硫堇/聚2,6-吡啶二甲醛膜电极检测DNA及其损伤

摘    要:在玻碳电极上电聚合2,6-吡啶二甲醛,再共价键合硫堇至聚2,6-吡啶二甲醛膜的表面上制备成修饰电极.用电化学交流阻抗表征了该修饰电极的制备过程.以差示脉冲伏安法研究了该修饰电极对鲱鱼精DNA的作用,发现该修饰电极对dsDNA具有选择性的伏安响应.在优化的实验条件下,修饰电极氧化峰电流与dsDNA浓度的对数在0.10~100mg/L范围内呈现良好的线性关系,检测限为0.06mg/L.将该修饰电极用于检测鲱鱼精DNA的化学损伤,其峰电流与DNA的损伤程度成正比.该修饰电极制备简单、检测快速,5支电极间相对标准偏差小于4.83%.

Abstract:A thionine/Poly(pyridine-2,6-dicarbaldehyde)modified electrode(Th/PPDA/GCE)was fabricated for detecting DNA and its damage.Th/PPDA/GCE was prepared by electropolymerizing pyridine-2,6-dicarbaldehyde on a glassy carbon electrode,and covalently immobilizing thionine on the surface of poly(pyridine-2,6-dicarbaldehyde)film.The preparation process and electrochemical responses of Th/PPDA/GCE were investigated by using electrochemical impedance spectroscopy and differential pulse voltammetry for herring sperm DNA...
