摘    要:以火炬松优良家系20692、20615、201036、20639不同球果采收期的含未成熟合子胚的雌配子体为外植体,利用4种针叶树体胚发生专利培养基AL、TX、LP、DS,分别与AL、TX、LP培养基激素组合及无激素组合形成16种诱导培养基,开展不同家系、基本培养基、球果采收期对火炬松体细胞胚胎发生率影响的研究。结果表明:合子胚发育的第2、3、4阶段为最理想外植体,不同家系、基本培养基、激素组合对ESM诱导率及外植体褐化率均存在显著影响,16种组合的培养基中,在LPLP、LPTX、ALTX培养基上可获得较高的体胚诱导率,分别为18%、16%、11%.

Abstract:Exploring somatic embryogenesis in loblolly pine is important in performing clonal propagation and developing the genetic transformation. The megagametophyte,i, e. , immature zygotic embryos from the different cone collection time of the elite families 20 -692,20 -615,20 - 1036 and 20 -639 was used as explants. The four patent media AL, TX, LP and DS for somatic embryogenesis were combined with different hormones and no hormone to form 16 initiation media. The effects of different patent media, families and Cone collection time on induction rate of somatic embryos were studied. The results showed that the best explants were immature zygotic embryos of development stage 2, 3, 4. There was significant difference in ESM induction rates, explants browning using different families, basic media and hormone combinations. Among the 16 combination media tested, the media LP - LP,PL - TX and AL - TX got higher induction rate,i.e. ,18%,16% and 11%, respectively.
