Abstract:Objective To probe into the synergy of pre-exposure lower-intensity ultrasound ( US), or LIUS, in high-intensity focused ultrasound (HIFU) ablation of rabbit VX2 liver tumors, and its influence influence on the growth of the residual tumors. Methods Liver VX2 tumor models were established in 52 rabbits, which were randomly divided into three groups 20 days after vaccination, 22 for pre-irradiation group (Group A) ,20 for pure HIFU group (Group B) ,and 10 for control group (Group C). Group A received LIUS with sound power of 100 W irradiation 24 hours before 180 W HIFU therapy;Group B received sham LIUS pre-irradiation,the other treatment parameters were the same with group A ; group C received sham treatment. Tumor size was measured by MRI at 1 day before HIFU treatment and 7 day after HIFU treatment, and the tumor growth rate was calculated. On the seventh day after HIFU ablation, prognosis was analyzed by detection of the vascular endothelial growth factor (VEGF) expression using immunohistochemistry technology, and the pulmonary metastasis was observed with microscope. Results Compared with group B, the average EEF value of group A reduced nearly two times. Tumor size of three groups got larger as times went on. 7 days after HIFU treatment, the tumor growth rates of group A and B were dramatically lower than those of goup C ( P 〈 0. O1 ), and group A was minimum(P 〈 0.05 ). Compared the three groups,the expression of VEGF was the highest in group C , the lowest in group A ( P 〈 0.05 ). The pulmonary metastasis of group C was obviously more than those of group A and B ( P 〈 0.05 ), but there was no significant difference between group A and group B ( P 〉 0.05 ). Conclusion Pre-exposure LIUS can enhance the effects of HIFU, and can control the tumor metastasis. This provides a new method to enhance the effect of HIFU in clinical use.
