富产海藻糖合成酶菌株的筛选

摘    要:研究了从土壤中分离获得一种富产海藻糖合成酶菌株的全过程.通过平板初筛和摇瓶产酶转化试验及离子色谱仪(HPAEC)检测,确证该酶能转化聚合度大于3的麦芽寡糖合成海藻糖,转化率约为40%,通过形态、结构特征分析以及16SrDNA基因全序列与参比菌株的基因序列比较,菌株JNU-1与食尼古丁节杆菌16S rDNA序列同源性达95.12%,故将该菌株定名为食尼古丁节杆菌JNU-1(Arthrobacter nicotinovorus JNU-1).

Abstract:The process of screening a bacterium of rich-producing trehalose synthase enzyme from soil was studied in this paper. After plate screening and conversion test in surging bottle and checking by HPAEC, it was approved that this enzyme was able to transform maltose to trehalose with yield of 40%. The morphological,cultural physiological characteristics of trehalose-producing were described in this paper.The full-lenth of 16S rDNA has been tested and compared with the type strains of all known Arthrobacter sp in Genebank. The results showed the similarity values of 16S rDNA sequence between bacterium JNU-1 and Arthrobacter nicotinovorus were 95.12%.Therefore bacterium JNU-1 was named Arthrobacter nicotinovorus JNU-1.
