| 摘 要： | 将酿酒酵母3-磷酸甘油脱氢酶基因GPDH1的编码序列构建到表达载体pPIC3．5K中，利用电转化法将构建好的表达载体转入巴斯德毕赤酵母中，利用含有G418的YPD平板筛选到两个阳性转化子，通过分子生物学及外源蛋白表达实验证明，酿酒酵母3-磷酸甘油脱氢酶基因GPDH1已经整合到巴斯德毕赤酵母的基因组上，并且得到了预期的表达。

| Abstract: | Expression plasmid, pPIC3.5K-ScGPD, which was constructed by inserting glycerol-3-phosphate dehydrogenase gene from Saccharomyces cerevisiae (Sc-GPDH1 ) into yeast expression vector pPIC3.5K,was transformed into Pichia pastoris by electroporation. Two positive transformants were screened in YPD- G418 plates. The results indicated glycerol-3-phosphate dehydrogenase gene from Saccharomyces cerevisiae (Sc-GPDH1) has been integrated into Pichia pastoris genome and expressed successfully . |
