摘    要:在有机溶剂中利用固定化脂肪酶Novozym435催化酯化反应以分离扁桃酸对映体,考察了反应时间、温度、摇床转速、加酶量、扁桃酸浓度及乙醇浓度对反应转化率和产物对映体过量值(eep)的影响.结果表明,在二氯乙烷中,拆分扁桃酸对映体的最佳条件为:加酶量40mg,扁桃酸20mM,乙醇80mM,转速150rpm,温度40℃.在此条件下,反应24h后,转化率为30%,产物中R-扁桃酸乙酯eep达到92%.

Abstract:The aim of this study is to resolute (R,S)-mandelic acid enantiomers in organic solvent using immobilized lipase Novozym 435. The enantiomers was esterified with ethanol. Conversion of mandelic acid and the enantiomeric excess of the product (eep) were used to estimate the reaction efficiency. Reaction conditions were optimized and determined to be: enzyme loading 40 mg, concentration of mandelic acid 20 mM, concentration of ethanol 80 mM, shaker speed 150 rpm, temperature 40 ℃, time 24 h. Under the optimal conditions, the conversion was 30% after 24 h of reaction and an eep of 92% was obtained.
