流通池光纤荧光光度法测定乳酸

摘    要:用明胶和戊二醛将乳酸脱氨酶(LDH)固定于PVC颗粒表面后,装成酶柱,以流通池光纤荧光光度法连续测定乳酸,检测乳酸的线性范围为2.0×10-5~4.0×10-1mol·L-1。该方法简便、快速,重现性、稳定性均好,酶的使用寿命长,已用于酿牛奶和食用白醋中乳酸的测定。

Abstract:A simple, rapld and accurate fiber - optic fluorimetry with flow cell for the contiually determination of lactic acid in food by using immobilized lactate dehydrogenase (LDH) has been established.The LDH is immobilized onto the pvc particle by covalently arosslinking via glutaric dialdehyde,then an enzymlc Colum is packed with the pvc particle.A good linear Calibration curve over the Iactate concentration range of 2.5×10-5 to 4. 0×10-4mol·L-1 is obtained.lactic acid in sour milk and vinegar was determined by this method.
