黄精属植物酯酶同工酶的研究

摘    要:垂直平板聚丙烯酰胺凝胶电泳法和K-K-J-G电泳光密度扫描法分析了黄精属三种不同植物——长梗黄精、多花黄精和玉竹的叶片,根状茎和根的酯酶同工酶谱。结果表明,无论是叶片、根状茎或根内的酶谱带,三种植物之间均存在明显差别。在叶片中,虽然三种植物的酶带总数量相同(10条),但酶带的位置和活性明显不同。长梗在C区中谱带最多,玉竹在B区中最多,多花在A区中最多。在根中,三者的酶带总数量和位置也有差异。在C区内,只有多花有一条酶带,而长梗和玉竹均无酶带,特别是D区的出现为根所特有,表明器官之间酶带的不同。作为主要药用部分的根状茎,三种植物的酶带,在B区和C区均存在显著差别,这些差别反映了各种之间遗传基质的差别,可以作为鉴别物种的生化标志之一。

Abstract:The esterase isoenzyme zymograms from leaf,rhizome and root among three species Polygonatum plants i.e.,Polygonatum filipes mear.,P.cyrtonema Hua.and P.odoratum(Mill.)Druce.,were obtained by polyacrylamide gel slab electrophoresis and distingushed by K-K-J-G electrophoresis photodensimeter scan. The results showed that there were significant differences among the esterase isozyme patterns of leaf,rhizome,and root of three species Polygonatum.At the leaf, although total band numbers of the esterase isozyme zymogram were the same(10 bands),but the differences in their peak area and distribution were found.In zone C, zymogram of P.filipes Mear.had the most bands among three species Polygon- atum.In zone B,that of P.odoratum(Mill.)Druce.had the most.In zone A,that of P.cyrtonema Hua.had the most.At the root the situation was similar,but zone D was found.It indicated that the isoenzymes were different between the organs.The paper suggested that the zymogram differences of esterase isozyme among varietal plants were decided by their genotypic matteral property,and esterase isozyme patterns thus could be used as a biochemical marker to identify plant species.
