蚯蚓乙酰胆碱酯酶的分离与固定化试验研究

摘    要:从蚯蚓中提取乙酰胆碱脂酶,通过硫酸铵盐析,离子交换层析等方法获得了较纯的胆碱酯酶,并对其部分性质进行测定.分别采用鸡蛋膜固定化法,活化载体-共价结合法和戊二醛交联法进行酶的固定.结果表明:蚯蚓体内乙酰胆碱酯酶含量较低,且纯化过程中损失较大,结果欠佳;活化载体-共价结合法固定得到的乙酰胆碱酯酶酶膜活性高,酶不易损失,重现性好.为建立快速的痕量有机磷类的酶法分析方法奠定了基础.

Abstract:Acetylcholinesterase was extracted from earthworms. Purer Acetylcholinesterase was obtained through salting out by ammonium sulphate and ion exchanging.Some characteristics of the enzyme and the fixation methods of the enzyme were also studied.The results shows that the concentration of Acetylcholinesterase in earthworms was low,it was not steady and its loss during purification process was great.The Acetylcholinesterase membrane fixed by activated carrier had higher enzyme activity and longer life-span.
