苦杏仁苷与人血清白蛋白相互作用的荧光光谱研究

摘    要:应用荧光光谱法,研究了苦杏仁苷与人血清白蛋白(HSA)在pH=7.40的Tris-HCl缓冲溶液中相互作用的情况.研究表明,苦杏仁苷通过静态机理猝灭了HSA的内源荧光.并计算得到苦杏仁苷与HSA的结合常数和结合位点数.通过热力学数据,推断出苦杏仁苷与人血清白蛋白之间主要靠氢键和范德华力结合.同时采用同步荧光技术考察了苦杏仁苷对HSA构象的影响.

Study on the Interaction between Amygdalin and Human Serum Albumin by Fluorescence Spectroscopy

Abstract:The interaction between amygdalin and human serum albumin(HSA)in buffer solution(pH =7.40)were studied by fluorescence spectroscopy.It was found that amygdalin quenched the fluorescence of HSA via a static quenching process.The binding constant and number of binding sites were found.Values of thermodynamic parameters were calculated.These dates indicated that hydrogen bonds and van der Waals force played a major role in the binding of amygdalin and HSA.Using synchronous fluorescence spectroscopy,the effect of amygdalin on the configuration of HSA was studied.
