The injection of ACTH 1--24 into the cerebral ventricles in rats markedly reduces the reaction time in the hot-plate test and the nociception threshold in the tail-stimulation test. Morphine antagonizes and naloxone potentiates this hyperalgesic effect of ACTH. It is proposed that ACTH peptides play a physiological role in nociception.  相似文献

Summary In the course of studying the plasma adrenocorticotropic hormone (ACTH) and corticosterone responses to synthetic corticotropin releasing factor (CRF), we noted some disparity in the responses. A higher dose (20 g compared with 5 g per rat i.a.) produced an equal plasma ACTH but greater plasma corticosterone response in adult male rats. Thus, we examined the possibility that CRF increases adrenocortical responsiveness to ACTH. CRF significantly (p<0.0005) increased the plasma corticosterone response to ACTH in rats pretreated with dexamethasone. Thus, synthetic CRF increases corticosterone secretion in rats not only by stimulating ACTH secretion, but also by increasing the adrenocortical responsiveness to ACTH.Acknowledgments. The male gametogenic development of the immature rat shows inhibitory effect by the exogenous infusion of ACTH (0.25 IU and 0.50 IU; i.p.). Seminiferous tubular degeneration, Leydig cell atrophy and Sertoli cell regression are very conspicuous in the dosage of 0.50 IU ACTH (i.p.). Cytometrical and histological evidence confirms that ACTH could be inhibited by FSH during male gametogenic development which leads to cellular degeneration of testes in the immature rat.  相似文献

Zusammenfassung Der Mechanismus der Verhinderung des ACTH-Effektes durch Protaminsulfat wurde untersucht. Papierchromatographisch konnte festgestellt werden, dass beide Substanzen einen Komplex bilden und infolgedessen ACTH seine Aktivität verliert. Während die proteinartigen ACTH-Präparate starke komplexibldende Eigenschaft besitzen, vermögen die peptidartigen Präparate nur in geringem Masse Komplexe zu bilden. Dies konnte auch durchin-vivo-Verhinderungsversuche (Sayers test) mit beiden Präparaten bestätigt werden.  相似文献

Summary We investigated whether hypothalamic prostaglandin E2 (PGE2) and corticotropin releasing factor (CRF) are responsible for the development of the adrenocorticotropic hormone (ACTH) response induced by interleukin-1 (IL-1). The present results show that ACTH responses induced by intravenous injection of IL-1 were suppressed by systemic pretreatment with indomethacin and that intrahypothalamic injection of PGE2 stimulates the secretion of ACTH. Furthermore, systemic pretreatment with anti-CRF antibody significantly suppressed the ACTH response induced by intrahypothalamic injection of PGE2. These data suggest that the ACTH response induced by IL-1 is mediated by CRF secretion stimulated by hypothalamic PGE2.  相似文献

Summary Impairment of 21-hydroxylation was observed when 4-C14-cholesterol was transformed into 4-C14-corticosteroids in slices of bovine adrenal cortex, whereas corticosteroid synthesis from 4-C14-progesterone proceeded freely. ACTH stimulated corticosteroid formation from 4-C14-cholesterol but not from 4-C14-progesterone. It is therefore suggested that ACTH facilitates the «release» of 21-desoxypregnenes, which are present in a «bound» form after synthesis from cholesterol in intact adrenocortical tissue, and which are unavailable for the following reactions without conditioning by ACTH.  相似文献

The effects of chronic ACTH treatment on the increase in the intracellular concentration of cyclic-AMP and cyclic-GMP acutely elicited by ACTH in the rat adrenal cortex were investigated. The immunocytochemical and histochemical characters of the corticotroph cells of the Turtle adenohypophysis have been studied. These cells are localised in the rostral part of the gland and are revealed by Is anti ACTH (1-24) and (17-39). They are also colored with lead hematoxyline and PAS-Orange G. The corticotroph nature of these cells is confirmed by the study of their modifications after treatment with amphenone and ACTH. The Is anti ACTH also reveal most of the cells of the pars intermedia; while the Is anti beta-MSH reveals only these cells and some scatter cells of the pars distalis.  相似文献

Resumen En animales tratados crónicamente con ACTH se aumentan o activan los sistemas de la adrenal que catabolizan la corticosterona y los que hidroxilan en C21, a la vez que disminuye la cantidad de radioactividad incorporada a la 18 OH DOC. Esto indica que el ACTH in vivo tiene sobre la adrenal incubada una acción distinta, a veces opuesta, a la del ACTH exógeno agregado in vitro al principio de la incubación.

Daily administration of synthetic ACTH (betaI-23 corticotrophine) from day 15 of pregnancy induces delay or inhibition of parturition in normal or ovariectomized Rats injected or not with estradiol. Parturition takes place at approximatively normal time, when ACTH is stopped and estradiol continued. Radio-immunological assays show an increase of blood progesterone level after ACTH administration in castrated, but not in castrated and adrenalectomized Rats. Summary The acute in vitro effects of ACTH and HGH in human adrenals were examined. HGH acts synergistically with ACTH in augmenting adrenal aldosterone output. This effect is not due to cAMP alterations in response to HGH. HGH slightly depresses the human adrenal cortisol response to ACTH.  相似文献

In the green Frog (Rana esculenta) the plasma contains a polypeptide immunologically related to human and porcine corticotropins. A radiommunoassay capable of detecting 4.10-12 g hog ACTH has been used for a direct plasma ACTH assay in the Frog. Using this method, the ACTH rate was determined both in untreated frogs and in animals under various experimental conditions.  相似文献

Adrenocorticotropin hormone (ACTH) receptor-like messenger RNA was localized in molluscan hemocytes and human peripheral blood mononuclear cells by in situ hybridization using a digoxigenin-labelled bovine complementary DNA probe. These findings suggest that the ACTH receptor gene has been highly conserved during evolution. Moreover, these data represent further support for a relationship between the immune and neuroendocrine systems in invertebrates, as documented in our previous studies [1].
