14株产淀粉酶的菌株,采用Yoo改良法对产透明圈较大的菌株进行酶活力测定,从中筛选出了一株产酶活性较高的菌株HM-22。经革兰氏染色、菌落形态观察、生理生化检测和16S r DNA序列比对鉴定该菌与Bacillus tequilensis的相似性为99.8%,属于芽孢杆菌属。对菌株HM-22产酶条件进行初步优化确定其产酶的最佳条件是:该菌产淀粉酶最适温度为40℃,最适碳源为可溶性淀粉,最适氮源为牛肉膏,最适反应pH为6.0,最适产酶培养时间为24 h。优化后菌株HM-22的酶活力从最初的的酶活力122.45 U/m L达到147.53 U/m L,酶活力提高了17%。该研究为从新疆盐湖细菌中筛选淀粉酶活力较高的菌种资源及应用提供了理论依据和参考。  相似文献

在实验室条件下通过富集培养共分离出121株菌进行筛选。利用需氧性、BOD2/COD0、胞外酶等指标进行筛选,最后筛选到7株对富营养有机物具有较高降解能力的细菌,它们均能产生酪蛋白酶、明胶酶、脂酶(Tween-80)、淀粉酶其中4株细菌还能产生卵磷脂酶。通过测定BOD5及COD来衡量7株细菌利用对虾饵料的能力,5d内对虾饵料培养基的CODMn去除率达到59.6%-79.2%。用常规生理生化方法将细菌鉴定到属,其中1株为弧菌属细菌(Vibrio spp.),6株为芽孢杆菌属细菌(Bacillus spp.)。  相似文献

通过唯一碳源法从来源于长白山森林的土壤中筛选到多株具有纤维素降解活力的细菌,选取其中的1株芽孢杆菌DS1309进行了鉴定及发酵条件的初步优化,结果显示该菌株为地衣芽孢杆菌(Bacillus licheniformis)。发酵产酶的最适条件为培养温度30℃,培养基初始p H6.0,最适碳源为葡萄糖,最适碳源为酵母粉。菌株在最适条件下培养48h达到产酶高峰,酶活力可达到1.82 IU/m L。  相似文献

为了筛选含ACC脱氨酶的根际促生细菌,以1-氨基环丙烷-1-羧酸(ACC)为唯一氮源,从绞股蓝根际土壤中富集和分离ACC脱氨酶阳性细菌,利用α-丁酮酸测定法和CAS平板法分别检测其ACC脱氨酶活性和产嗜铁素能力,利用菌液培养接种法检测细菌的促生作用,最后通过生理生化试验和16SrRNA基因序列分析对所筛选的菌株进行鉴定.经富集筛选法从绞股蓝根际土壤中分离出9株ACC脱氨酶阳性细菌,它们均具有不同程度的ACC脱氨酶活性,其中3株细菌能产生嗜铁素.促生试验结果表明,7株ACC脱氨酶活性细菌对水稻幼苗根系的生长发育表现出不同程度的促生作用,其中菌株JDG127的促生作用最明显,与对照组相比,水稻幼苗的根长和根鲜重增长了约1.6倍.鉴定结果表明,9株细菌中1株属于单胞菌属(Stenotrophomonas),1株属于类芽胞杆菌属(Paenibacillus),1株属于芽孢杆菌属(Bacillus),2株属于肠杆菌属(Enterobacter),4株属于不动杆菌属(Acinetobacter),与ACC脱氨酶相比,铁载体对根际细菌的促生作用更明显.  相似文献

作者从泸州油脂厂附近的土壤中,筛选到一株产碱性脂肪酶活性较高的细菌,经初步鉴定为假单胞菌(Pseudomonassp.).经对该菌株进行产酶条件和酶学性质研究发现,该菌株的最适产酶发酵培养基为(%):酵母浸出汁1.5,黄豆粉1,NaCl0.2,K2HPO40.2,NaH2PO40.2,MgSO4·7H2O0.01,初始pH8.5.100mL三角瓶装液10mL,30℃,150r/min摇床培养48h,酶活最高可达19.1U/mL.酶的最适反应温度40℃,最适pH8.5,该酶具有较好的热稳定性.  相似文献

采用平板透明圈法从 19株腐皮镰孢菌 (Fusariumsolani)中筛选到 1株产壳聚糖酶的菌株 ( 0 114 ) .摇瓶培养表明 ,该菌壳聚糖酶的产生不需要壳聚糖诱导 ;该菌培养 84h达到产酶高峰 ,酶的最适作用温度为 5 0℃ .4 5mU mL作用酶量 ,5 0℃下处理 6h ,壳聚糖的降解率达 2 6 .7% ,薄层层析分析表明酶解产物中壳寡糖含量较高  相似文献

为了探寻功能微生物,用几种不同培养基分离蜗牛肠道可培养细菌并筛选纤维素降解细菌,研究了纤维素酶学性质.结果表明,LB培养基培养蜗牛肠道细菌时菌落种类和数量最多,达3.3×104CFU/m L.刚果红染色显示,7种优势细菌全部具有产酶能力;菌S6粗酶液的内切葡聚糖酶(Cx)和滤纸酶(FPase)活性最高,分别为5.96和2.84 U/m L.对Cx酶学性质初步研究表明:该酶最适反应温度为35℃,在55~75℃时有较高的热稳定性;最适p H为6.0,在p H 3.0~8.0均有酶活力,p H 8.0相对酶活仍为60%;Mg~(2+)、Na~+、K~+和Ca~(2+)对酶反应具有促进作用,Zn~(2+)、Cu~(2+)和Mn~(2+)为酶反应的抑制剂.研究蜗牛肠道细菌的组成,从中分离高产纤维素酶菌株,为纤维素生物降解转化成可再生能源寻找新的思路.  相似文献

采用平板涂布法和划线分离法对青海湖水样进行了初步的分离纯化,筛选出可培养低温微生物14株,其中细菌9株,真菌3株,放线菌2株;经过初步的分类有8个耐冷菌和6个嗜冷菌;而且其中有不少产低温淀粉酶、脂酶、蛋白酶和氨基酸脱羧酶的有用微生物。  相似文献

通过富集培养、水解圈鉴定、酶活检测等步骤,从不同地点采集含木聚糖的土样中初筛出10株木聚糖酶产生菌,获得一株酶活最高的菌株Bacillus sp. X-18.对Bacillus sp. X-18木聚糖酶的部分酶学特性进行了研究,结果显示:菌株X-18酶活最适温度为50 ℃,在50 ℃保温1 h仍保持100%的酶活力.该酶pH值范围较广,在pH4.0-8.0范围内均能保持较高的活性,最适为pH5.0,与其相关报道的研究相比,该木聚糖酶具有良好的酸碱耐受性.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献
