人面果酮抗糖尿病活性的筛选

摘    要:为研究人面果中酮的抗糖尿病活性,筛选具有较强活性的化合物,采用MTT法检测了化合物对HepG2细胞增殖率的影响.用高浓度胰岛素诱导HepG2细胞,建立了胰岛素抵抗模型(HepG2/IR),并采用葡萄糖氧化酶法,检测了不同浓度化合物对HepG2细胞胰岛素抵抗模型的葡萄糖消耗的影响.结果表明:人面果中酮中化合物1、3、4(除化合物2、5)能够明显促进HepG2/IR细胞的葡萄糖消耗,具有潜在的抗糖尿病活性.

Abstract:To study the anti-diabetic activities of xanthones from Garcinia xanthochymus and to screen the active compounds, MTT assay was used to determine its effect on the survival rate of HepG2 cells. The HepG2 cells were induced by high concentration of insulin to establish the insulin-resistance cell model (HepG2/IR). Then the glucose consumption of cells treated with different concentration of compounds was measured by glucose oxidase and peroxidase (GOD-POD) assay. The results showed that compounds 1, 3, and 4 (except compounds 2 and 5) of the xanthones from Garcinia xanthochymus could significantly enhance the glucose consumption of HepG2/IR cell, which suggested that they had the potential anti-diabetic activities.
