摘    要:以3种不同细胞核分离缓冲液制备了水稻幼叶样品,通过流式细胞术比较了其DNA分辨率效果.结果发现:利用Otto buffer制备细胞核悬浮液的效果最好,不仅变异系数较低,碎片少,还对多倍性细胞核的完整性具有很好的稳定作用.该法可用于对水稻根和种子的快速准确的DNA含量测定和倍性鉴定.

Abstract:The rice young leaf was prepared by three different nucleus buffers and the resolution effect of nucleus suspensions were compared by flow cytometry. The results showed that the Otto buffer was the best solution of the three, with lower coefficient of variation and little fragment. Moreover, this buffer also stabilized well for the integrality of polyploidy nucleus. So this method could be used to determine the DNA content and identify the ploidy level from different rice root and seed fast and correctly.
