苦参素联合富硒酵母干预大鼠肝癌变进程中体征的动态变化

摘    要:以二乙基亚硝胺(n-nitroso-diethylamin,DENA)为诱变剂建立SD大鼠肝癌变动物模型,同时用苦参素(oxymatrine,OMT)联合富硒酵母(selenium enriched yeast)对其进行干预.自诱癌之日起,分别在2、4、6、8、10、12、14、16、18周观察相关体征后,处死大鼠,取其肝脏组织,制作病理切片,对大鼠在肝癌演进及药物干预过程中的体征及组织病理学的动态变化进行研究.结果显示,干预组大鼠在体重、毛色、精神状态、肝脏外形及组织显微结构等方面均优于诱癌组,且从11w开始干预组体重极显著高于诱癌组(P<0.01).说明干预组大鼠在肝癌变进程上要滞缓于诱癌组,在癌前病变及肝癌期的生存质量明显优于诱癌组.提示OMT联合富硒酵母减弱了DENA诱发肝癌的作用,在一定程度上延缓了肝癌的发生发展.

Abstract:Animal models of every pathologic stage during the development of SD rats' liver cancer were established by the gavage with n-nitroso-diethylamin(DENA).Meanwhile,treatment groups were administrated by oxymatrine and selenium enriched yeast(OMT+Se).After observing the related signs,animals were sacrificed respectively at 2,4,6,8,10,12,14,16 and 18 week from the first day of gavage and livers were harvested for pathological grading to explore the dynamic changes of signs and histopathological structures during the development of rats' liver cancer as well as the intervention treatment of OMT+Se.Results showed that the treatment group was better than the induced group on the weight,liver appearance and the microscopic structures.From the 11th week,the weights of the treatment group were significantly more than the induced group(P<0.01).It hinted that the treatment group was slow than the induced group in the progress of hepatocarcinogenesis and the qualities of life in the treatment group were much better especially in precancerosis and liver cancer.Furthermore,it suggested the intervention of OMT+Se reduced the carcinogenicity of DENA and delayed the progress of hepatocarcinogenesis to some extent.
