大豆胚芽中RNA提取方法的比较评价

摘    要:从细胞中分离完整性好、纯度高的RNA是进行基因克隆和基因表达研究的基础,不同植物、不同组织的RNA提取难点各异,适宜方法也不尽相同,大豆胚芽富含蛋白质、脂质、多糖、酚类化合物及未知次级代谢产物,其组成成分的复杂性增加了从大豆胚芽中提取高质量的RNA的难度,本文采用了尿素-LiCi法、异硫氰酸胍法及改进的异硫氰酸胍法等三种方法从大豆胚芽中提取了RNA,从RNA的完整性、产率和纯度等方面对这三种方法进行了比较和评价,结果发现改进的异硫氰酸胍法是从大豆胚芽中提取RNA的适宜方法。

Abstract:Isolating RNA with high-integrity and high-purity from cells is the base for studies on gene cloning and the regulation of gene expression. The difficulties of extracting RNA from different plants and different tissues are different, thus the appropriate methods are not always the same. Soybean embryo is rich in protein, lipid, polysaccharide, phenolic compounds and some unidentified secondary metabolites. The complexity of the components makes it more difficult to extracting RNA from soybean embryo. We used three methods including Urea-LiCl method,guanidine thiocyanate method and the modified guanidine thiocyanate method to isolate RNA from soybean embryo. The three methods are evaluated by determinating quality,purity and integrity of RNA sample and the results suggest that the modified guanidine thiocyanate method is more fit for extracting total RNA from soybean embryo.
