怀地黄HPLC指纹图谱研究

摘    要:目的:建立并比较怀地黄不同栽培品种HPLC指纹图谱,为强化怀地黄质量控制和品种鉴定与划分建立图谱标准.方法:采用HPLC,以0.4%醋酸溶液和乙腈进行梯度洗脱,检测波长为260nm;柱温:30℃;流速:1mL/min.使用"中药色谱指纹图谱相似度评价系统"进行数据处理,对不同栽培品种怀地黄的HPLC指纹图谱进行了相似度和聚类分析.结果:获得了HPLC优化技术条件;建立了含有18个特征指纹峰的怀地黄HPLC化学对照指纹图谱;9个农家品种划分为2大类,又可细分为4类.结论:该图谱可作为怀地黄质量控制的标准图谱,为怀地黄的质量控制、品种鉴别及划分提供理论和技术支撑.

Abstract:Objective:To study the HPLC fingerprint of Rehmannin gtutinosa and establish a chromatographic standard for the quality control as well as to comparative study the HPLC fingerprint among various farm cultivars.Methods:The HPLC method was used.The mobile phase is 0.3%acetic acid and methyl cyanide,The wavelength is 260nm.The temprature is 30℃,and the flow rate is 1mL/min.The chromatographic fingerprint similarity evaluation system was adopted for data analysis.Results:Obtaining the HPLC optimization technology conditions and containing 20characteristic fingerprint peaks Rehmannia HPLC chemical control fingerprint;9farm species is divided into two categories,subdivided into four categories.The standard fingerprint chromatogram for Henan trueborn Rehmannin gtutinosa which contain 20characteristic peaks was built. Conclusion:The HPLC fingerprint method summaried in this article can be used as the typical chromatography and provide the theoretical and technical support for species identification and division of Rehmannin gtutinosa.
