铁棍山药试管苗快繁培养基的优化

摘    要:研究了不同植物生长调节剂(TDZ、6-BA、KT和NAA)浓度及其组合对铁棍山药试管苗快繁的影响,结果表明:KT和NAA组合时,在MS+KT 2mg·L-1+NAA 0.02mg·L-1培养基中试管苗高度达7.67cm,但繁殖系数只有2.67;在KT和NAA组合的基础上再添加TDZ后,在MS+KT 2mg·L-1+NAA 0.02mg·L-1+TDZ 0.02mg·L-1培养基中试管苗繁殖系数提高到6.00,试管苗健壮且生长旺盛;在比较不同细胞分裂素(KT,6-BA和TDZ)与NAA组合对试管苗快繁的影响时发现,6-BA与NAA组合最不适合试管苗的生长,KT与NAA组合中的试管苗生长缓慢,繁殖系数低,TDZ与NAA组合的培养基中有类原球茎形成.因此,综合以上研究结果,本实验认为KT、TDZ和NAA组合的培养基(MS+KT 2mg·L-1+NAA 0.02mg·L-1+TDZ 0.02mg·L-1)更适于铁棍山药试管苗的快繁.

Abstract:Studies were carried out with the aim of evaluating in vitro the effects of growth regulators-auxin(NAA) and cytokinin(TDZ,KT,6-BA) combined at different levels on virus-free Dioscorea opposita Thunb.cv Tiegun rapid propagation on Murashige and Skoog media.The results were as follows: KT(2 mg·L-1) in combination with NAA(0.02 mg·L-1) showed most increase,shoot length in vitro cultured on the medium reached 7.67 cm and frequency of proliferation were 2.67.Addition of TDZ(0.02 mg·L-1) to the MS+KT 2 mg·L-1+NAA 0.02 mg·L-1,could increase propagation coefficient which reached 6.00,plantlets were vigorous and robust.Through comparing the effects on rapid propagation of virus-free plant materials by different cytokinin(KT,6-BA,TDZ) along with NAA,we found that the efficiency of propagating virus-free plant materials was the worst with the hormone combination of 6-BA and NAA,KT in combination with NAA showed slow growth and gave a lower propagation coefficient,TDZ in combination with NAA can form some protocorm-like bodies.Thus,on the basis of the results of the current study,we can conclude that explants of virus-free Dioscorea opposita Thunb.cv Tiegun cultured on MS+KT 2 mg·L-1+NAA 0.02 mg·L-1+TDZ 0.02 mg·L-1showed the best response to shoot multiplication.
