生物表面活性剂产生菌的筛选及鉴定

摘    要:从大庆油田采集的原油样品中筛选出一株产表面活性剂的菌株HD-322-2,根据形态特征、生理生化性质和16S rDNA序列分析对菌株进行鉴定,并运用红外光谱法和排油圈法对产物进行分析。菌株HD-322-2的16S rDNA序列与枯草芽孢杆菌的相似性达到98%,生理生化性质分析的结果亦与枯草芽孢杆菌相同,认定该菌株属于枯草芽孢杆菌。红外光谱分析结果表明:菌株HD-322-2产生的生物表面活性剂的主要成分为脂肽。该菌株具有较强的排油和降黏能力。

Abstract:A surfactant producing strain called HD-322-2 is screening from daqing oilfield.The strain was identified using the morphological characteristics,biochemical and physiological characteristics and 16S rRNA sequences analysis,and the products were analyzed by oil displacement method and infrared spectroscopy.The strain HD-322-2 is the closest phylogenetic neighbor with 98%,and even biochemical and physiological as similer as Bacillus subtilis,so the strain belong to Bacillus Subtilis.The strain HD-322-2 produced bio-surfactant was lipopeptide by infrared spectrum.The results of experiment show that the strain HD-322-2 has greater capacity of oil expulsion and viscosity reduction.
