微生物学实验综合模块的设计研究

摘    要:对微生物学实验“产生蛋白酶菌株的筛选、鉴定、培养和选育”进行了教学改革,改变了以往的内容安排以及考核方式,并将该实验综合为一个统一的实验模块.模块是由8个紧密相连的实验所组成的一个综合性实验模块,其每个实验的具体内容分别为:玻璃器皿的清洗、包扎,培养基的配制及灭茵,产蛋白酶茵株的筛选、纯化及保藏,产蛋白酶茵的革兰氏染色及镜检,产蛋白酶茵的PCR鉴定,16SrDNA序列的比对及其分析,产蛋白酶细菌的生理生化鉴定.产蛋白酶茵的培养优化实验以及其诱变选育.

Abstract:Contents and examination forms of teaching comprehensive experiment of microbiology, which focuses on isolation, identification, cultivation and breeding of protease-producing strains, are introduced in this paper. The comprehensive experiment was composed of eight closely linked parts, which were as follows: clean and pack of glassware, and preparation and sterilization of media; screening, purification and preservation of protease-producing strains; gram stain and examination of microscope; PCR identification of strains; alignment and analysis of 16S rDNA sequence of the strains; identification of physiological and biochemical characters of the strains; optimization of the culture medium and mutation breeding of the strains.
