摘    要:建立HPLC测定重楼块根中4种重楼皂苷含量的方法。结果表明:重楼皂苷Ⅰ质量浓度在0.054~0.540 mg·mL-1范围内线性关系良好,r=0.999 6 (n=5),回收率为99.12%,RSD为1.95%;重楼皂苷Ⅱ质量浓度在0.043 4~0.434 mg·mL-1范围内线性关系良好,r=0.990 8 (n=5),回收率为99.10%,RSD为1.89%;重楼皂苷Ⅵ质量浓度在0.012 6~0.126 mg·mL-1范围内线性关系良好,r=0.993 3 (n=5),回收率为99.46%,RSD为2.01%;重楼皂苷Ⅶ质量浓度在0.010 3~0.103 mg·mL-1范围内线性关系良好,r=0.999 1 (n=5),回收率为96.52%,RSD为2.25%。HPLC法能够准确检测出重楼块根的主要皂苷成分及其含量,且操作简便,可为重楼中4种主要皂苷含量的检测提供科学方法。

Abstract:An HPLC method was developed for the determination of contents of four steroidal saponins in root tubers of it. The results showed good linearity within the measured concentration range of 0.054~0.54 mg·mL-1 for saponin I (r =0.999 6), with the rate of recovery of 99.12% and RSD of 1.95%. The results showed good linearity within the measured concentration range of 0.0434-0.434mg/mL for saponin II (r =0.990 8), with the rate of recovery of 99.10% and RSD of 1.89%. The results showed good linearity within the measured concentration range of 0.0126-0.126mg/mL for saponin VI (r =0.993 3), with the rate of recovery of 99.46% and RSD of 2.01%. The results showed good linearity within the measured concentration range of 0.0103-0.103mg/mL for saponin VII (r =0.999 1), with the rate of recovery of 96.52% and RSD of 2.25%. The results indicated that the HPLC method could accurately detect the contents of major steroidal saponins in root tubers of P. polyphylla, and the operation is simple. This provided scientific methods for the determination of four major saponins in P.polyphylla.
