硒酸酯多糖和亚硒酸钠抗白血病效应的实验研究

摘    要:应用MTT比色分析法和细胞克隆形成法比较研究了有机硒化物——硒酸酯多糖(Kappa-selenocarrageenan,KSC)和无机硒化物——亚硒酸钠(Na2SeO3)对白血病K562细胞的作用.结果表明:KSC和Na2SeO3体外在实验浓度范围内均对K562细胞的增殖具有显著的抑制作用,并有剂量反应关系(p<0.001);K562细胞中具有自我更新能力的细胞群体(克隆形成细胞)对硒的抑制作用更敏感;KSC的抗白血病效应明显高于含同样硒量的Na2SeO3,KSC的作用约为Na2SeO3的5倍.本研究证明药理剂量的硒可显著抑制白血病细胞的增殖,有机硒比无机硒具有更高的抗白血病活性

Abstract:The antileukemia effects of kappa selenocarrageenan (KSC), an organic selenium compound, and its inorganic form sodium selenite(Na 2SeO 3) were studied in K 562 cells in culture with MTT colorimetric assay(MTT) and colony forming rate assay (CFR). The data showed that both KSC and Na 2SeO 3 significantly inhibited the proliferation of K 562 cells in vitro ( p <0 001); The colony forming cell in K 562 cells was more sensitive to selenium (Se). Compared with sodium selenite containing the same amount of Se, KSC was more effective in antileukemia activity, its effect was about 5 times as high as that of Na 2SeO 3. These results indicate that pharmacologic concentration of selenium can effectively inhibit the growth of K 562 cells, and the organic form of Se is more effective than its inorganic form.
