In vitro translation of human placental pregnancy-specific beta 1-glycoprotein (SP1)

Synthesis of SP1-glycoprotein by the human placenta was directly demonstrated, by in vitro translation of RNA extracted from full term and from early placentas in a cell-free wheat germ system followed by specific immunoprecipitation of the radioactively labeled nascent peptides. De novo synthesized SP1-glycoprotein in both RNA preparations accounted for 1-1.3% of total protein synthesis.     

Intrinsic fluorescence of mitochondrial F1-ATPase

Summary The emission maximum of the fluorescence spectrum of the mitochondrial F₁-ATPase in shifted from 305 to 334 nm when the excitation wavelength is altered from 270 to 300 nm. This indicates that both tyrosine and tryptophan contribute to the intrinsic fluorescence of the F₁-ATPase.Acknowledgments. The authors wish to thank F. Binding of aflatoxins B1 and G1 to human serum proteins

Ileal absorption of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) and disodium dichloromethylene diphosphonate (Cl2 MDP) in the chick

Prostaglandin E1 induced salivary secretion

Summary PGE₁ but not PGF₂ at 500–1000 g/kg induced a slow and sparse flow from the parotid and no flow at all from submaxillary glands. Composition of PGE₁-induced parotid saliva was quite different from that evoked by any autonomic agonists. The present study suggests that PGE₁ might act directly on parotid acinar cells.Acknowledgment. This work was supported by NIDR grant DE05633. Protease-activated-receptor-2 affects protease-activated-receptor-1-driven breast cancer

Mammalian protease-activated-receptor-1 and -2 (PAR₁ and PAR₂) are activated by proteases found in the flexible microenvironment of a tumor and play a central role in breast cancer. We propose in the present study that PAR₁ and PAR₂ act together as a functional unit during malignant and physiological invasion processes. This notion is supported by assessing pro-tumor functions in the presence of short hairpin; shRNA knocked-down hPar2 or by the use of a truncated PAR₂ devoid of the entire cytoplasmic tail. Silencing of hPar2 by shRNA-attenuated thrombin induced PAR₁ signaling as recapitulated by inhibiting the assembly of Etk/Bmx or Akt onto PAR₁-C-tail, by thrombin-instigated colony formation and invasion. Strikingly, shRNA-hPar2 also inhibited the TFLLRN selective PAR₁ pro-tumor functions. In addition, while evaluating the physiological invasion process of placenta extravillous trophoblast (EVT) organ culture, we observed inhibition of both thrombin or the selective PAR₁ ligand; TFLLRNPNDK induced EVT invasion by shRNA-hPar2 but not by scrambled shRNA-hPar2. In parallel, when a truncated PAR₂ was utilized in a xenograft mouse model, it inhibited PAR₁–PAR₂-driven tumor growth in vivo. Similarly, it also attenuated the interaction of Etk/Bmx with the PAR₁-C-tail in vitro and decreased markedly selective PAR₁-induced Matrigel invasion. Confocal images demonstrated co-localization of PAR₁ and PAR₂ in HEK293T cells over-expressing YFP-hPar2 and HA-hPar1. Co-immuno-precipitation analyses revealed PAR₁-PAR₂ complex formation but no PAR₁-CXCR4 complex was formed. Taken together, our observations show that PAR₁ and PAR₂ act as a functional unit in tumor development and placenta-uterus interactions. This conclusion may have significant consequences on future breast cancer therapeutic modalities and improved late pregnancy outcome.     

Hypotensive activity of histidine-containing analogues of C-terminal hexapeptide of Substance P

Summary Four new hexapeptide analogues of C-terminal Substance P fragment with increased solubility in aqueous solutions are described. The peptides contain histidine in positions 6, 8, 9 and 10, respectively. The effect of the structural changes on the hypotensive activity and antigenic properties of analogues was compared. It was found that substitution of amino acid residues in various positions in the C-terminal hexapeptide of Substance P resulted in different effects on the hypotensive and antigenic properties, respectively. Only the [His⁶] SP_6-11 analogue had an unchanged antigenic structure when compared with the C-terminal region of Substance P, but it showed an almost total loss of hypotensive activity. The [His⁹] SP_6-11 analogue retained 50% of the hypotensive activity of the C-terminal hexapeptide but showed a markedly reduced expression of the antigenic epitope localized in this region of Substance P.     

RNA biosynthesis in isolated prothoracic glands ofTenebrio molitor in vitro

Summary The prothoracic Glands ofTenebrio molitor synthesize in vitro mainly rRNA. The rate of RNA synthesis reaches a maximum at day 10 of the last larval instar, which coincides with the event of apolysis.The work was supported by the Deutsche Forschungsgemeinschaft. Effect of benzimidazole on nicotinamide adenine dinucleotide phosphate phosphomonosterase activity in wheat leaves

Summary Nicotinamide adenine dinucleotide phosphate phosphomonoesterase was isolated and partially purified from wheat (Triticum aestivum L. var. Selkirk) leaves. The enzyme hadK _NADP value of 1.4×10^–4 M and a pH optimum of 5.9.In vitro activity of this enzyme was unaffected by precursors of NAD (nicotinamide and nicotinic acid) or cytokinis (kinetin and benzimidazole). However, when detached wheat leaves were treated with solutions of these compounds, the precursors lowered the specific activity while the cytokinins enhanced the activity. It is suggested that spatial separation and compartmentation of the enzyme and its substrate NADP account for the similar effect of benzimidazole on both.This work was supported by a grant No. A2698 from the National Research Council, Canada.     

The structures of minor congeners of detoxin complex,the selective antagonist of blasticidin S1

Summary The structures of the minor congeners of detoxin complex, viz., detoxins E₁, C₁, C₂, C₃, B₁, B₃ and A₁ have been established on the basis of spectral and degradative evidence.Acknowledgment. Production and characterization of antibody against aflatoxin M1

Summary Antibody against aflatoxin M₁ was obtained after immunization of rabbits with bovine serum albumin-afla M₁ oxime conjugate. The antibody has greatest binding efficiency for afla M₁, and was less efficient for afla B₁. Cross-reaction of antibody with aflatoxin Q₁, aflatoxicol, and aflatoxin B_2a was weak. Aflatoxin B₂, G₁, and G₂ and afla B₁-guanine adduct showed almost no cross-reaction with the antibody. The sensitivity of the binding assay for aflatoxin M₁ detection is in the range of 1–10 ng per assay. Specific immunosuppression of IgE response to hapten DNP by DNP linked to monoclonal IgG1 in rats

Summary The induction of the anti-DNP IgE in rat was suppressed by pretreatment of rats with the tolerogen synthesized by coupling DNP to rat IgG, i.e.; DNP_7–10-IgG. It was found that DNP₁₀-IgG₁ was an effective tolerogen, whereas other DNP conjugates, i.e. DNP₉-IgM, DNP₉-IgA, DNP₁₀-IgE, DNP₁₀-IgG_2c and DNP₁₀-IgG_2a were ineffective.This work was partly supported by a research grant from the Medical Council of Canada to W.Y.L.     

Resolution of small G1 and G2 populations of L1210 leukemia by flow microfluorometric analysis aided by velocity sedimentation

Summary L1210 leukemic cells grown in vitro were subjected to kinetic analysis using a flow microfluorometer. A single broad peak was found for the DNA content distribution if unfractionated cells were used; prior fractionation using lg velocity sedimentation allowed the separation of small peaks with smaller (G₁) and larger (G₂) DNA contents from the dominant S phase peak with intermediate DNA content.This work was supported by grant number 5P01CA13053 awarded by the National Cancer Institute, DHEW USA, and by grant num ber RBI 76-1 from the Queen Wilhelmina Fund, The Netherlands.     

XY1Y2, a new sex-chromosome system among caraboid beetles

Summary Calathus ascendens (Caraboidea, Pterostichiae, Sphodrini) has 2n=20, XY₁Y₂: XX. This multiple sexchromosome system seems to be derived from a neo-XY one through an X-autosome fusion.Thanks go to A. Machado for collecting some specimens from Last Lagunets. The effect of pentagastrin on the A1-cells of the pancreatic islets in guinea-pigs

Zusammenfassung Meerschweinchen wurden für 5 Tage mit Pentagastrin injiziert und die Kerngrössen in den Pankreas-Inseln mit Karyometrie bestimmt. Die Kerne der A₁-Zellen nahmen in den behandelten Tieren am Anfang ab und schienen kleiner als in den Kontrollen. In den A₂-und B-Zellen wurden keine Veränderungen nach Pentagastrin notiert. Die Resultate stützen die Auffassung, dass Gastrin in den A₁-Zellen produziert wird.

Reduction of aflatoxin B1 with zinc borohydride: An efficient preparation of aflatoxicol

Summary A simple and mild reduction of aflatoxin B₁, involving treatment of aflatoxin B₁ with ethereal zinc borohydride to give 57–65% yield of diastereomeric aflatoxicols, is described.This work was supported by the College of Agricultural and Life Sciences, the University of Wisconsin, Madison, and by Public Health Service Research, grant No. CA 15064, from the National Cancer Institute, and by funds from contributions of food industries to the Food Research Institute. Influence of 2-methyl-2-(p-1,2,3,4-tetrahydro-1-naphthylphenoxy)propionic acid on the oxidation of cholesterol by rat liver mitochondria

Zusammenfassung Es wird gezeigt, dass bei männlichen Wistar-Ratten im 3-Wochen-Diätexperiment mit Zusatz von 0,3% 2-Methyl-2-(p-1,2,3,4-Tetrahydro-1-naphthylphenoxy)-Propionsäure (Su-13437) die Oxydation von 26-¹⁴C-Cholesterin zu¹⁴CO₂ durch Lebermitochondrien (bezogen auf mg/N) ähnlich wie bei den Kontrollen war. Bei den Versuchstieren war jedoch bei Abwesenheit von Cytosol im System die Oxydation durch Lebermitochondrien erhöht.

Beta1-adrenoceptor mediated salivary gland enlargement in the rat

Summary In the rat, prolonged activation of ₁-adrenoceptors causes a gain in salivary gland weight, whereas prolonged blockade of these receptors causes a reduction in weight.This work was supported by grants to J.E. from the Medical Faculty in Lund and the Swedish Medical Research Council (05927).     

Effects of plant lectins on cellular defense reactions of ascidian hemocytes

The effects of plant lectins on the three cellular defense reactions of hemocytes of the solitary ascidian,Halocynthia roretzi (hemocyte aggregation, phagocytosis, and an allogenic reaction), were investigated. Concanavalin A inhibited aggregation, while wheat germ agglutinin and ricin inhibited the allogenic reaction. Neither of the lectins showed inhibitory effects on phagocytosis, but ricin promoted phagocytosis. These effects of the lectins were diminished by the addition of sugars specific for the respective lectins. These results strongly suggest that different surface carbohydrates are involved in the recognition mechanisms of threeH. roretzi cellular defense reactions.
