Synthesis of SP1-glycoprotein by the human placenta was directly demonstrated, by in vitro translation of RNA extracted from full term and from early placentas in a cell-free wheat germ system followed by specific immunoprecipitation of the radioactively labeled nascent peptides. De novo synthesized SP1-glycoprotein in both RNA preparations accounted for 1-1.3% of total protein synthesis.  相似文献

Summary The emission maximum of the fluorescence spectrum of the mitochondrial F1-ATPase in shifted from 305 to 334 nm when the excitation wavelength is altered from 270 to 300 nm. This indicates that both tyrosine and tryptophan contribute to the intrinsic fluorescence of the F1-ATPase.Acknowledgments. The authors wish to thank F. Résumé Une proportion non négligeable de disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) et de disodium dichloromethylenediphosphonate (Cl2MDP) peut être absorbée au niveau de l'ileum de poulet. L'absorption du Cl2MDP est significativement supérieure à celle de l'EHDP.

Summary PGE1 but not PGF2 at 500–1000 g/kg induced a slow and sparse flow from the parotid and no flow at all from submaxillary glands. Composition of PGE1-induced parotid saliva was quite different from that evoked by any autonomic agonists. Mammalian protease-activated-receptor-1 and -2 (PAR1 and PAR2) are activated by proteases found in the flexible microenvironment of a tumor and play a central role in breast cancer. We propose in the present study that PAR1 and PAR2 act together as a functional unit during malignant and physiological invasion processes. This notion is supported by assessing pro-tumor functions in the presence of short hairpin; shRNA knocked-down hPar2 or by the use of a truncated PAR2 devoid of the entire cytoplasmic tail. Silencing of hPar2 by shRNA-attenuated thrombin induced PAR1 signaling as recapitulated by inhibiting the assembly of Etk/Bmx or Akt onto PAR1-C-tail, by thrombin-instigated colony formation and invasion. Strikingly, shRNA-hPar2 also inhibited the TFLLRN selective PAR1 pro-tumor functions. In addition, while evaluating the physiological invasion process of placenta extravillous trophoblast (EVT) organ culture, we observed inhibition of both thrombin or the selective PAR1 ligand; TFLLRNPNDK induced EVT invasion by shRNA-hPar2 but not by scrambled shRNA-hPar2. In parallel, when a truncated PAR2 was utilized in a xenograft mouse model, it inhibited PAR1–PAR2-driven tumor growth in vivo. Similarly, it also attenuated the interaction of Etk/Bmx with the PAR1-C-tail in vitro and decreased markedly selective PAR1-induced Matrigel invasion. Confocal images demonstrated co-localization of PAR1 and PAR2 in HEK293T cells over-expressing YFP-hPar2 and HA-hPar1. Co-immuno-precipitation analyses revealed PAR1-PAR2 complex formation but no PAR1-CXCR4 complex was formed. Taken together, our observations show that PAR1 and PAR2 act as a functional unit in tumor development and placenta-uterus interactions. This conclusion may have significant consequences on future breast cancer therapeutic modalities and improved late pregnancy outcome.  相似文献

Summary Four new hexapeptide analogues of C-terminal Substance P fragment with increased solubility in aqueous solutions are described. The peptides contain histidine in positions 6, 8, 9 and 10, respectively. The effect of the structural changes on the hypotensive activity and antigenic properties of analogues was compared. It was found that substitution of amino acid residues in various positions in the C-terminal hexapeptide of Substance P resulted in different effects on the hypotensive and antigenic properties, respectively. Only the [His6] SP6-11 analogue had an unchanged antigenic structure when compared with the C-terminal region of Substance P, but it showed an almost total loss of hypotensive activity. The [His9] SP6-11 analogue retained 50% of the hypotensive activity of the C-terminal hexapeptide but showed a markedly reduced expression of the antigenic epitope localized in this region of Substance P.  相似文献

Summary Nicotinamide adenine dinucleotide phosphate phosphomonoesterase was isolated and partially purified from wheat (Triticum aestivum L. var. Selkirk) leaves. The enzyme hadK NADP value of 1.4×10–4 M and a pH optimum of 5.9.In vitro activity of this enzyme was unaffected by precursors of NAD (nicotinamide and nicotinic acid) or cytokinis (kinetin and benzimidazole). However, when detached wheat leaves were treated with solutions of these compounds, the precursors lowered the specific activity while the cytokinins enhanced the activity. It is suggested that spatial separation and compartmentation of the enzyme and its substrate NADP account for the similar effect of benzimidazole on both.This work was supported by a grant No. A2698 from the National Research Council, Canada.  相似文献

Summary Antibody against aflatoxin M1 was obtained after immunization of rabbits with bovine serum albumin-afla M1 oxime conjugate. The antibody has greatest binding efficiency for afla M1, and was less efficient for afla B1. Cross-reaction of antibody with aflatoxin Q1, aflatoxicol, and aflatoxin B2a was weak. Aflatoxin B2, G1, and G2 and afla B1-guanine adduct showed almost no cross-reaction with the antibody. The sensitivity of the binding assay for aflatoxin M1 detection is in the range of 1–10 ng per assay. Summary The induction of the anti-DNP IgE in rat was suppressed by pretreatment of rats with the tolerogen synthesized by coupling DNP to rat IgG, i.e.; DNP7–10-IgG. It was found that DNP10-IgG1 was an effective tolerogen, whereas other DNP conjugates, i.e. DNP9-IgM, DNP9-IgA, DNP10-IgE, DNP10-IgG2c and DNP10-IgG2a were ineffective.This work was partly supported by a research grant from the Medical Council of Canada to W.Y.L.  相似文献

Summary L1210 leukemic cells grown in vitro were subjected to kinetic analysis using a flow microfluorometer. Zusammenfassung Meerschweinchen wurden für 5 Tage mit Pentagastrin injiziert und die Kerngrössen in den Pankreas-Inseln mit Karyometrie bestimmt. Die Kerne der A1-Zellen nahmen in den behandelten Tieren am Anfang ab und schienen kleiner als in den Kontrollen. In den A2-und B-Zellen wurden keine Veränderungen nach Pentagastrin notiert. Die Resultate stützen die Auffassung, dass Gastrin in den A1-Zellen produziert wird.

Zusammenfassung Es wird gezeigt, dass bei männlichen Wistar-Ratten im 3-Wochen-Diätexperiment mit Zusatz von 0,3% 2-Methyl-2-(p-1,2,3,4-Tetrahydro-1-naphthylphenoxy)-Propionsäure (Su-13437) die Oxydation von 26-14C-Cholesterin zu14CO2 durch Lebermitochondrien (bezogen auf mg/N) ähnlich wie bei den Kontrollen war. Bei den Versuchstieren war jedoch bei Abwesenheit von Cytosol im System die Oxydation durch Lebermitochondrien erhöht.

The effects of plant lectins on the three cellular defense reactions of hemocytes of the solitary ascidian,Halocynthia roretzi (hemocyte aggregation, phagocytosis, and an allogenic reaction), were investigated. Concanavalin A inhibited aggregation, while wheat germ agglutinin and ricin inhibited the allogenic reaction. Neither of the lectins showed inhibitory effects on phagocytosis, but ricin promoted phagocytosis. These effects of the lectins were diminished by the addition of sugars specific for the respective lectins. These results strongly suggest that different surface carbohydrates are involved in the recognition mechanisms of threeH. roretzi cellular defense reactions.  相似文献
